The Small non-coding Vault rna1-1 Acts as a Riboregulator of Autophagy

Vault rnas (vtrna) are small non-coding rnas transcribed by rna polymerase III found in many eukaryotes. Although they have been linked to drug resistance, apoptosis, and viral replication, their molecular functions remain unclear. Here, we show that vault rnas directly bind the autophagy receptor sequestosome-1/p62 in human and murine cells. Overexpression of human vtrna1-1 inhibits, while its antisense LNA-mediated knockdown enhances p62-dependent autophagy. Starvation of cells reduces the steady-state and p62-bound levels of vault rna1-1 and induces autophagy. Mechanistically, p62 mutants that fail to bind vtrnas display increased p62 homo-oligomerization and augmented interaction with autophagic effectors. Thus, vtrna1-1 directly regulates selective autophagy by binding p62 and interference with oligomerization, a critical step of p62 function. Our data uncover a striking example of the potential of rna to control protein functions directly, as previously recognized for protein-protein interactions and post-translational modifications.

A large body of evidence has highlighted the role of non-coding rnas in neurodevelopment and *** evidence has led to increasing speculation that non-coding rnas may be involved in the pathophysiological mechanisms underlying hydrocephalus,one of the most common neurological conditions *** this review,we first outline the basic concepts and incidence of hydrocephalus along with the limitations of existing treatments for this ***,we outline the definition,classification,and biological role of non-coding ***,we analyze the roles of non-coding rnas in the formation of hydrocephalus in ***,we have focused on the potential significance of non-coding rnas in the pathophysiology of hydrocephalus,including glymphatic pathways,neuroinflammatory processes,and neurological dysplasia,on the basis of the existing ***,we review the potential of non-coding rnas as biomarkers of hydrocephalus and for the creation of innovative treatments.

Ovarian cancer (OC) is the most fatal gynecological malignancy because of its early asymptomatic nature and acquired resistance to chemotherapy. Small extracellular vesicles (sEVs) are a heterogeneous group of biological vesicles with a diameter <200 nm released by cells under physiological or pathological conditions. sEVs-derived non-coding rnas (ncrnas) are the essential effectors in the biological environment. sEVs-ncrnas have critical roles in tumor progression via regulating mrna expression of target cells to affect cell signaling. In addition, the status of parental cells can be disclosed via analyzing the composition of sEVs-ncrnas, and their "cargoes" with specific changes can be used as key biomarkers for the diagnosis and prognosis of OC. Accumulating evidence has demonstrated that sEVs-ncrnas are involved in multiple key processes that mediate the development of metastasis and chemotherapeutic resistance in OC: epithelial-mesenchymal transition;tumorigenicity of mesenchymal stem cells;immune evasion;angiogenesis. The nanomedicine delivery system based on engineering sEVs is expected to be a novel therapeutic strategy for OC. Insights into the biological roles of sEVs-ncrnas in the invasion, metastasis, immune regulation, and chemoresistance of OC will contribute to discovery of novel biomarkers and molecular targets for early detection and innovative therapy. In this review, we highlight recent advances and applications of sEVs-ncrnas in OC diagnosis and treatment. We also outline current challenges and knowledge gaps.

Cardiovascular diseases are the leading cause of death worldwide. They are non-transmissible diseases that affect the cardiovascular system and have different etiologies such as smoking, lipid disorders, diabetes, stress, sedentary lifestyle and genetic factors. To date, lncrnas have been associated with increased susceptibility to the development of cardiovascular diseases such as hypertension, acute myocardial infarction, stroke, angina and heart failure. In this way, lncrnas are becoming a very promising point for the prevention and diagnosis of cardiovascular diseases. Therefore, this review highlights the most important and recent discoveries about the mechanisms of action of the lncrnas ANRIL, H19 and TUG1 and their clinical relevance in these pathologies. This may contribute to early detection of cardiovascular diseases in order to prevent the pathological phenotype from becoming established.

CRISPR-mediated silencing of non-coding rnas: A novel putative treatment for prostate cancer

non-coding rnas affect carcinogenic processes in diverse tissues, such as prostate. Several of these transcripts act as oncogenes driving prostate cancer. Thus, they are putative targets for treatment of this type of cancer. CRISPR/Cas9 technology has provided new tools for modulation of expression of these oncogenes in order to combat several aspects of carcinogenesis, including invasion cascades and metastasis. This review aimed to describe novel achievements in modulation of expression of non-coding rnas using CRISPR/Cas9 technology in prostate cancer.

In recent years, ischemic stroke (IS) has been one of the major causes of disability and mortality worldwide. The general mechanism of IS is based on reduced blood supply to neuronal tissue, resulting in neuronal cell damage by various pathological reactions. One of the main techniques for acute IS treatment entails advanced surgical approaches for restoration of cerebral blood supply but this is often associated with secondary brain injury, also known as ischemic reperfusion injury (I/R injury). Many researches have come to emphasize the significant role of long non-coding rnas (lncrnas) in IS, especially in I/R injury and their potential as therapeutic approaches. Lncrnas are non-protein transcripts that are able to regulate cellular processes and gene expression. Further, lncrnas have been shown to be involved in neuronal signaling pathways. Several lncrnas are recognized as key factors in the physiological and pathological processes of IS. In this review, we discuss the role of lncrnas in neuronal injury mechanisms and their association with brain neuroprotection. Moreover, we identify the lncrnas that show the greatest potential as novel therapeutic approaches in IS, which therefore merit further investigation in preclinical research.

Prognostic significance of non-coding rnas related to the tumorigenic epithelial-mesenchymal transition (EMT) process among ovarian cancer patients: A systematic review and meta-analysis

Introduction: Ovarian cancer is the seventh most prevalent cancer among women. It has high mortality and morbidity and imposes a great burden on healthcare systems worldwide. Unraveling the mechanisms behind the Epithelial-Mesenchymal Transition and finding a panel for predicting the prognosis of the disease may help find the appropriate treatment approaches for the management of the disease. The overarching aim of this systematic review was to define a panel of different types of EMT-associated non-coding rnas (ncrnas) with significant prognostic value in all types of ovarian cancers. Methods: We searched PubMed, Web of Science, Scopus, and Embase till Jun 2024 to retrieve relevant papers. Two independent reviewers screened papers, and discrepancies were resolved by consensus. Publications related to the dysregulation of different types of ncrnas, including micrornas, lncrnas, and circrnas, only in patients with ovarian cancer were included. The participation of ncrnas in epithelial-mesenchymal transformation should be assessed via methods evaluating different EMT-related proteins. To assess the quality and risk of bias for the included case-control and cohort studies, refined Newcastle-Ottawa Scale (NOS) and Quadas-2 were recruited. A bivariate meta-analysis was performed to analyze extracted data. Results: A total of 37 studies with overall 42 non-coding rnas (15 microrna, 24 long non-coding rnas, and 3 circular rnas) were entered into the analysis. Overall diagnostic odds ratio for ncrnas in lymph node metastasis, distant metastasis, TNM stage, and clinical stage were 4.19, 3.80, 6.52, and 3.97, respectively. Also, a hazard ratio of 1.39 (P = 0.32) for overall survival was observed. Bioinformatic analyses on the Pan-cancer database demonstrated a significant correlation between low expression of mirna and high expression of lncrnas with poor prognosis of ovarian cancer. Epigenetic regulation of switchgrass ( Panicum virgatum L.) in multiple drought stresses

Drought memory serves as an adaptive mechanism that plants have evolved to endure drought stress. Preliminary research has identified drought stress memory in the seedling stages of switchgrass. However, current research on the epigenetic regulation of switchgrass under multiple drought stresses is limited. Here, we revealed the epigenetic regulation of drought memory genes in switchgrass through the data analysis of high-throughput rna-seq and Whole-Genome Bisulphite Sequencing (WGBS) under multiple drought stresses. In this study, we identified 2388 drought memory genes, 48 memory mirnas, 702 memory lncrnas, and three memory circrnas, and constructed a competing endogenous rna (cerna) network of differentially expressed (DE) lncrnasDE mirnas-memory genes. WGBS data showed that methylated regions were found on the promoter and gene body of 2315 memory genes (96.94 %), and the expression of memory genes showed a correlation with the changes in methylation level on the gene body. By integrating the data from high-throughput rna-seq and WGBS, further analysis found that drought memory genes and their epigenetic regulation participated in the lignin biosynthesis, as well as biosynthesis and signal transduction of abscisic acid (ABA), auxin (IAA), brassinosteroids (BR) and jasmonic acid (JA). In summary, a comprehensive network involving non-coding rna (ncrna), DNA methylation, and the regulation of memory genes has been systematically constructed in switchgrass under multiple drought stresses, which provides theoretical basis and genetic resources for the thorough analysis of the complex mechanisms of drought memory formation in switchgrass and other plants.

Viral non-coding rnas: Stealth strategies in the tug-of-war between humans and herpesviruses

Oncogenic DNA viruses establish lifelong infections in humans, and they cause cancers, often in immunocom-promised patients, despite anti-viral immune surveillance targeted against viral antigens. High-throughput sequencing techniques allowed the field to identify novel viral non-coding rnas (ncrnas). ncrnas are ideal factors for DNA viruses to exploit;they are non-immunogenic to T cells, thus viral ncrnas can manipulate host cells without evoking adaptive immune responses. Viral ncrnas may still trigger the host innate immune response, but many viruses encode decoys/inhibitors to counter-act and evade recognition. In addition, ncrnas can be secreted to the extracellular space and influence adjacent cells to create a pro-viral microenvironment. In this review, we present recent progress in understanding interactions between oncoviruses and ncrnas including small and long ncrnas, micrornas, and recently identified viral circular rnas. In addition, potential clinical applications for ncrna will be discussed. Extracellular ncrnas are suggested to be diagnostic and prognostic biomarkers and, with the realization of the importance of viral ncrnas in tumorigenesis, approaches to target critical viral ncrnas are emerging. Further understanding of viral utilization of ncrnas will advance anti-viral therapeutics beyond conventional medication and vaccination.
