Host factors and genetic susceptibility to infections due to intracellular bacteria and fastidious organisms

While genetic polymorphisms play a paramount role in tuberculosis (TB), less is known about their contribution to the severity of diseases caused by other intracellular bacteria and fastidious microorganisms. We searched electronic databases for observational studies reporting on host factors and genetic predisposition to infections caused by intracellular fastidious bacteria published up to 30 May 2014. The contribution of genetic polymorphisms was documented for TB. This includes genetic defects in the mononuclear phagocyte/T helper cell type 1 (Th1) pathway contributing to disseminated TB disease in children and genome-wide linkage analysis (GWAS) in reactivated pulmonary TB in adults. Similarly, experimental studies supported the role of host genetic factors in the clinical presentation of illnesses resulting from other fastidious intracellular bacteria. These include IL-6 -174G/c or low mannose-binding (MBL) polymorphisms, which are incriminated in chronic pulmonary conditions triggered by ***, type 2-like cytokine secretion polymorphisms, which are correlated with various clinical patterns of *** infections, and genetic variation in the NOD2 gene, which is an indicator of tubal pathology resulting from chamydia trachomatis infections. Monocyte/macrophage migration and T lymphocyte recruitment defects are corroborated to ineffective granuloma formation observed among patients with chronic Q fever. Similar genetic polymorphisms have also been suggested for infections caused by *** although not confirmed yet. In conclusion, this review supports the paramount role of genetic factors in clinical presentations and severity of infections caused by intracellular fastidious bacteria. Genetic predisposition should be further explored through such as exome sequencing.

Fecundity of the sea louse caligus rogercresseyi on its native host Eleginops maclovinus captured near salmon farms in southern chile

The sea louse (caligus rogercresseyi) is the most significant parasitic pathogen in chilean salmon farms, and it infects farmed salmon and native host fish. Fecundity is one of the most important parameters for understanding the population dynamics of a species;however, this information is scarce for this parasite. The fecundity of c. rogercresseyi females collected from native hosts (Eleginops maclovinus) captured near salmon farms in southern chile was measured to evaluate the reproductive output of this parasite on this host fish. From June 2008 to May 2009, 212 specimens of E. maclovinus were examined. Each fish was measured, and all its parasites were collected, sorted and counted. Seventy-nine ovigerous c. rogercresseyi females (OFs) were measured. Total body length, egg string length and total number of eggs per string were recorded for each parasite. Ovigerous females body length varied between 3.9 and 5.0 mm. Fecundity varied between 12 and 56 eggs string-1, and it was correlated with OF body length. Temporal variations in OF fecundity were explained by co-variation in OF body length, but not by month. Ovigerous females on E. maclovinus were smaller and showed lower fecundity than OFs on farmed salmon. Our results suggest that native hosts play a secondary role in c. rogercresseyi egg production in codigue bay.

Neurobeachin (NBEA) was initially identified as a candidate gene for autism. Recently, variants in NBEA have been associated with neurodevelopmental delay and childhood epilepsy. Here, we report on a novel NBEA missense variant (c.5899G > A, ***1967Arg) in the Domain of Unknown Function 1088 (DUF1088) identified in a child enrolled in the Undiagnosed Diseases Network (UDN), who presented with neurodevelopmental delay and seizures. Modeling of this variant in the caenorhabditis elegans NBEA ortholog, sel-2, indicated that the variant was damaging to in vivo function as evidenced by altered cell fate determination and trafficking of potassium channels in neurons. The variant effect was indistinguishable from that of the reference null mutation suggesting that the variant is a strong hypomorph or a complete loss-of-function. Our experimental data provide strong support for the molecular diagnosis and pathogenicity of the NBEA ***1967Arg variant and the importance of the DUF1088 for NBEA function. (c) 2021 The Authors. FUNcTIONAL REcONSTITUTION OF PHOTOSYNTHETIc cYcLIc ELEcTRON-TRANSFER IN LIPOSOMES

The interaction of solubilized reaction centers from the phototrophic bacterium, Rhodobacter sphaeroides,and the solubilized ubiquinol-cytochrome c oxidoreductase of Rhodobacter capsulatus was studied in solution and after coreconstitution in liposomes prepared from Escherichia coli phospholipids. Under both conditions, the ubiquinol-cytochrome c oxidoreductase increased the light-induced cyclic electron transfer, induced be reaction centers with 2,3-dimethoxy-5-methyl-6-(prenyl)2-1,4-benzoquinone and cytochrome c as redox mediators. This effect was more pronounced at acid pH values. The light-induced cyclic electron transfer in these liposomes resulted in the generation of a protonmotive force. Under conditions where the protonmotive force was composed of a membrane potential only, the highest membrane potential (approx. -200 mV) was generated when 2,3-dimethoxy-5-methyl-6-(prenyl)10-1,4-benzoquinone was used as redox mediator and when both electron transfer proteins were co-reconstituted in a 2:1 molar ratio. At acid pH non-transitent membrane potentials could be generated only in liposomes containing both reaction centers and the ubiquinol-cytochrome c oxidoreductase. These observations show that the pH-dependent direct oxidation of cytochrome c by ubiquinol in the liposomes was indeed catalyzed by the ubiquinol-cytochrome c oxidoreductase and that this oxidoreductase participates in proton pumping. This could also be concluded from the stimulating effect of 2,3-dimethoxy-5-methyl-6(prenyl)10-1,4-benzoquinone on the membrane-potential-generating capacities in liposomes containing both electron transfer complexes. Such a stimulation was not observed in liposomes containing only reaction centers. The presence of cytochrome c in the co-reconstituted system was found to be essential for proton pumping.

Polymorphism of HLA class I genes in Meizhou Han population of Guangdong, china

Human leukocyte antigen (HLA) is an invaluable marker for anthropological studies because of its extreme polymorphism. Most of the studies carried out in chinese populations are about HLA class II genes, but few about class I genes. In the present study, we investigated HLA class I polymorphism using polymerase chain reaction-sequencing-based typing (PcR-SBT) method in 104 unrelated Han individuals in Meizhou of Guangdong, southern china. Twenty-three HLA-A, 43 HLA-B and 27 HLA-c alleles were identified and allele frequencies and two-locus (c/B) and three-locus (A/c/B) haplotypes were statistically analysed. The most frequent HLA-A allele is A*110101 with a frequency of 30.3%, followed by A*24020101 (22.2%) and A*2420 (11.6%). Among the 43 detected HLA-B alleles, B*5801 (17.0%), B*400101 (15.5%) and B*4601 (10.0%) were frequently observed. Among the 27 detected c alleles, the most predominant one is cw*07020101 (25.8%), followed by cw*0717 (14.7%). The most frequent HLA-c/B two-locus haplotype is cw*07020101/B*400101 (10.1%). The most common HLA-A/c/B three-locus haplotype in Meizhou Han is A*110101/cw*07020101/B*400101 (3.4%). Phylogenetic tree based on HLA class I allele frequencies genetically suggested that Meizhou Han has an affinity to southern Asian populations. The result may also reflect an admixture of Han and ethnic minorities of southern china.

Optimising nutrients in the culture medium of Rhodosporidium toruloides enhances lipids production

Rhodosporidium toruloides is a useful oleaginous yeast, but lipids production is affected by various factors including nutrients in the culture medium. Herein, the R-ZL2 high-yield mutant strain was used to investigate the effects of different carbon sources (sucrose, glucose, xylose), nitrogen sources (ammonium sulphate, ammonium nitrate), and c/N ratio on lipids production capacity, get the following conclusion (1) compared with glucose and xylose, sucrose was a superior carbon source for lipids production;(2) When using ammonium sulphate as the nitrogen source, a c/N ratio of 200:1 achieved the highest biomass, lipids production and lipids content (10.7 g/L, 6.32 g/L and 59%, respectively), and lipids produced under different c/N conditions have potential for biodiesel production (except for c/N = 40 and c/N = 80);(3) When using ammonium nitrate as the nitrogen source, a c/N ratio of 200:1 achieved the highest biomass, lipids production and lipids content (12.1 g/L, 8.25 g/L and 65%, respectively), and lipids produced under different c/N ratio conditions have potential for biodiesel production. Thus, a combination of sucrose and ammonium nitrate was optimal for the lipid accumulation in R-ZL2. The findings will lay a foundation for further improving lipids yields.

Germline Maintenance Through the Multifaceted Activities of GLH/Vasa in caenorhabditis elegans P Granules

Vasa homologs are ATP-dependent DEAD-box helicases, multipotency factors, and critical components that specify and protect the germline. They regulate translation, amplify piwi-interacting RNAs (piRNAs), and act as RNA solvents;however, the limited availability of mutagenesis-derived alleles and their wide range of phenotypes have complicated their analysis. Now, with clustered regularly interspaced short palindromic repeats (cRISPR/cas9), these limitations can be mitigated to determine why protein domains have been lost or retained throughout evolution. Here, we define the functional motifs of /Vasa in caenorhabditis elegans using 28 endogenous, mutant alleles. We show that 's helicase activity is required to retain its association with P granules. remains in P granules when changes are made outside of the helicase and flanking domains, but fertility is still compromised. Removal of the glycine-rich repeats from GLH proteins progressively diminishes P-granule wetting-like interactions at the nuclear periphery. Mass spectrometry of -associated proteins implies conservation of a transient piRNA-amplifying complex, and reveals a novel affinity between and three structurally conserved PcI (26S Proteasome Lid, cOP9, and eIF3) complexes or "zomes," along with a reciprocal aversion for assembled ribosomes and the 26S proteasome. These results suggest that P granules compartmentalize the cytoplasm to exclude large protein assemblies, effectively shielding associated transcripts from translation and associated proteins from turnover. Within germ granules, Vasa homologs may act as solvents, ensuring mRNA accessibility by small RNA surveillance and amplification pathways, and facilitating mRNA export through germ granules to initiate translation.

carbon Stock in Sandy and Loamy Soils of coniferous-Broadleaved Forests at Different Succession Stages

A comparative assessment of the carbon stock in loamy soils (Albic Retisols) of coniferous-broadleaved forests on the Moskvoretsko-Oksky Plain and sandy soils (Albic Podzols) in the Bryansk region is given. The carbon accumulation in soils is related to the texture of their mineral horizons and biotic factors, such as the quality of litter and activity of earthworms. The forests on sandy soils were characterized by a high carbon stock in the plant litter, which is explained by the low biomass of earthworms. The carbon content was significantly higher in the mineral horizons of loamy soils owing to high share of fine particles. The carbon stock in the litter and mineral horizons of loamy and sandy soils turned out to be comparable, except for the sandy soils of mixed forests due to higher clay content. In the course of succession of the coniferous-deciduous forests on the loamy soil, the carbon accumulation in the litter was high only at the late stage due to the high input of spruce needles. The carbon stock in the humus horizons of the loamy soils varied due to changes in their density, which was regulated by earthworms. When comparing different succession stages/types of the forests on sandy soils, it was found that the carbon stock in the litter of the pine forests at the early stage of succession was significantly higher than that of the mixed and broadleaved forests. Trends of carbon accumulation in the mineral horizons of sandy soils were mainly explained by their thickness, which was related to the amount of percolating precipitation regulated by woody plants.

Three quinolinone alkaloid - phenylpropanoid adducts from Melicope pteleifolia

Preliminary in vitro cytotoxic test on different extracts of Melicope pteleifolia collected at Dak Nong province, Vietnam showed that the n-hexane one was the most potent. From this n-hexane extract, three new quinolinone alkaloid-phenylpropanoid derivatives (1-3) and three known compounds (4-6) were isolated. Based on NMR and HR-MS analysis, their chemical structures were elucidated as melicoptines A-c (1-3), flindersine (4), 3,4,5-trimethoxybenzoic acid (5) and (24S)-methylcholestan-1 alpha,3 beta-diol (6). Isolated compounds (1-4) were evaluated for their anti-bacterial and cytotoxic activities against human non-small cell lung cancer (A549), human cervical cancer (HeLa), human Burkitt's lymphoma (Raji) and normal fibroblasts (NIH-3T3). All of them were inactive.

Isotropic and linear dichroic triplet-minus-singlet (T - S) spectra of reaction centers of Rhodobacter sphaeroides 2.4.1 and Rhodospirillum rubrum S1 have been measured with magneto-optical difference spectroscopy (MODS) in the wavelength range 350-950 nm as a function of the temperature (10-288 K). The (T - S) spectra allowed the determination of the carotenoid triplet yield and the probability of triplet transfer between the primary donor bacteriochlorophyll a (Bchl a) dimer, D, and the carotenoid as a function of the temperature. The ***.3c transfer can be described with an equilibrium model, without introducing additional triplet states. The ***.3c transfer has an activation enthalpy of 0.020 eV, while the activation enthalpy of the backward transfer is 0.15 eV for Rb. sphaeroides 2.4.1 and 0.08 eV for R. rubrum S1 and apparently depends on the type of carotenoid present in the reaction center (spheroidene in Rb. sphaeroides 2.4.1, spirilloxanthin in R. rubrum S1). The energeis of the spheroidene and the spirilloxanthin triplet states in the reaction center are 0.81 and 0.88 eV, respectively. The protection against free oxygen by the carotenoid at physiological temperatures is therfore better in Rb. sphaeroides 2.4.1 than in R. rubrum S. The mechanism of triplet transfer between dimer and carotenoid is discussed. It is suggested that one possible mechanism may involve a change in triplet localization of 3D, populating the higher-energy triplet exciton state of 3D. Evidence has been found that 3c shares its triplet exciton with an accessory Bchl a and that with increasing temperature the triplet state on 3D becomes more delocalized. The temperature-dependence of the energy and bandwidth of the low-exciton band of D, D(-), is interpreted to be mainly a consequence of thermal broadening and expansion of the Bchl a dimer.
