Advances in research regarding the roles of non-coding rnas in non-syndromic cleft lip with or without cleft palate: A systematic review

Objective: To investigate by means of a literature review, what non-coding rnas engage in non-syndromic cleft lip with or without cleft palate (NSCL/P) and how they lead to the occurrence of this ***: A literature search of online databases (Medline via PubMed, Web of Science, Scopus, and Embase) was performed using appropriate keywords (e.g. non-coding rna, mirna, lncrna, NSCL/P, non-syndromic cleft lip only, and non-syndromic orofacial cleft). The risk of bias in the included studies was then assessed, and a comprehensive review of reported non-coding rnas associated with NSCL/P was ***: The initial search retrieved 133 studies reporting non-coding rnas associated with NSCL/P;after excluding 18 replicates and 77 ineligible studies, 35 remained. Of these, 16 studies fulfilled all the criteria and were included in the systematic review. These studies established the roles of non-coding rnas in the development of craniofacial structures. The differential expression of these non-coding rnas could lead to orofacial clefts, indicating their significance in NSCL/P and their profound research value. Conclusion: There is evidence that non-coding rnas are involved in the formation of NSCL/P. Specifically, they play significant roles in the regulation of genes and signalling pathways related to NSCL/P.

Role of Hedgehog and Hippo signaling pathways in cancer: A special focus on non-coding rnas

Despite advanced clinical and translational oncology research, mortality rates are still increasing worldwide. Recently, a class of non-coding rnas (ncrnas), such as micrornas (mirnas), long non-coding rnas (lncrnas), and circular rnas (circrnas), have been well investigated in regulating biological, molecular, and cellular signaling pathways. This review article provided the current research progress on how mirnas, lncrnas, and circrnas regulate Hedgehog (Hh) and Hippo signaling pathways in various cancers. These ncrnas target both pathways' key downstream molecules and may be used for targeted cancer treatment. Moreover, Hh and Hippo signaling pathways crosstalked with each other through Gli1 of Hh pathways and YAP1/TEAD molecules of Hippo pathways during cancer progression. Additionally, Hh and Hippo signaling pathways regulate resistance against the chemo, radio, and immune therapies for several types of cancer via inducing GLI and YAP/TAZ proteins level. Therefore, to improve the treatment regime, we presented the role of various prominent phyto-chemicals such as curcumin, resveratrol, genistein, quercetin, paclitaxel, and silibinin in regulating lncrnas, mirnas, circrna through Hedgehog and Hippo signaling pathways' constituents in cancers. We believe that knowledge obtained from this review may help make new drugs for cancer treatment in the future.

Microrna in cancer therapy: breakthroughs and challenges in early clinical applications

Micrornas (mirnas) have emerged as pivotal regulators in cancer biology, influencing tumorigenesis, progression, and resistance to therapy. Their ability to modulate multiple oncogenic and tumor-suppressive pathways positions them as promising therapeutic tools or targets. This review examines the dual role of mirnas in solid and hematological malignancies, starting from their dysregulation in various cancer types. Therapeutic approaches, including mirna replacement and inhibition strategies, are discussed alongside innovative delivery systems such as lipid nanoparticles and exosomes. Despite their transformative potential, challenges persist, including off-target effects, immune activation, and delivery inefficiencies. Recent clinical trials demonstrate both progress and hurdles, underscoring the need for advanced strategies to optimize specificity and minimize toxicity. This review provides an updated comprehensive overview of the current landscape of mirna-based therapies under early clinical investigation and explores future directions for integrating these approaches into precision oncology.

IntroductionProstate cancer is a serious threat to the health of older adults worldwide. The quality of life and survival time of patients sharply decline once metastasis occurs. Thus, early screening for prostate cancer is very advanced in developed countries. The detection methods used include Prostate-specific antigen (PSA) detection and digital rectal examination. However, the lack of universal access to early screening in some developing countries has resulted in an increased number of patients presenting with metastatic prostate cancer. In addition, the treatment methods for metastatic and localized prostate cancer are considerably different. In many patients, early-stage prostate cancer cells often metastasize due to delayed observation, negative PSA results, and delay in treatment time. Therefore, the identification of patients who are prone to metastasis is important for future clinical *** coveredthis review introduced a large number of predictive molecules related to prostate cancer metastasis. Extracellular vesicles: The next generation in gene therapy delivery

Extracellular vesicles (EVs) are esteemed as a promising deliv-ery vehicle for various genetic therapeutics. They are relatively inert, non-immunogenic, biodegradable, and biocompatible. At least in rodents, they can even transit challenging bodily hurdles such as the blood-brain barrier. Constitutively shed by all cells and with the potential to interact specifically with neighboring and distant targets, EVs can be engineered to carry and deliver therapeutic molecules such as proteins and rnas. EVs are thus emerging as an elegant in vivo gene therapy vector. Deeper understanding of basic EV biology-including cellular production, EV loading, systemic distribution, and cell deliv-ery-is still needed for effective harnessing of these endogenous cellular nanoparticles as next-generation nanodelivery tools. However, even a perfect EV product will be challenging to pro-duce at clinical scale. In this regard, we propose that vector transduction technologies can be used to convert cells either ex vivo or directly in vivo into EV factories for stable, safe mod-ulation of gene expression and function. Here, we extrapolate from the current EV state of the art to a bright potential future using EVs to treat genetic diseases that are refractory to current therapeutics.

Objectives: In contrast to mirna expression, little attention has been given to piwirna (pirna) expression among endometriosis patients. The aim of the present study was to explore the human pirnaome and to investigate a potential pirna saliva-based diagnostic signature for ***: Data from the prospective "ENDOmirna" study (*** Identifier: NCT04728152) were used. Saliva samples from 200 patients were analyzed in order to evaluate human pirna expression using the pirna bank. Next Generation Sequencing (NGS), barcoding of unique molecular identifiers and both Artificial Intelligence (AI) and machine learning (ML) were used. For each pirna, sensitivity, specificity, and ROC AUC values were calculated for the diagnosis of ***: 201 pirnas were identified, none had an AUC > 0.70, and only three pirnas (piR-004153, piR001918, piR-020401) had an AUC between > 0.6 and < 0.70. Seven were differentially expressed: piR-004153, piR001918, piR-020401, piR-012864, piR-017716, piR-020326 and piR-016904. The respective correlation and accuracy to diagnose endometriosis according to the F1-score, sensitivity, specificity, and AUC ranged from 0 to 0.862 %, 0-0.961 %, 0.085-1, and 0.425-0.618. A correlation was observed between the patients' age (>35 years) and piR-004153 (p = 0.002) and piR-017716 (p = 0.030). Among the 201 pirnas, four were differentially expressed in patients with and without hormonal treatment: piR-004153 (p = 0.015), piR-020401 (p = 0.001), piR-012864 (p = 0.036) and piR-017716 (p = 0.009).Conclusion: Our results support the link between pirnas and endometriosis physiopathology and establish its utility as a potential diagnostic biomarker using saliva samples. Per se, pirna expression should be analyzed along with the clinical status of a patient.

Centromeric and pericentric transcription and transcripts: their intricate relationships, regulation, and functions

Centromeres are no longer considered to be silent. Both centromeric and pericentric transcription have been discovered, and their rna transcripts have been characterized and probed for functions in numerous monocentric model organisms recently. Here, we will discuss the challenges in centromere transcription studies due to the repetitive nature and sequence similarity in centromeric and pericentric regions. Various technological breakthroughs have helped to tackle these challenges and reveal unique features of the centromeres and pericentromeres. We will briefly introduce these techniques, including third-generation long-read DNA and rna sequencing, protein-DNA and rna-DNA interaction detection methods, and epigenomic and nucleosomal mapping techniques. Interestingly, some newly analyzed repeat-based holocentromeres also resemble the architecture and the transcription behavior of monocentromeres. We will summarize evidences that support the functions of the transcription process and stalling, and those that support the functions of the centromeric and pericentric rnas. The processing of centromeric and pericentric rnas into multiple variants and their diverse structures may also provide clues to their functions. How future studies may address the separation of functions of specific centromeric transcription steps, processing pathways, and the transcripts themselves will also be discussed.

noncoding rnas are typically more than 200 nucleotides in lengths with indiscernible protein coding potential defined as long non-coding rna (lncrnas). Lncrnas play a versatile role in plant growth, development, and responses to various ecological perturbations particularly cold. However, a comprehensive identification of lncrnas responsive to cold stress in B. napus has not been identified. The aim of the present study was to identify potential lncrnas in cold tolerant (CT-C18) and cold-sensitive (CS-C6) B. napus varieties under cold stress. A total of 11073 lncrnas were identified in both varieties of Brassica. In CT-C18 variety, 2394 and 3508 lncrnas were expressed at 1d and 7d after cold stress, respectively. Furthermore, 3347 and 3872 lncrnas were expressed at 1d and 7d in CS-C6 variety, respectively. Fifteen lncrnas were found to be target mimics of 23 mirnas. Additionally, a total of 16 lncrnas were predicted as precursors of B. napus mirnas seven mirna family. Moreover, weighted gene co-expression network analysis revealed that four modules correlate with the expression status of protein-coding genes with lncrnas. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analysis shown that out of those 170 gene were tagged with 60 Enzyme Code, and 101 GO terms. Our study increased the understanding of the expression and characterization of B. napus lncrnas in response to cold stress, which may use as a reference for functional-based studies of targeting putative lncrnas particularly under cold stress.

piwirna is a kind of non-coding rna (ncrna) that cannot be translated into proteins. It helps in understanding the study of gametes generation and regulation of gene expression over both transcriptional and post-transcriptional levels. piwirna has the function of instructing deadenylation, animal fertility, silencing transposons, fighting viruses, and regulating endogenous genes. Due to the great significance of piwirna, prediction of piwirna is essential for crucial cellular functions. Several predictors were established for prediction of piwirna. However, improving the prediction of piwirna is highly desirable. In the current study, we developed a more promising predictor named, BLP-piwirna. The features are explored by reverse complement k-mer, gapped-k-mer composition, and k-mer composition. The feature set of all descriptors is fused and the best features are selected by cascade and relief feature selection strategies. The best feature sets are provided to random forest (RF), deep neural network (DNN), and support vector machine (SVM). The models validation are examined by 10-fold test. DNN with optimal features of Cascade feature selection approach secured the highest prediction results. The results illustrate that BLP-piwirna effectively outperforms the existing studies. The proposed approach would be beneficial for both research community and drug development industry. BLP-piwirna would serve as novel biomarkers and therapeutic targets for tumor diagnostics and treatment.
