This study aims to advance our mechanistic understanding of electrical stimulation of dorsal root ganglia (DRG) for treating chronic pain. While DRG stimulation has shown moderate clinical success in managing certain types of chronic pain, the underlying neural mechanism remains inconclusive, hindering the further development of the technology to treat a broader range of chronic pain symptoms and benefit a larger patient population. In this study, we conducted computational simulations in the NEURON simulation environment to assess the neuromodulatory effect of DRG stimulation on action potential transmission in Aδ-fiber and C-fiber sensory afferents. Our simulation incorporates Markov-type state models to capture the subtle gating characteristics of voltage-gated sodium channel subtypes, especially NaV1.6, the anatomical distribution of which was revealed by our immunohistological staining on sparsely labeled afferents. Our simulation results indicate that DRG stimulation causes a significant increase in intra-axonal Na + concentration and a reduction in K + concentration, collectively disrupting the transaxonal ionic gradients. This disruption resulted in activitydependent conduction slowing, leading to the eventual conduction block in both Aδ- and C-fiber afferents. This research marks a crucial step forward in unraveling the intricate mechanisms underlying DRG stimulation, presenting a framework for the further development of innovative pain modulation strategies that target the DRG.

The purpose of this biomedical engineering study is to examine interventional and surgical approaches for complex and palliative solutions in infants with patent ductus arteriosus and pulmonary artesia diagnosis with mathematical modeling and computational fluid mechanics methods. In the study, we visualized hemodynamically using patient-specific preoperative cross-sectional image data and computer-aided design techniques and discussed the differences between repair methods through mathematical models and virtual repairs. We discussed the significance of blood flow analysis in the treatment planning of infants with ductal-dependent pulmonary flow.

computational modeling of the Chlamydial Developmental Cycle Reveals a Potential Role for Asymmetric Division

Chlamydia trachomatis is an obligate intracellular bacterium that progresses through an essential multicell form developmental cycle. Infection of the host is initiated by the elementary body (EB). Once in the host, the EB cell differentiates into the noninfectious, but replication-competent, reticulate body, or RB. After multiple rounds of replication, RBs undergo secondary differentiation, eventually producing newly infectious EBs. Here, we generated paired cell-type promoter reporter constructs and determined the kinetics of the activities of the euo, hctA, and hctB promoters. The paired constructs revealed that the developmental cycle produces at least three phenotypically distinct cell types, the RB (euoprom(+)), intermediate body (IB;hctAprom(+)), and EB (hctBprom(+)). The kinetic data from the three dual-promoter constructs were used to generate two computational agent-based models to reproduce the chlamydial developmental cycle. Both models simulated EB germination, RB amplification, IB formation, and EB production but differed in the mechanism that generated the IB. The direct conversion and the asymmetric production models predicted different behaviors for the RB population, which were experimentally testable. In agreement with the asymmetric production model, RBs acted as stem cells after the initial amplification stage, producing one IB and self-renewing after every division. We also demonstrated that IBs are a transient cell population, maturing directly into EBs after formation without the need for cell division. The culmination of these results suggests that the developmental cycle can be described by a four-stage model, EB germination, RB amplification/maturation, IB production, and EB *** Chlamydia trachomatis is an obligate intracellular bacterial pathogen responsible for both ocular and sexually transmitted infections. Background Expert elicitation (EE) has been used across disciplines to estimate input parameters for computational modeling research when information is sparse or conflictual. Objectives We conducted a systematic review to compare EE methods used to generate model input parameters in health research. Data Sources PubMed and Web of Science. Study Eligibility modeling studies that reported the use of EE as the source for model input probabilities were included if they were published in English before June 2021 and reported health outcomes. Data Abstraction and Synthesis Studies were classified as "formal" EE methods if they explicitly reported details of their elicitation process. Those that stated use of expert opinion but provided limited information were classified as "indeterminate" methods. In both groups, we abstracted citation details, study design, modeling methodology, a description of elicited parameters, and elicitation methods. Comparisons were made between elicitation methods. Study Appraisal Studies that conducted a formal EE were appraised on the reporting quality of the EE. Quality appraisal was not conducted for studies of indeterminate methods. Results The search identified 1520 articles, of which 152 were included. Of the included studies, 40 were classified as formal EE and 112 as indeterminate methods. Most studies were cost-effectiveness analyses (77.6%). Forty-seven indeterminate method studies provided no information on methods for generating estimates. Among formal EEs, the average reporting quality score was 9 out of 16. Limitations Elicitations on nonhealth topics and those reported in the gray literature were not included. Conclusions We found poor reporting of EE methods used in modeling studies, making it difficult to discern meaningful differences in approaches. Improved quality standards for EEs would improve the validity and replicability of computational models.

Teachers' Disciplinary Boundedness in the Implementation of Integrated computational modeling in Physics

This study explored teachers' conceptualizations of integrated computational modeling in secondary physics by exposing twelve experienced physics teachers to programming and then analyzing interview responses. Responses revealed that teachers fell along a spectrum of disciplinary boundary-stretching mentalities. This paper presents a preliminary conceptual framework for exploring both horizontal (interdisciplinary) and vertical (intradisciplinary) boundary stretching, as well as for identifying bounded mentalities as teachers consider integration. Horizontal boundary stretchers envisioned opportunities to use computational modeling to shift their curriculum or pedagogical approaches in physics to help students enhance skills underlying multiple fields, while vertical boundary stretchers considered how computing might allow students to explore physics concepts more deeply. Teachers with more boundary-stretching indicators at the outset of an integrated curriculum development workshop were more likely to persist in the implementation of computational modeling-integrated materials in their physics classrooms than those who expressed more bounded thinking. These findings emphasize the importance of considering teachers' perceptions about how their own science discipline is connected to similar fields and provide implications about how to identify potential adopters of innovative teaching approaches.

BACKGROUND: Motivational deficit is a core clinical manifestation of depression and a strong predictor of treatment failure. However, the underlying mechanisms, which cannot be accessed through conventional questionnaire-based scoring, remain largely unknown. According to decision theory, apathy could result either from biased subjective estimates (of action costs or outcomes) or from dysfunctional processes (in making decisions or allocating resources).METHODS: Here, we combined a series of behavioral tasks with computational modeling to elucidate the motiva-tional deficits of 35 patients with unipolar or bipolar depression under various treatments compared with 35 matched healthy control ***: The most striking feature, which was observed independent of medication across preference tasks (likeability ratings and binary decisions), performance tasks (physical and mental effort exertion), and instrumental learning tasks (updating choices to maximize outcomes), was an elevated sensitivity to effort cost. By contrast, sensitivity to action outcomes (reward and punishment) and task-specific processes were relatively ***: These results highlight effort cost as a critical dimension that might explain multiple behavioral changes in patients with depression. More generally, they validate a test battery for computational phenotyping of motivational states, which could orientate toward specific medication or rehabilitation therapy, and thereby help pave the way for more personalized medicine in psychiatry.

Implementing Laboratory Management Systems (LMS) involves an accreditation process whereby the laboratory documents its processes and demonstrates the quality and reliability of those processes. The ISO/IEC 17025 standard specifies the accreditation requirements in an international context. Since accreditation processes are complex, expensive, involve multiple steps and variables and a heavy documentation burden, it would be desirable to have some form of quick a priori assessment of a laboratory before it embarks on the accreditation journey. This paper presents a computational model based on Bayesian Networks (BN) for this purpose. The formalism focused on the use of Bayesian statistics and decision graphs. Identification of the BN variables, conditional dependencies, probabilistic information and domain knowledge were obtained from field investigations. Model evaluation was performed using a set of known scenarios that represent unequivocal prescriptions. The model enables predicting the results of implementing a management system, and simulating the accreditation process results. This development also identified the variables that have a significant influence on the expected results of the LMS accreditation process. © 2022 IEEE.

Carotid atherosclerotic plaque growth leads to the progressive luminal stenosis of the vessel, which may erode or rupture causing thromboembolism and cerebral infarction, manifested as stroke. Carotid atherosclerosis is considered the major cause of ischemic stroke in Europe and thus new imaging-based computational tools that can improve risk stratification and management of carotid artery disease patients are needed. In this work, we present a new computational approach for modeling atherosclerotic plaque progression in real patient-carotid lesions, with moderate to severe degree of stenosis (>50%). The model incorporates for the first time, the baseline 3D geometry of the plaque tissue components (e.g. Lipid Core) identified by MR imaging, in which the major biological processes of atherosclerosis are simulated in time. The simulated plaque tissue production results in the inward remodeling of the vessel wall promoting luminal stenosis which in turn predicts the region of the actual stenosis progression observed at the follow-up visit. The model aims to support clinical decision making, by identifying regions prone to plaque formation, predict carotid stenosis and plaque burden progression, and provide advice on the optimal time for patient follow-up screening.

computational modeling has great advantages in human behavior research, such as abstracting the problem space, simulating the situation by varying critical variables, and predicting future outcomes. Although much research has been conducted on driver behavior modeling, relatively little modeling research has appeared at the Auto-UI Conferences. If any, most work has focused on qualitative models about manual driving. In this workshop, we will first describe why computational driver behavior modeling is crucial for automotive research and then, introduce recent driver modeling research to researchers, practitioners, and students. By identifying research gaps and exploring solutions together, we expect to form the basis of a new modeling special interest group combining the Auto-UI community and the computational modeling community. The workshop will be closed with suggestions on the directions for future transdisciplinary work.

Implantable neuromodulation devices that interface with the peripheral nervous system are a promising approach to restore functions lost to nerve damage. Existing nerve stimulation electrodes require direct contact with the target nerve and are associated with mechanical nerve damage and fibrous tissue encapsulation. Endovascularly delivered electrode arrays may provide a less invasive solution. Using a hybrid tissue conductor-neuron model and computational simulations, this study demonstrates the feasibility of delivering electrical stimulation of a peripheral nerve from a blood vessel in the vicinity of the target and predicts that the stimulation intensity required strongly depends on nerve-vessel distance and relative orientation, which are important factors to consider when screening candidate blood vessels for electrode implantation.
