Emerging Involvement of long non-coding rnas in gastrointestinal associated inflammatory disorders

Gastrointestinal (GI) disorders including a wide range of infectious, inflammatory, autoimmune, etc. disorders. Inflammatory bowel and celiac disease are non-fatal but overwhelming GI associated disorders. IBD and celiac's complications, besides the great suffering, disturb the normal life of the patients and make them involved in mental and physical problems. The emerging role of genetic content is deniable for GI inflammatory disorders incidence, and long non-coding rnas (lncrnas) function is the recent topic for its association. Analyzing of absolute lncrnas interference in GI inflammatory appearance remains in infancy, and more studies are requested. Here, we concisely performed a systematic review in the last knowledge up to 2020 to identify all of the significant lncrnas associated with the initiation and progression of GI inflammatory diseases. Accordingly, this assay attempted to refer to the expression of lncrnas changing from the normal state, discovery of genetic mechanisms, and main effectors that would trigger associated IBD and celiac expression and immune responses would be effective for therapeutic approaches. It could be useful for prognostic and diagnostic purposes of GI associated inflammatory disorders.

rna transcripts that not undergo translation into polypeptides recently came into focus of research. Long non-coding rnas (lncrnas), micrornas (mirnas) and circular rnas (circrnas) comprise the most important groups of these transcripts. Lncrnas have a length over 200 nucleotides and like mrnas, have regulated transcription in a tissue specific manner. Biogenesis and function of lncrnas is related to cell differentiation, response to stimuli and regulation of immune responses. Lncrnas can interact with both mirnas and mrnas. Mirnas are characterized by a length of 22-24 nucleotides. Mirnas regulate expression of genes at the post-transcriptional level. Lncrnas together with mirnas are considered as regulators of the immune system. Alterations in their biogenesis is an important mechanism in the development immune related disorders. Circrnas are products of aberrant maturation of protein-coding transcripts in a process of back-splicing, in which a single strand rna molecule attains a closed circle shape. Despite a low expression, some circrna were found to titrate mirnas and interfere with maturation of legitimate protein-coding transcripts. We summarize the current knowledge on the role of non-coding transcripts in allergic disorders: asthma, atopic dermatitis, allergic rhinitis and urticaria. The reviewed data suggest lncrna and mirnas as therapeutic targets and biomarkers of allergic disorders.

Micrornas and long non-coding rnas as novel regulators of ribosome biogenesis

Ribosome biogenesis is the fine-tuned, essential process that generates mature ribosomal subunits and ultimately enables all protein synthesis within a cell. Novel regulators of ribosome biogenesis continue to be discovered in higher eukaryotes. While many known regulatory factors are proteins or small nucleolar ribonucleoproteins, micrornas (mirnas), and long non-coding rnas (lncrnas) are emerging as a novel modulatory layer controlling ribosome production. Here, we summarize work uncovering non-coding rnas (ncrnas) as novel regulators of ribosome biogenesis and highlight their links to diseases of defective ribosome biogenesis. It is still unclear how many mirnas or lncrnas are involved in phenotypic or pathological disease outcomes caused by impaired ribosome production, as in the ribosomopathies, or by increased ribosome production, as in cancer. In time, we hypothesize that many more ncrna regulators of ribosome biogenesis will be discovered, which will be followed by an effort to establish connections between disease pathologies and the molecular mechanisms of this additional layer of ribosome biogenesis control.

The final steps in the Renin-Angiotensin-Aldosterone signaling System (RAAS) involve binding of the corticosteroid hormone, aldosterone to its mineralocorticoid receptor (MR). The bound MR interacts with response elements to induce or repress the transcription of aldosterone-regulated genes. Along with the classic genomic targets of aldosterone that alter mrna and protein expression, aldosterone also regulates the expression of noncoding rnas (ncrnas). Short ncrnas termed micrornas (miRs) have been shown to play a role in transducing aldosterone's actions via MR signaling. The role of miRs in homeostatic regulation of aldosterone signaling, and the potential for aldosterone-regulated miRs to act as feedback regulators of MR have been recently reported. In this review, the role of miRs in RAAS signaling and feedback regulation of MR in kidney epithelial cells will be discussed.

non-coding rnas (ncrnas) are key players in variety of biogenesis and biological functions. Their aberrant expression has been implicated in disease progression. Ncrnas can be divided into short ncrnas whose subtypes are mainly microrna (mirna), long non-coding rna (lncrna) and circular rna (circrna). They are involved in cellular processes, including gene regulation, development and disease. The retina is a remarkably sophisticated instrument with interconnected cell types and is the primary target of many genetic diseases. In addition, in terms of retinal dyshomeostasis and inflammation, ncrnas seems to play critical roles in many retinal diseases. Here, we provide an overview of ncrnas in developing retina. We also review how does these ncrnas function in various retinal diseases including animal and human models. These data indicate that ncrnas regulate cellular processes including cell proliferation, differentiation, apoptosis and contribute to initiation and progression of retinal diseases.

Celiac disease (CeD) is a prevalent gluten-induced autoimmune condition that affects approximately 1%-2% of the global population. Recent research has emphasized the pivotal role of non-coding rnas (ncrnas) in regulating inflammatory responses and modulating autoimmune processes. This diverse group contains micrornas (mirnas), long non-coding rnas (lncrnas), and circular rnas (circrnas),and serves as critical regulators in various physiological and disease-related pathways.A group of validated non-coding rnas have emerged as promising biomarkers forthe diagnosis and treatment *** review aims to elucidate the pathogenesis of CeD and investigate the potential of mirnas, lncrnas, and circrnas as diagnostic biomarkers for this condition. By doing so, we hope to offer valuable insights into the underlying mechanisms of CeD and demonstrate the utility of ncrnas as diagnostic and therapeutic tools in managing this autoimmune disorder.

Ageing is a complex process common to all living organisms, influenced by different environmental and genetic factors which are difficult to understand. Epigenetic modifications such as DNA methylation, histone posttranslational modification and non-coding rna affect ageing. Eusocial insects provide an ideal platform for analysing the impact of epigenetic changes on ageing due to their phenotypic plasticity. This study summarizes most of the data published so far on epigenetic changes during ageing in eusocial insects.

Colorectal cancer (CRC) is one of the most common types of cancer which affects the colon and the rectum. Approximately one third of annual CRC mortality occurs due to the late detection of this type of cancer. Therefore, there is an urgent need for more powerful diagnostic and prognostic tools for identification and treatment of colorectal tumorigenesis. non-coding rnas (ncrnas) have been implicated in the pathology of CRC and also linked to metastasis, proliferation, differentiation, migration, angiogenesis and apoptosis in numerous cancers. Recently, attention has turned towards ncrnas as specific targets for diagnosis, prognosis and treatment of various types of cancers, including CRC. In this review, we have tried to outline the roles of ncrnas, and their involvement in signaling pathways responsible for the progression of CRC.

N-6-Methyladenosine (m(6)A) rna modification has emerged in recent years as a new layer of regulatory mechanism controlling gene expression in eukaryotes. As a reversible epigenetic modification found not only in messenger rnas but also in non-coding rnas, m(6)A affects the fate of the modified rna molecules and plays important roles in almost all vital bioprocesses, including cancer development. Here we review the up-to-date knowledge of the pathological roles and underlying molecular mechanism of m(6)A modifications (in both coding and non-coding rnas) in cancer pathogenesis and drug response/resistance, and discuss the therapeutic potential of targeting m(6)A regulators for cancer therapy.

Long non-coding rnas and TGF-β signaling in cancer

Cancer is driven by genetic mutations in oncogenes and tumor suppressor genes and by cellular events that develop a misregulated molecular microenvironment in the growing tumor tissue. The tumor microenvironment is guided by the excessive action of specific cytokines including transforming growth factor-beta (TGF-beta), which normally controls embryonic development and the homeostasis of young or adult tissues. As a consequence of the genetic alterations generating a given tumor, TGF-beta can preserve its homeostatic function and attempt to limit neoplastic expansion, whereas, once the tumor has progressed to an aggressive stage, TGF-beta can synergize with various oncogenic stimuli to facilitate tumor invasiveness and metastasis. TGF-beta signaling mechanisms via Smad proteins, various ubiquitin ligases, and protein kinases are relatively well understood. Such mechanisms regulate the expression of genes encoding proteins or non-coding rnas. Among non-coding rnas, much has been understood regarding the regulation and function of micrornas, whereas the role of long non-coding rnas is still emerging. This article emphasizes TGF-beta signaling mechanisms leading to the regulation of non-coding genes, the function of such non-coding rnas as regulators of TGF-beta signaling, and the contribution of these mechanisms in specific hallmarks of cancer.
