Accurate correction method and algorithm of fluorescence secondary inner filter effect (sIEF) in fluorescence quantitative analysis

Fluorescence spectroscopy is a reliable and widely used analytical method. The fluorescence inner filter effect (IFE) is one of the main obstacles in the application of fluorescence spectroscopy and an error source in fluo-rescence analysis, resulting in the fluorescence spectrum distortion, the spectral shape distortion, and the nonlinearity between fluorescence intensity and fluorophore concentration. An optimized parameter reflecting the self-absorption effect -the fluorescence attenuation absorption index of secondary inner filter effect (sIFE) nopt -is proposed in this paper. Considering the received fluorescence in a direction perpendicular to the incident light, it is related to the solute-solvent system of the fluorescent substance, neither the geometric parameters of the cuvette and the light beam nor the concentration of the fluorescent substance. nopt can accurately reflect the degree to which the fluorescence is affected by the sIFE and correct for any non-ideality of the shapes of exci-tation/emission beams. The principle and determination method of nopt are explained in detail. Accordingly, an algorithm for the fluorescence spectroscopic correction by nopt is designed. To verify the method, the fluorescence spectra and absorbance spectra of the solutions of fluorescein sodium, rhodamine B, rhodamine 6G, and chlorophyll-a with a series of concentration gradients were measured, respectively. The influence of solvent effect on sIFE correction was also studied. The experiments show that different solute-solvent systems of the fluorescent substances have their own nopt. The novel algorithm can determine the nopt, correct the intensity attenuation and the peak red-shift of the fluorescence spectrum caused by the sIFE, expand the linear range of the concentration predicted by the fluorescence intensity, reduce the error of the prediction model, and improve the measurement accuracy.

Research question: Is there a precise and efficient haplotyping method to expand the application of preimplantation genetic testing (PGT)? Design: In this study, eight cell-line families and 18 clinical families including 99 embryos were used to construct whole-genome haplotyping based on link-read sequencing (Phbol-seq) and optimized analytical workflow with a correction algorithm. The correction algorithm was based on a differentiation of assembly errors and homologous recombination, in which the main feature of parental assembly error was that all embryos (embryo number >= 2) had breakpoints at the same chromosome position. Results: With Phbol-seq, parental assembly errors and homologous recombination were accurately distinguished and corrected. Using the link-reads (>25% long-reads were >= 30 kilobases [kb]), complete genome-wide parental haplotypes were constructed, and the consistency of the typing results of each chromosome with a conventional method requiring other family members was more than 95%. In addition, the length of N50 contigs was 11.03-16.2 million bases (mb), which was far beyond the N50 contigs from long-read sequencing (148-863 kb). The complete haplotype analysis of all embryos could be performed by Phbol-seq and revealed 100% concordance with the available diagnostic results obtained by the conventional method requiring other family members.

A feature of diffraction grating spectrometers in the visible and near IR range is that the second-order diffraction spectrum overlaps the first-order diffraction spectrum. This leads to distortions of the recorded spectrum in its long-wavelength region. We propose a method for correction of the second-order diffraction, based on representing the spectrum as the sum of Gaussians with parameters that are experimentally determined for individual wavelengths, followed by numerical interpolation. We also use parametrization of the second-order line shape. We describe the details of the experiment required, and test the performance of the method using an SSP-600N spectrometer as an example.

CT metal artefact reduction for hip and shoulder implants using novel algorithms and machine learning: A systematic review with pairwise and network meta-analyses

Introduction: Many tools have been developed to reduce metal artefacts in computed tomography (CT) images resulting from metallic prosthesis;however, their relative effectiveness in preserving image quality is poorly understood. This paper reviews the literature on novel metal artefact reduction (MAR) methods targeting large metal artefacts in fan-beam CT to examine their effectiveness in reducing metal artefacts and effect on image quality. Methods: The PRISMA checklist was used to search for articles in five electronic databases (MEDLINE, Scopus, Web of Science, IEEE, EMBASE). Studies that assessed the effectiveness of recently developed MAR method on fan-beam CT images of hip and shoulder implants were reviewed. Study quality was assessed using the National Institute of Health (NIH) tool. Meta-analyses were conducted in R, and results that could not be meta-analysed were synthesised narratively. Results: Thirty-six studies were reviewed. Of these, 20 studies proposed statistical algorithms and 16 used machine learning (ML), and there were 19 novel comparators. Network meta-analysis of 19 studies showed that Recurrent Neural Network MAR (RNN-MAR) is more effective in reducing noise (LogOR 20.7;95 % CI 12.6-28.9) without compromising image quality (LogOR 4.4;95 % CI-13.8-22.5). The network meta-analysis and narrative synthesis showed novel MAR methods reduce noise more effectively than baseline algorithms, with five out of 23 ML methods significantly more effective than Filtered Back Projection (FBP) (p < 0.05). Computation time varied, but ML methods were faster than statistical algorithms. Conclusion: ML tools are more effective in reducing metal artefacts without compromising image quality and are computationally faster than statistical algorithms. Overall, novel MAR methods were also more effective in reducing noise than the baseline reconstructions. Implications for practice: Implementation research is needed to establish the clinical suitability of ML MAR in

Comparative analysis of methods for constructing correction algorithms for terrestrial navigation systems by digital road maps

Results are presented for analysis of the accuracy of contemporary approaches to forming constraint equations used with correction of systems for terrestrial navigation by digital road maps. On the basis of analysing the results of statistical tests, correction algorithms are proposed for terrestrial navigation systems obtained in accordance with the approaches analysed. Fields of application are determined for the approaches in question.

Simple algorithm for the correction of MRI image artefacts due to random phase fluctuations

Purpose: Fast Field-Cycling (FFC) MRI is a novel technology that allows varying the main magnetic field B-0 during the pulse sequence, from the nominal field (usually hundreds of millitesla) down to Earth's field or below. This technique uses resistive magnets powered by fast amplifiers. One of the challenges with this method is to stabilise the magnetic field during the acquisition of the NMR signal. Indeed, a typical consequence of field instability is small, random phase variations between each line of k-space resulting in artefacts, similar to those which occur due to homogeneous motion but harder to correct as no assumption can be made about the phase error, which appears completely random. Here we propose an algorithm that can correct for the random phase variations induced by field instabilities without prior knowledge about the phase error. Methods: The algorithm exploits the fact that ghosts caused by field instability manifest in image regions which should be signal free. The algorithm minimises the signal in the background by finding an optimum phase correction for each line of k-space and repeats the operation until the result converges, leaving the background free of signal. Conclusion: We showed the conditions for which the algorithm is robust and successfully applied it on images acquired on FFC-MRI scanners. The same algorithm can be used for various applications other than Fast Field-Cycling MRI. (C) 2017 The Authors. Published by Elsevier Inc.

Over the past few years, a considerable number of studies have focused on estimation of arterial stiffness by using a digital volume pulse (DVP). This can generally be achieved by a linear regression analysis with several pulse parameters. Among various pulse parameters, since time-related pulse parameters, such as reflected wave arrival time, are varied with pulse rate, it can provide incorrectly estimated clinical information. In order to complement this problem, many previous studies suggested the following correction algorithms, which are widely used in ECG analysis: Bazett's formula, Fridericia's formula, and linear regression equation. Although these correction algorithms has been applied and tested in the ECG signal, they must be validated in the DVP since they can provide different performance in the DVP. Therefore, we have shown a comparison among commonly used correction algorithms for estimation of arterial stiffness in this paper. In order to compare the performance of the correction algorithms, we have carried out the correlation analysis by using a SPSS tool, which is typically used in statistical analysis. As a result, we can notice that it is reasonable to use both the Bazett's formula and linear regression equation for estimation of arterial stiffness by using time-related pulse parameters extracted from the DVP.

Error characteristics of the atmospheric correction algorithms used in retrieval of sea surface temperatures from infrared satellite measurements:: Global and regional aspects

A database of cotemporal, collocated satellite and in situ observations is used to examine the association between the brightness temperature differences measured by the thermal infrared channels (T45) of the Advanced Very High Resolution Radiometer (AVHRR) and water vapor estimates (omega) derived from the Special Sensor Microwave Imager (SSM/I). This channel difference is used to estimate the atmospheric correction (due mostly to water vapor absorption) in sea surface temperature (SST) algorithms. The association between T45 and vis found to be greatest for tropical latitudes;for mid- and high latitudes, the association is best during summer. However, the association tends to decrease toward mid- and higher latitudes during other periods. SST residual errors (satellite 2 buoy) show a negative mean in the Tropics, suggesting undercorrection for water vapor attenuation in the Tropics. This underestimation is explicitly shown for SST residuals in the high-water-vapor regimes of the Arabian Sea. In mid- and high latitudes, the variability of atmospheric water vapor and air-sea temperature difference contributes to the weaker association between T45 and v and results in positive mean SST residual errors. A differential form of SST algorithm that incorporates the use of a "first-guess estimate'' that correlates with SST is observed to give the least residual error.
