Ischemic stroke represents a highly perilous cerebrovascular disorder, involving a variety of complex pathophysiological mechanisms. OIP5 antisense rna 1 (OIP5-AS1) is a long non-coding rna (Lncrna) that has been shown to play a pivotal role in a variety of disease systems. However, there are relatively few studies on ischemic stroke. This research aimed to elucidate the direct impact of OIP5-AS1 on neuronal cells following cerebral ischemia-reperfusion. Our study revealed a significant reduction in OIP5-AS1 expression in mouse neurons following middle cerebral artery occlusion/reperfusion (MCAO/R). Overexpression of OIP5-AS1 in neurons inhibits neuronal apoptosis induced by cerebral ischemia-reperfusion injury (CIRI) and exerts a neuroprotective role. Mechanistically, OIP5-AS1 may play a neuroprotective role after CIRI by up-regulating the expression of TAK1 binding protein 2 (TAB-2), reducing neuronal mitochondrial damage, and inhibiting apoptosis. OIP5-AS1 may become a novel therapeutic target for ischemic stroke.

BackgroundGranulosa cell proliferation and survival are essential for normal ovarian function and follicular development. long non-coding rnas (lncrnas) have emerged as important regulators of cell proliferation and differentiation. Nuclear paraspeckle assembly transcript 1 (NEAT1) has been implicated in various cellular processes, but its role in granulosa cell function remains *** investigated the function of lncrna NEAT1 in human ovarian granulosa-like tumor cells (KGN). The effects of NEAT1 overexpression or silencing on cell proliferation and cell cycle were evaluated using CCK-8 assays and flow cytometry. The interaction between NEAT1, miR-29a-3p, and IGF1 was examined using dual-luciferase reporter assays, qRT-PCR, and Western blot ***1 promoted granulosa cell proliferation and cell cycle progression by indirectly upregulated IGF1 expression through acting as a molecular sponge for miR-29a-3p. Cell proliferation and G2/M phase proportions were increased by overexpression of NEAT1, whereas cell proliferation and G2/M phase proportions decreased with NEAT1 silencing. The effects of NEAT1 on cell proliferation and cell cycle-related proteins (CCNB1 and CDK2) were partially reversed by miR-29a-3p mimic, while miR-29a-3p inhibitor rescued the effects of NEAT1 *** NEAT1 could promote ovarian granulosa cell proliferation and cell cycle progression via the miR-29a-3p/IGF1 axis in polycystic ovary syndrome. Further investigation of this mechanism in clinical samples may have implications for understanding ovarian physiology and pathology.

Osteosarcoma, the most common primary bone cancer in children and adolescents, presents significant challenges, particularly in metastasis and recurrence, resulting in poor survival rates. This study explores the role of PANoptosis-a complex cell death mechanism involving pyroptosis, apoptosis, and necroptosis-in osteosarcoma by identifying relevant long non-coding rnas (lncrnas) and their prognostic significance. Bioinformatics analyses used rna expression data from the GEO and TARGET databases to identify differentially expressed genes (DEGs) and PANoptosis-related genes (PRGs). Co-expression analysis revealed lncrnas linked to PRGs, forming a risk prognostic model. Five PRGs and two lncrnas were significantly associated with prognosis, with the model showing high predictive accuracy (AUC 0.876, 0.787, and 0.794 for 1, 3, and 5 years). Notably, lncrna AC133552.2 was downregulated in osteosarcoma tissues, correlating with poor survival and reduced immune infiltration. Silencing AC133552.2 promoted cell proliferation and migration, while overexpression inhibited tumor growth and metastasis, confirmed in xenograft models. AC133552.2 emerges as a potential biomarker and therapeutic target, with future research needed to explore its molecular mechanisms and clinical application.

Acute coronary events show a diurnal rhythm, and atherosclerotic plaque vulnerability, as a histomorphological characteristic of major adverse cardiovascular events, is a key target for intervention. Although oscillating micrornas reduce plaque stability by facilitating macrophage apoptosis in lesions, whether rhythmic long noncoding rna (lncrna) can regulate diurnal oscillations in plaque stability and the potential underlying mechanism remain unclear. In this study, we examined whether rhythmic lncrnas are involved in the pathogenesis and progression of atherosclerosis and detected a novel circadian lncrna-AK023617, which is positively correlated with the peak occurrence of major adverse cardiovascular events. Transfection of short interfering rna specific to lnc-AK023617 into THP-1 cells dampened the oscillation of immunity-related GTPase family M protein 1 (Irgm1), which is negatively related to plaque stability. In ApoE- /- mice fed a high-fat diet for 12 weeks, diurnal variations in lncAK023617 were consistent with the proportions of necroptotic cells in atherosclerotic plaques. In addition, reduced expression of lncAK023617 inhibited P-RIP3 and P-MLKL in THP-1 cells. Mechanistically, lncAK023617 interacted with the core molecular clock Bmal1 and promoted nuclear translocation of Bmal1, which could directly bind to the E-BOX elements in the Irgm1 promoter. Thus, oscillating lncAK023617 in macrophages can affect plaque stability by regulating necroptosis, which regulates circadian expression of the target gene Irgm1 by increasing the transcriptional activity of Bmal1, ultimately determining the diurnal oscillations in plaque stability. Therefore, lncAK023617 may serve as a specific target to ameliorate atherosclerotic plaque vulnerability.

ObjectiveThis study aimed to investigate the role of the autophagy-related long noncoding rna (lncrna) MIR210HG in hepatocellular carcinoma and its potential as a therapeutic target. MethodsLncrna MIR210HG expression and its correlation with survival outcomes in hepatocellular carcinoma patients were analyzed using data from The Cancer Genome Atlas (TCGA). Kaplan-Meier and Cox regression analyses were conducted to assess survival correlations. Quantitative reverse transcription PCR was used to measure lncrna MIR210HG expression in liver cancer cells and normal liver cells. Functional assays, including CCK-8, Transwell, flow cytometry, and western blot, were performed to evaluate the effects of lncrna MIR210HG on cell proliferation, invasion, apoptosis, and autophagy in hepatocellular carcinoma. ResultsElevated lncrna MIR210HG expression correlated with poor overall survival in hepatocellular carcinoma patients. Lncrna MIR210HG expression was significantly up-regulated in hepatocellular carcinoma cells compared to normal liver cells. Knockdown of lncrna MIR210HG inhibited cell proliferation and autophagy, while promoting apoptosis in hepatocellular carcinoma cells, findings that were confirmed through both in vitro and in vivo studies. ConclusionThe findings suggest that lncrna MIR210HG contributes to hepatocellular carcinoma progression by regulating autophagy and could serve as a promising therapeutic target in hepatocellular carcinoma treatment strategies.

Screened of long non-coding rna related to wool development and fineness in Gansu alpine fine-wool sheep

Wool growth and fineness regulation is influenced by some factors such as genetics and environment. At the same time, lncrna participates in numerous biological processes in animal production. In this research, we conducted a thorough analysis and characterization of the microstructure of wool, along with long non-coding rnas (lncrnas), their target genes, associated pathways, and Gene Ontology terms pertinent to the wool fineness development. The investigation utilized scanning electron microscopy and transcriptomic technology, focusing on two distinct types in Gansu alpine fine-wool sheep: coarse type (group C, MFD = 22.26 +/- 0.69 mu m, n = 6) and fine type (group F, MFD = 16.91 +/- 0.29 mu m, n = 6), which exhibit differing wool fiber diameters. The results showed that fine type wool fiber scales were more regularly distributed in rings with large scale spacing and smooth edges, while coarse type wool fiber scales were more irregularly arranged in tiles with relatively rougher edges, and the density of wool scales was greater than that of fine type wool. Furthermore, a comprehensive analysis revealed 164 differentially expressed lncrnas along with 146 potential target genes linked to these lncrnas in the skin tissues from groups C and F. Utilizing functional enrichment analysis on the target genes, we successfully identified a number of target genes might be associated with the improvement of wool fineness, such as FOXN1, LIPK, LOC101116068, LOC101106296, KRTAP5.4, KRT71, KRT82, DNASE1L2, which are related to hair follicle development, histidine metabolism, epidermal cell differentiation, oxidative phosphorylation and hair cycle process. Additionally, the interoperability network involving lncrnas-mrnas indicated lncrnas (MSTRG.17445.2, XR_006060725.1, MSTRG.871.1, MSTRG.10907.4) might play a significant role in the wool growth development and fineness improvement process. The expression and clinical significance of serum exosomal-long non-coding rna DLEU1 in patients with cervical cancer

Background: Accumulating evidence has demonstrated that the long non-coding rna (lncrna) lymphocytic leukaemia deletion gene 1 (DLEU1) is abnormally overexpressed in many cancer types, including cervical cancer (CC). However, the potential clinical significance of DLEU1 in serum exosomes of patients with CC remains unclear. Methods: The expression of serum exosomal DLEU1 was detected by quantitative real-time polymerase chain reaction (qRT-PCR). A receiver operating characteristic (ROC) curve was plotted to evaluate the clinical diagnostic efficacy of DLEU1. The Kaplan-Meier survival curve and Cox proportional hazards model were used to assess the effect of DLEU1 on postoperative recurrence, metastasis and prognosis among patients with CC. Results: Our research showed that DLEU1 expression in the serum exosomes of patients with CC was significantly upregulated compared to that in patients with cervical intraepithelial neoplasia (CIN) and healthy controls (HCs) (both p < .001). DLEU1 relative expression was significantly correlated with tumour size, cervical invasion depth, pathological grade, International Federation of Gynecology and Obstetrics (FIGO) stage and lymph node metastasis among patients with CC (p < .01 all). The combined detection of DLEU1, carbohydrate antigen 125 (CA-125) and squamous cell carcinoma (SCC) exhibited significantly higher diagnostic efficiency (p < .01). Furthermore, the overall survival (OS) and disease-free survival (DFS) of CC patients in the high DLEU1 expression group were markedly lower than those in the low DLEU1 expression group (both p < .01). Cox univariate and multivariate regression analyses indicated that DLEU1 was an independent risk factor for postoperative recurrence and metastasis in CC patients. Conclusions: Our findings suggest that serum exosome DLEU1 has certain clinical value for diagnosing, monitoring recurrence and metastasis, and evaluating CC prognosis.

The development of effective immunotherapies for solid tumors remains a significant challenge. In previous studies, we identified PVT1, a long non-coding rna, with the peptide HF10 derived from PVT1, presented by HLA-A24. This study aims to develop a single-chain variable fragment (scFv) that specifically recognizes the HLA-A24/HF10 complex (HF10 scFv) and to evaluate its specificity, reactivity, and therapeutic potential as part of a T cell engaging bispecific antibody (HF10xCD3) in vitro and in vivo. Using a scFv phage display library, we screened for scFv clones targeting the HLA-A24/HF10 peptide complex. The selected HF10 scFv was engineered into an IgG1 format (HF10-hIgG1), which demonstrated high affinity (KD = 2.18 x 10(-8) M) and specific detection of the HLA-A24/HF10 complex on HLA-A24( +)/PVT1( +) tumor cell lines. Furthermore, HF10 scFv was incorporated into a T cell engaging bispecific antibody (HF10xCD3), which induced cytotoxicity in these tumor cell lines. In a mouse xenograft model, HF10xCD3 administration exhibited significant anti-tumor activity. In conclusion, HF10xCD3 represents a promising candidate for immunotherapy targeting solid tumors.

Cervical cancer (CC) and ovarian cancer (OC) are among the most common gynecological cancers with significant mortality in women, and their incidence is increasing. In addition to the prominent role of the malignant aspect of these cancers in cancer-related women deaths, chemotherapy drug resistance is a major factor that contributes to their mortality and presents a clinical obstacle. Although the exact mechanisms behind the chemoresistance in these cancers has not been revealed, accumulating evidence points to the dysregulation of noncoding rnas (ncrnas), particularly long non-coding rnas (lncrnas) and circular rnas (circrnas), as key contributors. These ncrnas perform the roles of regulators of signaling pathways linked to tumor formation and chemoresistance. Strong data from various recent studies have uncovered that the main mechanism of these ncrnas in the induction of chemoresistance of CC and OC is done through a dysregulated mirna sponge activity as competing endogenous rna (cerna) in the competing endogenous rna networks (ceRNETs), where a mirna regulating a messenger rna (mrna) is trapped, thereby removing its inhibitory effect on the desired mrna. Understanding these mechanisms is essential to enhancing treatment outcomes and managing the problem of drug resistance. This review provides a comprehensive overview of lncrna- and circrna-mediated ceRNETs as the core process of chemoresistance against the commonly used chemotherapeutics, including cisplatin, paclitaxel, oxaliplatin, carboplatin, and docetaxel in CC and OC. Furthermore, we highlight the clinical potential of these ncrnas serving as diagnostic indicators of chemotherapy responses and therapeutic targets.

Glioma is the most common primary malignant brain tumor globally, with a poor prognosis due to its complex interaction with the tumor microenvironment (TME). Despite advancements in glioma treatments, patient survival remains low, highlighting the need for additional molecular investigations. long non-coding rna Colorectal Neoplasia Differentially Expressed (lncrna-CRNDE) has been linked to tumorigenesis in various cancers, influencing tumor progression via the tumor microenvironment (TME). Its role in tumor-associated macrophages in gliomas is unclear. This study investigates whether lncrna-CRNDE promotes glioma progression by modulating these macrophages. Using the TCGA database, we assessed lncrna-CRNDE expression and its prognostic significance in glioma, and its association with the immune microenvironment. We collected 42 glioma samples of different grades from our institution. Using RT-qPCR, Western blotting, ELISA, and other cell biology techniques, we analyzed lncrna-CRNDE's effects on glioma proliferation and invasion and validated its role in glioma progression through animal model experiments. TCGA bioinformatic analysis showed higher lncrna-CRNDE expression in glioma than in normal tissues, which correlated with poorer prognosis. Clinical glioma samples via RT-qPCR also showed elevated lncrna-CRNDE in tumors, which was associated with reduced survival. Cellular experiments confirmed lncrna-CRNDE's promotion of glioma proliferation and invasion, which was associated with the tumor microenvironment (TME). Further studies revealed lncrna-CRNDE's role in macrophage polarization in the TME, enhancing glioma progression. Animal experiments confirmed lncrna-CRNDE's role in glioma progression. Lncrna-CRNDE is highly expressed in M2 macrophages and strongly associated with glioma progression. It is a potential prognostic marker and therapeutic target for glioma.
