Colorimetric sensor array for sensitive detection and identification of bacteria based on the etching of triangular silver nanoparticles regulated by Cl^- at various concentrations

A colorimetric sensor array is proposed for ultrasensitive detection and identification of bacteria by using Cl- at various concentrations as sensing elements and triangular silver nanoparticles (T-AgNPs) as a single sensing nanoprobe. T-AgNPs are easily etched by Cl-. However, in the presence of bacteria, the etching process will be hindered. Different bacteria have differential protective effects on T-AgNPs due to their interactions, resulting in different etching degrees of T-AgNPs by Cl-, and visual color changes. By adjusting the antagonistic action between bacteria protection on T-AgNPs and the etching by using Cl- at various concentrations, different bacteria had their own color response patterns. Combined with linear discriminant analysis (LDA) and orthogonal partial least squares-discriminant analysis (OPLS-DA), the bacteria could be identified. The method was also used for bacteria mixtures identification and showed high sensitivity (OD600 = 1.0 x 10-6) for V. parahaemolyticus detection. Finally, the sensor array was successfully utilized in the identification of bacteria in pure and mineral bottled water. The method is low-cost, simple, sensitive, visual, and has potential application in point-of-care testing of bacteria.

Experimental study on effect of different parameters on size and shape of triangular silver nanoparticles prepared by a simple and rapid method in aqueous solution

This paper continues our previous work on preparation of truncated triangular silver nanoparticles. The method proceeds with reaction of silver nitrate with hydrazine in the presence of sodium citrate in aqueous solution, in which triangular nanoparticles are formed in a few minutes with some spherical ones. In particular range of reactants, especially high reductant concentration, only spherical nanoparticles are formed. In further investigation we observed that spherical nanoparticles shape could change to triangular by aging. This means that controlled growth of nanoparticles could lead to the formation of triangular ones. Therefore, a method was devised to slow down the rate of reduction by adding Fe3+ to the reaction solution. The results show that in this case more triangular nanoparticles are formed compared to the original one. This result also confirms that with the increasing hydrazine concentration, growth becomes less important compared to nucleation and smaller triangles are formed. (C) 2009 King Saud University. There is always a substantial effort to develop a point of care detection fora severe and acute disease like dengue. In this work, we have described the detection of serotype-specific dengue virus using multicolor triangular silver nanopartides (TAg) which could be a potential diagnosis method to distinguish between serotype. Functionalized TAg was prepared with polyA(10) DNA through pH-mediated process. Further, it was characterized with Transmission Electron Microscopy (TEM), Ultra Visible-visible spectroscopy (UV-vis) and gel electrophoresis and its stability towards NaCl concentration. The colorimetric detection was carried out based on the interaction of TAg-DNA probe with specific complimentary strand which was designed to form a network assembly between DNA probes and sample RNA. The hybridization process was enhanced in presence of NaCI (0.4 M) which makes the hybridization process more stable over probe compared to un-hybridization. Therefore a simple and effective colorimetric method was developed to detect the different dengue serotype RNA with high sensitivity and specificity. (C) 2018 Elsevier B.V. All rights reserved.

Fast and simple synthesis of triangular silver nanoparticles under the assistance of light

Herein, the fast and simple formation of triangular silver nanoparticles (T-Ag NPs) was systematically studied by a chemical reduction process with the assistance of light irradiation. The effects of sodium borohydride (NaBH4) concentration, pH medium, and light source irradiation are investigated via the localized surface plasmon resonances (LSPRs) detection of silver nanoparticles (Ag NPs). Results indicated that pH and light source are the two primary factors for the morphology of Ag NPs while NaBH4 plays an important role to control the formation rate of T-Ag NPs. Moreover, the photoreduction time for synthesis of T-Ag NPs is 180 min, and this method can be easily synthesized in any laboratory.

This system utilizes an innovative platform consisting of a poly (methyl methacrylate) (PMMA) plate adorned with triangular silver nanoparticles, enabling the sensitive and specific detection of prostate-specific antigen (PSA), carcinoembryonic antigen (CEA), and cancer antigen 15-3 (CA 15.3). To achieve this, the PMMA polymer microarrays were precisely cut using a laser cutting machine to create an optimized number of micro-arrays. These designed wells were embellished with triangular silver nanoparticles (TA-AgNPs) featuring prism morphology. The antigens could be easily detected based on antigen-antibody-induced aggregation of TA-AgNPs and resulting in a visible color change due to the alteration of surface plasmon resonance properties. Consequently, utilizing the immunocomplex of antigen-antibody in the presence of the optical probe (TA-AgNPrs) and a portable photochemical sensor, candidate cancer biomarkers (PSA, CEA, and CA 15.3) were successfully identified with linear ranges of 1-60, 1-75, and 1-125 mu g/mL, respectively. By employing digital image analysis using a smartphone, we achieved a highly favorable low limit of quantification of 1 mu g/mL for PSA, CEA, and CA 15.3 as biomarkers for prostate, colon, and breast cancer, respectively. This groundbreaking research introduces a pioneering digital image-based colorimetry immunoassay, the first of its kind in the field, providing a transformative approach to the early-stage diagnosis of various cancer biomarkers through a single opto-device. To the best of our knowledge, no smartphone ambient light sensor has yet been specifically designed to identify PSA, CEA, and CA 15.3 proteins as cancer biomarkers. Therefore, a sensory micro-array was developed to open new possibilities for the rapid and simple colorimetric identification of PSA, CEA, and CA 15.3 antigens in real samples, offering a sensitive and specific monitoring strategy for these biomarkers through colorimetry.

Trace-level detection of Dopamine (DA) in human body fluids is crucial for diagnosing neurological disorders, understanding mental health, and advancing drug discovery, but found challenging with rapid and low-cost detection technologies. Herein, an innovative paper-based photochemical method has been developed for the detection of DA using triangular silver nanoprism (AgNPrs), as an optical nanoprobe, in human blood plasma and urine samples. The redox reaction between DA and AgNPrs was evaluated through a color change at two pHs (5.35and 6.14), which can be analyzed by UV -Vis spectrophotometry and microfluidic paper-based colorimetric device ( mu PCD) with low limit of quantification of 0.01 mu M. This study presents pioneering work with the potential to inspire the development of simple and effective on -site semi quantifying recognition devices for other neurotransmitters, requiring only a lightweight piece of paper and a single drop of the necessary reagent.

Observation of Size-Dependent Electron-Phonon Scattering and Temperature-Dependent Photoluminescence Quenching in triangular-Shaped silver nanoparticles

Research studies on plasmonic properties of triangular-shaped silver nanoparticles might lead to several interesting applications. However, in this work, triangular-shaped silver nanoparticles have been synthesized by simple solvothermal technique and reported the effect of size on the electron-phonon scattering in the synthesized materials by analyzing their temperature-dependent photoluminescence (PL) emission characteristics. It has been observed that total integrated PL emission intensity is quenched by 33 % with the increase in temperature from 278 to 323 K. The observed decrease in PL emission intensity has been ascribed to the increase of electron-phonon scattering rate with the increase in temperature. The values of electron-phonon coupling strength (S) for synthesized samples have been evaluated by theoretical fitting of the experimentally obtained PL emission data. Smaller sized triangular nanoparticle has been found to exhibit stronger temperature dependence in PL emission, which strongly suggests that smaller sized triangular silver nanostructures have better electron-phonon coupling.

With the ever-growing threat of bacterial infections, especially for multidrug-resistant microbial infections, the development of highly effective treatment modalities to inhibit the infections is challenging. Although silver nanoparticles have been intensively applied as antimicrobial agent for decades, the therapeutic efficacy toward multidrug-resistant bacteria is still unsatisfactory. Here, we show that near-infrared (NIR) laser-excited silver triangular nanoparticles (Tri-Ag) can efficiently kill Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus both in vitro and in vivo. Notably, multidrug-resistant bacterial clinical isolates, including methicillin-resistant S. aureus and extended spectrum beta-lactamase E. coli strain were significantly inhibited by the combined treatment of the Tri-Ag with NIR laser irradiation due to their synergistic antibacterial ability. Taking the advantage of its strong near-infrared absorbance, photothermal treatment is also conducted with Tri-Ag, achieving a remarkable synergistic antibacterial effect to inhibit various bacteria at a rather low concentration of this agent. Given the above advantages, the combination therapy of Tri-Ag with assistance of NIR laser may find potential applications to strengthen the antimicrobial arsenal for fighting bacterial infections.

Radiolytic formation of highly luminescent triangular Ag nanocolloids

In this work, luminescent triangular silver nanoparticles were synthesized by radiolytic reduction method. The results showed that by variation of irradiation dose, morphology of silver nanoparticles can be converted from spherical to triangular. These shape variations mainly arise from competition between adsorption rate of polymer chains on (111) facets and reduction rate of the Ag+ ions along (110) facets during increasing dose. Furthermore, the dramatically enhanced photoluminescence spectra were observed from triangular Ag nanoparticles. This unusual behavior can be explained by excitation of dipolar and quadrupolar resonance in triangular nanoparticles which increase the electric fields at the surface.

Most nanoparticle based colorimetric sensor array utilize several sensor elements and static response for discrimination of target analytes. This approach can be complicated and costly to synthesize or functionalize different nanoparticles for providing wide color variation. Herein, triangular silver nanoparticles (TSNPs) were used to develop a colorimetric sensor array by time-dimension responses. The principle of this sensor array is based on the diverse etching process of TSNPs in the presence of three halide ions, including bromide (Br-), iodide (I-) and chloride (Cl-). Various etchings of TSNPs induced color changes at different reaction time in-tervals, which produced a colorimetric pattern for each ion. Therefore, using time dependent etching responses of TSNPs as a single sensing component can produce a wide color variation which can be distinguished by naked eyes. The colorimetric responses of TSNPs upon the addition of different concentrations of halide ions have been analyzed by PLS regression (PLS-R) and PLS discriminant analysis (PLS-DA). The analytical figures of merit confirmed that the developed chrono-colorimetric TSNPs-based sensor array is successful in both the discrim-ination and quantitative detection of halide ions. At the final step, the three halide ions were accurately determined in a real water sample, which verified the potential of the developed sensor in a real sample.
