Organic Nanoflowers from a Wide Variety of Molecules Templated by a Hierarchical Supramolecular Scaffold

Nanoflowers (NFs) are flowered-shaped particles with overall sizes or features in the nanoscale. Beyond their pleasing aesthetics, NFs have found a number of applications ranging from catalysis, to sensing, to drug delivery. Compared to inorganic based NFs, their organic and hybrid counterparts are relatively underdeveloped mostly because of the lack of a reliable and versatile method for their construction. We report here a method for constructing NFs from a wide variety of biologically relevant molecules (guests), ranging from small molecules, like doxorubicin, to biomacromolecules, like various proteins and plasmid DNA. The method relies on the encapsulation of the guests within a hierarchically structured particle made from supramolecular G-quadruplexes. The size and overall flexibility of the guests dictate the broad morphological features of the resulting NFs, specifically, small and rigid guests favor the formation of NFs with spiky petals, while large and/or flexible guests promote NFs with wide petals. The results from experiments using confocal fluorescence microscopy, and scanning electron microscopy provides the basis for the proposed mechanism for the NF formation.

Nodular fasciitis (NF) is a clonal self-limited neoplastic proliferation characterized by rearrangements of the USP6 locus in most examples. To our knowledge well-documented malignant behavior has never been previously observed in NF. In this report we present an unusual case of NF with classical histologic features that showed a protracted clinical course characterized by multiple recurrences and eventual metastatic behavior over a period of 10 years. Molecular analyses revealed the presence and amplification of the novel PPPR6-USP6 gene fusion, which resulted in USP6 mRNA transcriptional upregulation. These findings further support the oncogenic role of the USP6 protease in mesenchymal neoplasia and expand the biologic potential of NF. (C) 2016 Wiley Periodicals, Inc.

network function virtualization (NFV) brings significant benefits by outsourcing network functions (NFs) to virtualized datacenters and enabling new business models, e.g. NF as a service (NFaaS). In this respect, network service embedding (i.e., the mapping of service chains) raises significant challenges due to the need to preserve correctness and satisfy NF location dependencies. The latter creates the need for service mapping across multiple NF Providers (NFPs), given the limited footprint of each NFP. This further exacerbates the network service embedding (NSE) problem, due to the NFPs' local policies and the confidentiality of network topology and resource availability. To address these issues, we present DistNSE, a distributed NSE framework that enables the collaboration among NFPs for NSE, while maintaining the privacy and autonomy of each NFP. DistNSE ensures competitive pricing by letting different NFPs compete for different NFs of a service chain. To this end, DistNSE decomposes NSE in two steps: (i) service chain partitioning using an algorithm that fosters competition across NFPs, and (ii) the assignment of chain segments to datacenter networks, while ensuring compliance with the NFP's policy. We further couple the proposed embedding methods with a communication protocol for interoperability and collaboration among the participating NFPs. We use simulations to assess the efficiency of DistNSE and identify significant gains over an existing distributed embedding framework (i.e., Polyvine).

Selective Nucleation of Poly(3-hexyl thiophene) Nanofibers on Multilayer Graphene Substrates

We demonstrate that graphene surfaces provide highly selective nucleation of poly(3-hexyl thiophene) (P3HT) nanofibers (NFs) from supersaturated solutions. Solvent conditions are identified that give rise to a wide hysteresis between crystallization and melting centered around room temperature, yielding metastable solutions that are stable against homogeneous nucleation for long periods of time but that allow for heterogeneous nucleation by graphene. Selective growth of P3HT crystals is found for multilayer graphene (MLG) supported on either Si or ITO substrates, with nucleation kinetics that are more rapid for MLG on Si but slower in both cases than for highly oriented pyrolytic graphite (HOPG). Although the NFs grow vertically from the substrate with face-on orientation of P3HT chains, we observe edge-on orientation in dried films, presumably due to capillary forces that cause collapse of the NFs onto the substrate during solvent evaporation.

Scalable network file systems with load balancing and fault tolerance for web services

Because of the rapid growth of the World Wide Web and the popularization of smart phones, tablets and personal computers, the number of web service users is increasing rapidly. As a result, large web services require additional disk space, and the required disk space increases with the number of web service users. Therefore, it is important to design and implement a powerful network file system for large web service providers. In this paper, we present three design issues for scalable network file systems. We use a variable number of objects within a bucket to decrease internal fragmentation in small files. We also propose a free space and access load-balancing mechanism to balance overall loading on the bucket servers. Finally, we propose a mechanism for caching frequently accessed data to lower the total disk I/O. These proposed mechanisms can effectively improve scalable network file system performance for large web services. (C) 2014 Elsevier Inc. All rights reserved.

The deployment of micro-datacenters for network function virtualization (NFV) by Internet Service Providers creates opportunities for flow processing along the traffic path. On-path processing requires the discovery of the middle boxes that will be traversed by each flow and the assignment of network functions (NFs) to middleboxes, while preserving the order of the NFs as specified in the service chain. NF location dependencies may require flow processing establishment across multiple NF Providers (NFPs). This entails additional challenges for middle box discovery and selection, stemming from the NFPs' restrictions in information disclosure and interoperability. To address these issues, we present MIDAS, an architecture for the coordination of middle box discovery and selection across multiple NFPs. MIDAS relies on a centralized middle box controller in each NFP to provide interoperability among NFPs for flow processing setup. MIDAS establishes on-path processing via middlebox signaling, controller chaining, and Multi-Party Computation (MPC) based middle box selection. We particularly employ MPC to preserve the confidentiality of middlebox utilization across the NFPs. We study the feasibility of MIDAS using a prototype implementation and further present simulation results to assess the efficiency of our middle box selection approach.

network Service Chaining (NSC) is a service concept which promises increased flexibility and cost-efficiency for future carrier networks. The two recent developments, network Function Virtualization (NFV) and Software-Defined networking (SDN), are opportunities for service providers to simplify the service chaining and provisioning process and reduce the cost (in CAPEX and OPEX) while introducing new services as well. One of the challenging tasks regarding NFV-based services is to efficiently map them to the components of a physical network based on the services specifications/constraints. In this paper, we propose an efficient cost-effective algorithm to map NSCs composed of network Functions (NF) to the network infrastructure while taking possible decompositions of NFs into account. NF decomposition refers to converting an abstract NF to more refined NFs interconnected in form of a graph with the same external interfaces as the higher-level NF. The proposed algorithm tries to minimize the cost of the mapping based on the NSCs requirements and infrastructure capabilities by making a reasonable selection of the NFs decompositions. Our experimental evaluations show that the proposed scheme increases the acceptance ratio significantly while decreasing the mapping cost in the long run, compared to schemes in which NF decompositions are selected randomly.

Kirchhoff Pre-Stack Depth Migration (KPSDM), a widely used seismic imaging algorithm in petroleum industry, is a typical IO-bound application since a large amount of seismic data and travel timetable data needs to be read from the storage system iteratively during runtime. We present an optimized high performance KPSDM implementation called HKPSDM based on Hadoop where a large I/O aggregated bandwidth is offered cheaply to replace our previous implementation based on network Attached Storage (NAS) appliances over NFS which have a limited I/O bandwidth when hundreds of processes participate in a migration job. In our implementation, MapReduce facilitates the travel timetable rearrangement and HDFS provides a stable and scalable storage system for seismic data as well as travel timetable data. Various optimizations are applied to HKPSDM to reduce the global bandwidth consumption of HDFS and overhead caused by some disadvantages of HDFS. Experimental results show that HKPSDM can scale better when the number of computing cores ranges from 160 to 800. HKPSDM performs more than 5 times better than NAS-based one when rearranging the travel timetables. And a 37% efficiency improvement is observed when 800 cores are used for migration.

The popular network file system (NFS) protocol is 30 years old. The latest version, NFSv4, is more than ten years old but has only recently gained stability and acceptance. NFSv4 is vastly different from its predecessors: it offers a stateful server, strong security, scalability/WAN features, and callbacks, among other things. Yet NFSv4's efficacy and ability to meet its stated design goals had not been thoroughly studied until now. This paper compares NFSv4.1's performance with NFSv3 using a wide range of micro- and macro-benchmarks on a testbed configured to exercise the core protocol features. We (1) tested NFSv4's unique features, such as delegations and statefulness; (2) evaluated performance comprehensively with different numbers of threads and clients, and different network latencies and TCP/IP features; (3) found, fixed, and reported several problems in Linux's NFSv4.1 implementation, which helped improve performance by up to 11X; and (4) discovered, analyzed, and explained several counter-intuitive results. Depending on the workload, NFSv4.1 was up to 67\% slower than NFSv3 in a low-latency network, but exceeded NFSv3's performance by up to 2.9X in a high-latency environment. Moreover, NFSv4.1 outperformed NFSv3 by up to 172X when delegations were used.

Discrimination of Pterocephalus hookeri collected at flowering and non-flowering stages using GC-MS-based fatty acid profiling

For a herbal medicine, harvesting at different growth stage affects its quality and efficiency. We hypothesized that fatty acid profiling might be used to discriminate herbal samples according to their growth stages. To test the hypothesis, fatty acids of Pterocephalus hookeri samples collected at the flowering stage (FS) and non-flowering stages (NFS) were characterized and compared using gas chromatography-mass spectrometry (GC-MS) and followed by multivariate statistical analysis. A total of 14 fatty acids were identified and quantified in all the P. hookeri samples. Both the relative and absolute compositions of the 14 fatty acids varied greatly between the FS and NFS groups, suggesting each group has its own fatty acid pattern. Orthogonal partial least squares discriminant analysis (OPLS-DA) and hierarchical clustering analysis (HCA) based on data sets of relative and absolute composition of fatty acids showed that 13 tested samples could be clearly classified into two clusters, in terms of their growth stages. More importantly, a-linolenic acid, a plant-derived n-3 polyunsaturated fatty acid (PUFA), was identified as the potential fatty acid biomarker for its greatest contribution to the group's separation. In addition, to evaluate the quality of P. hookeri at the FS and NFS, oleanolic acid (OA) and ursolic acid (UA) were determined by HPLC, as described in the Chinese Pharmacopoeia (version 2010). A higher total concentration of OA and UA could be found in the P. hookeri samples at the flowering stage, which was suggested to be a better quality. These findings demonstrated that GC-MS-based fatty acid profiling, coupled with multivariate statistical analysis, provides a reliable platform to discriminate the herb collected at different growth stages, which is helpful for ensuring its efficacy.
