Fed-batch and perfusion culture processes: Economic, environmental, and operational feasibility under uncertainty

This article evaluates the current and future potential of batch and continuous cell culture technologies via a case study based on the commercial manufacture of monoclonal antibodies. The case study compares fed-batch culture to two perfusion technologies: spin-filter perfusion and an emerging perfusion technology utilizing alternating tangential flow (ATF) perfusion. The operational, economic, and environmental feasibility of whole bioprocesses based on these systems was evaluated using a prototype dynamic decision-support tool built at UCL encompassing process economics, discrete-event simulation and uncertainty analysis, and combined with a multi-attribute decision-making technique so as to enable a holistic assessment. The strategies were compared across a range of scales and titres so as to visualize how their ranking changes in different industry scenarios. The deterministic analysis indicated that the ATF perfusion strategy has the potential to offer cost of goods savings of 20% when compared to conventional fed-batch manufacturing processes when a fivefold increase in maximum viable cell densities was assumed. Savings were also seen when the ATF cell density dropped to a threefold increase over the fed-batch strategy for most combinations of titres and production scales. In contrast, the fed-batch strategy performed better in terms of environmental sustainability with a lower water and consumable usage profile. The impact of uncertainty and failure rates on the feasibility of the strategies was explored using Monte Carlo simulation. The risk analysis results demonstrated the enhanced robustness of the fed-batch process but also highlighted that the ATF process was still the most cost-effective option even under uncertainty. The multi-attribute decision-making analysis provided insight into the limited use of spin-filter perfusion strategies in industry. In the present study, a series of reflectance spectra of postmortem lividity, pallor, and putrefaction-affected skin for 195 investigated cases in the course of cooling down the corpse has been collected. The reflectance spectrometric measurements were stored together with their respective metadata in a mysql database. The latter has been managed via a scientific information repository. We propose similarity measures and a criterion of similarity that capture similar spectra recorded at corpse skin. We systematically clustered reflectance spectra from the database as well as their metadata, such as case number, age, sex, skin temperature, duration of cooling, and postmortem time, with respect to the given criterion of similarity. Altogether, more than 500 reflectance spectra have been pairwisely compared. The measures that have been used to compare a pair of reflectance curve samples include the Euclidean distance between curves and the Euclidean distance between derivatives of the functions represented by the reflectance curves at the same wavelengths in the spectral range of visible light between 380 and 750 nm. For each case, using the recorded reflectance curves and the similarity criterion, the postmortem time interval during which a characteristic change in the shape of reflectance spectrum takes place is estimated. The latter is carried out via a software package composed of Java, Python, and MatLab scripts that query the mysql database. We show that in legal medicine, matching and clustering of reflectance curves obtained by means of reflectance spectrometry with respect to a given criterion of similarity can be used to estimate the postmortem interval.

According to daily scientific research and communication needs of the author's laboratory, the author puts forward a system for providing a unified platform for daily management and record of scientific research. The system contains records of daily reports, papers, patents, projects and multimedia files storage function. Compared with existing laboratory systems, the system focuses on needs of graduate students in laboratory, replaces equipment management and fund management of traditional laboratory for daily reporting and scientific research of graduate students in laboratory. The system is built on cloud platform. There are two innovations in this system. Firstly, the perspective of this paper is professional laboratory for postgraduate students. A set of systems are set to meet demands of graduate students in laboratory, which are different and at the same time have common points, so as strengthen communication between teachers and students and strengthen exchange among students, as well as improve the quality of postgraduate training. Secondly, as the system is applied in a number of laboratories, they have common features. Therefore, this system conforms to the development trend of technology, and the combination of cloud computing technology becomes preliminary exploration for the development of laboratory management system.

Due to rapid socioeconomic development in recent years, the influence of human activities on the urban ecosystem and environment is becoming more pronounced, causing increases in accumulated pollutants, resource consumption, and built-up land area. At this time, cities are facing complex ecological threats. We designed a platform to classify and assess the various types of ecological risks that cities and urban agglomerations may encounter. We used PHP to develop a web-based application, with nginx as the platform server, and mysql to manage the background database. The platform provides users with a clearly structured, visual platform to manage ecological risk. By using this platform, users can quickly identify regional risk sources, find and describe risk factors, and predict possible consequences caused by these risks based on simulation modeling. The forecasting model and database can be synchronously updated and adjusted according to the actual situation. The visual web management platform and user security login system employ the MD5 encryption algorithm. The platform is designed to provide convenient, accurate, and scientific decision-making services for urban planning, construction, environmental protection, and other related areas.

Development of an Accounting Skills Simulation Practice System Based on the B/S Architecture

Nowadays, companies are more in need of accounting professionals with both theoretical knowledge and practical abilities. However, the traditional accounting teaching model in colleges and universities cannot train students in both aspects. In order to improve the practical abilities of accounting major students in colleges and universities and promote the accounting teaching upgrade and reform, this paper proposes an accounting skills simulation practice system based on the B/S architecture, aiming at improving the comprehensive abilities of accounting students and teaching efficiency. The technical roadmap of this system adopts the B/S as the network architecture of the teaching system, HTML and Java Script language as the system programming language and mysql database as the data storage and management database. The system consists of user administration, knowledge base management, teaching content management, practical test score management and other modules, which not only achieves the self-evaluation on the full set of accounting skills, but also improves the accounting teaching efficiency and helps teachers gain more control over the teaching process. This study can serve as some guidance to colleges and universities in cultivating innovative and practical accounting talents.

This paper focuses on the design of methods which enable unlicensed devices to use the frequency spectrum already allocated to rotating radar systems without causing interference to those radars. We first present a database-assisted spectrum sharing/co-existence mechanism for radar spectrum bands. This mechanism takes into account the real spectrum usage behavior of different radars, and is also implemented by prototyping a real-time rotating radar beam emulator, a radar spectrum database using mysql software, and spectrum access algorithm on wireless open access research platform. Then we propose a cloud-based unified channel access (UCA) method and a distributed UCA method for co-existence of multiple competing users in unlicensed and rotating radar spectrum bands. We model the UCA problem as a game and propose iterative methods (i.e., algorithms) to obtain the solution of the game. We study the stability of the proposed methods under several different scenarios. We show that the proposed UCA game is a potential game, and also show that the proposed methods guarantee convergence to a Nash equilibrium. Our results show that the cloud-based method allows fast convergence and can achieve performance that is close to optimal solution.

Deep foundation pits in coastal areas have the characteristics of high groundwater level, strong formation permeability, and large changes in foundation pit water level. The real-time monitoring of the water level of the foundation pit is an important basis for ensuring the safe operation of the foundation pit project. Traditional monitoring methods have disadvantages such as unstable equipment, high cost, and poor scalability. Therefore, in this study, a set of online monitoring system for deep foundation pit water level was developed by combining Raspberry Pi and IoT technology. The system mainly consists of data acquisition layer, data management layer, equipment control layer and data display layer, and has good expansibility. By embedding self-compiled Python Program in Raspberry Pi, the system can be controlled remotely, and the water level of foundation pit can be collected, stored and transmitted in real time and effectively. At the same time, the foundation pit pumping is controlled according to the water level of the foundation pit, so as to realize the fine control of the foundation pit pumping and achieve the effect of energy saving and emission reduction. Based on relational database mysql and B/S architecture, it realizes centralized data management, calculation and analysis, charting and other functions. The practical application test shows that the system has good stability, reliability and expansibility, improves the water level monitoring efficiency of the deep foundation pit in the coastal area, and provides a guarantee for the safe construction of the project.

In this study, we present a quality control system devoted for radiotherapy services. Cancer treatment field concerns also Chemotherapy, Surgery and Other but they didn't do the object of this work. The Cancer is an enormous and growing global public health problem. The majority of cancers are treated in the radiotherapy (or radiation oncology) services. The radiation oncology includes the diagnostic imaging involving the safe use of ionizing radiation and nuclear medicine procedures. The strategies for treatment management and terminal cure must be established in a comprehensive and multidisciplinary approach for a total cancer care program. The system taken as an example consists of a set of 6 personnel computers (PC's), 3 linear accelerators (VARIAN model), 2 scanner simulators (SIEMENS and General Electric). Currently the problem of the quality services and the accidents in radiotherapy services in the world is not solved. This system is used in order to improve the quality of services and to increase the medical care. Its main purpose is to avoid mistake's treatment which could kill patients. Accidents occur till now even in developed countries (USA, France, England, etc). This project is in its first step. Some results are obtained and described in this study. The PC's are put in a network and five of them are used to control the check-list of the apparatus (linear accelerators and scanner simulators). One PC is used as a data server in order to store all data of the system (the check-list of the devices, the temperature and the moisture data, physicians and patients' management).
