This work has evaluated the potential superiority of a morphomolecular classification based on the combination of clinicopathologic and molecular features of colorectal cancers. A cohort of 126 colorectal carcinomas was investigated by unsupervised hierarchical clustering analysis to combine 13 routinely assessed clinicopathologic features and all five molecular markers recently suggested by Jass' classification to distinguish four molecular subtypes of sporadic colorectal carcinomas. Survival analysis was assessed by a Cox proportional hazards model. A clear separation into three prognostically significant groups was identified: cluster A and cluster C were associated with good prognosis and cluster B with poor prognosis (P=0.006). Clinicopathologic and molecular features of cluster A and cluster B tumors were strongly concordant with colorectal cancer profiles characterized by microsatellite instability or by chromosomal instability, respectively. The clinicopathologic features of cluster C tumors were suggestive of a less aggressive disease than cluster B tumors. Genetically, they appeared intermediate between cluster A and cluster B tumors, as they were mainly microsatellite stable tumors showing high levels of both MGMT methylation and loss of heterozygosity. Chromosomal instability was significantly lower in cluster C than in cluster B tumors. A more accurate tumor classification should combine the prognostic power of clinicopathologic parameters with molecular biomarkers that provide information regarding the natural history of the cancer. hierarchical clustering seems to be a useful, promising and powerful tool for further translational studies and should lead us to define a diagnostic and prognostic signature for different carcinomas. Twin and family studies are typically used to elucidate the relative contribution of genetic and environmental variation to phenotypic variation. Here, we apply a quantitative genetic method based on hierarchical clustering, to blood plasma lipidomics data obtained in a healthy cohort consisting of 37 monozygotic and 28 dizygotic twin pairs, and 52 of their biological nontwin siblings. Such data are informative of the concentrations of a wide range of lipids in the studied blood samples. An important advantage of hierarchical clustering is that it can be applied to a high-dimensional 'omics' type data, whereas the use of many other quantitative genetic methods for analysis of such data is hampered by the large number of correlated variables. For this study we combined two lipidomics data sets, originating from two different measurement blocks, which we corrected for block effects by 'quantile equating'. In the analysis of the combined data, average similarities of lipidomics profiles were highest between monozygotic (MZ) cotwins, and became progressively lower between dizygotic (DZ) cotwins, among sex-matched nontwin siblings and among sex-matched unrelated participants, respectively. Our results suggest that (1) shared genetic background, shared environment, and similar age contribute to similarities in blood plasma lipidomics profiles among individuals;and (2) that the power of quantitative genetic analyses is enhanced by quantile equating and combination of data sets obtained in different measurement blocks. Simultaneous Determination of Four Triterpenoid Saponins in Aralia elata Leaves by HPLC-ELSD Combined with hierarchical clustering analysis

Introduction - Aralia elata leaves are known to have several biological activities, including anti-arrythmia, antitumor, antiinflammatory, anti-fatigue, antimicrobial and antiviral effects. Our previous study found that triterpenoid saponins from the leaves of A. elata had antitumor effects. Quantification of the triterpenoids is important for the quality control of A. elata leaves. Objective - To establish high-performance liquid chromatography coupled with evaporative light scattering detection (HPLC-ELSD) for the simultaneous determination of four major triterpenoid saponins, including Aralia-saponin IV, Aralia-saponin VI, 3-O-beta-D-glucopyranosyl-(1 -> 3)-beta-D-glucopyranosyl-(1 -> 3)-beta-D-glucopyranosyl oleanolic acid 28-O-beta-D-glucopyranoside (Aralia-saponin TTP) and Aralia-saponin V. Methodology - The separation was carried out on a Dikma Diamonsil C-18 column (4.6 mm x 250 mm, 5 mu m) efficiently with gradient elution consisting of acetonitrile and water. Results - All calibration curves showed good linear regression (R-2 > 0.9996) within the ranges of tested concentrations. This validated method was applied to determine the contents of the four major triterpenoid saponins in 53 samples from different regions of northeast China. hierarchical clustering analysis was first used to classify and differentiate Aralia elata leaves. Conclusion - The method developed was successfully applied to analyse four major triterpenoid saponins in Aralia elata leaves which is helpful for quality control of the herb. Maintenance and repairing in actual engineering for long-term used structures, such as pipelines and bridges, make structural damage detection indispensable, as an unanticipated damage may give rise to a disaster, leading to huge economic loss. A new approach for detecting structural damage using transmissibility together with hierarchical clustering and similarity analysis is proposed in this study. Transmissibility is derived from the structural dynamic responses characterizing the structural state. First, for damage detection analysis, hierarchical clustering analysis is adopted to discriminate the damaged scenarios from an unsupervised perspective, taking transmissibility as feature for discriminating damaged patterns from undamaged ones. This is unlike directly predicting the structural damage from the indicators manifestation, as sometimes this can be vague due to the small difference between damaged scenarios and the intact baseline. For comparison reasons, cosine similarity measure and distance measure are also adopted to draw out sensitive indicators, and correspondingly, these indicators will manifest in recognizing damaged patterns from the intact baseline. Finally, for verification purposes, simulated results on a 10-floor structure and experimental tests on a free-free beam are undertaken to check the suitability of the raised approach. The results of both studies are indicative of a good performance in detecting damage that might suggest potential application in actual engineering real life.

Fingerprint Chromatogram analysis of Radix Glehniae by LC Coupled with hierarchical clustering analysis

For the first time a validated liquid chromatography method coupled with hierarchical clustering analysis has been developed for the study of fingerprint chromatograms of extracts from the Radix Glehniae of Glehnia littorali. Liquid chromatography with gradient elution was performed on extracts from 23 plant samples collected from different geographical locations. Ten of twenty-three plant samples were selected using hierarchical clustering analysis and employed to establish the fingerprint. The fingerprint was established and ten chromatographic peaks were selected as characteristic peaks and panaxynol was used as reference standard compounds. The fingerprint chromatograms had a good stability, precision, and reproducibility. This method is a very reliable and useful method for assessment of the quality of Radix Glehniae.

Self-adaptable hierarchical clustering analysis and differential evolution for optimal integration of renewable distributed generation

In a previous paper, we have introduced a simulation and optimization framework for the integration of renewable generators into an electrical distribution network. The framework searches for the optimal size and location of the distributed renewable generation units (DG). Uncertainties in renewable resources availability, components failure and repair events, loads and grid power supply are incorporated. A Monte Carlo simulation-optimal power flow (MCS-OPF) computational model is used to generate scenarios of the uncertain variables and evaluate the network electric performance with respect to the expected value of the global cost (ECG). The framework is quite general and complete, but at the expenses of large computational times for the analysis of real systems. In this respect, the work of the present paper addresses the issue and introduces a purposely tailored, original technique for reducing the computational efforts of the analysis. The originality of the proposed approach lies in the development of a new search engine for performing the minimization of the ECG, which embeds hierarchical clustering analysis (HCA) within a differential evolution (DE) search scheme to identify groups of similar individuals in the DE population and, then, ECG is calculated for selected representative individuals of the groups only, thus reducing the number of objective function evaluations. For exemplification, the framework is applied to a distribution network derived from the IEEE 13 nodes test feeder. The results show that the newly proposed hierarchical clustering differential evolution (HCDE) MCS-OPF framework is effective in finding optimal DG-integrated network configurations with reduced computational efforts. Data Fusion of ion Mobility Spectrometry Combined with hierarchical clustering analysis for the Quality Assessment of Apple Essence

A fingerprinting approach was developed by means of high-performance ion mobility spectrometry with direct electrospray ionization (ESI-HPIMS) for the quality consistency and authentication of apple essence which contain many analytes. Thirty-four apple essence samples of the same brand and commercialized as a same product but brewed in four different manufacturers were used to establish the fingerprints. hierarchical clustering analysis (HCA) was performed to evaluate the similarity and variation of these samples. A combined data matrix was constructed with the use of individual data matrix of ion mobility spectrum in positive and negative ion modes. For comparison, ion mobility spectrum fingerprints in positive and negative ion modes respectively were also applied to the quality assessment of the same samples. Finally, our study demonstrated that the fusion of fingerprints did indeed provide more information and could be used to comprehensively conduct the quality consistency evaluation and discrimination of apple essences from similar products. It is suggested that this fingerprint approach is suitable for analysis of other complex, multianalyte substances.

A validated liquid chromatography (LC) method coupled with hierarchical clustering analysis (HCA) has been established for investigating the fingerprint chromatograms of ten kinds of animal horns. The method showed good performances in terms of precision, repeatability, linearity, accuracy, and stability. The results showed that the LC fingerprint method coupled with HCA could be used to classify animal horns, which could make it simple and feasible to search substitution of precious horns.

Chemical Fingerprinting and hierarchical clustering analysis of Centella asiatica from Different Locations in China

Centella asiatica (L.) Urban is a widely distributed herbaceous plant with great medicinal value that has been extensively used in traditional Chinese medicine. For effective quality control of C. asiatica, a feasible approach and control system is necessary. In this paper, a chemical fingerprint method (column liquid chromatography) was developed for investigating and demonstrating the variance of chemical components among different populations of C. asiatica, from 14 locations in China. The LC data showed considerable variation of chemical constituents among C. asiatica populations. Four chemotypes were visually developed from the chromatographic profiles. The hierarchical clustering analysis further suggested that the samples were divided into four major groups. The grouping of 14 C. asiatica populations in hierarchical clustering analysis was in good agreement with the visual comparison of their chromatograms, as demonstrated by chemotypes. In addition, the bioactive compound asiaticoside in this herb was quantitatively determined by a validated reversed-phase liquid chromatography analysis. Chemotype I was identified as asiaticoside-rich chemotype and exhibited significantly higher amounts of asiaticoside. These findings and results provide a solid basis to establish good agriculture practice and select geo-authentic crude drug for C. asiatica. The established method was considered to be suitable for fingerprint analysis for the quality control of C. asiatica.

Comparative studies on Ophiopogonis and Liriopes based on the determination of 11 bioactive components using LC-MS/MS and hierarchical clustering analysis

Radix Ophiopogonis and Radix Liriopes are two widely used herbal medicines and functional foods with similar pharmacological activities and clinical efficacies. In order to figure out whether phytoequivalence of species leads to therapeutic equivalence and consistency, in this study, a novel sensitive and selective high performance liquid chromatography-tandem mass spectrometric method (HPLC-MS/MS) has been developed for the simultaneous analysis of 11 bioactive components (6 steroidal saponins and 5 flavonoids) and then was applied for comparing in Radix Ophiopogonis and Radix Liriopes from different habitats. Separation was performed using a C-18 analytical column with gradient elution of acetonitrile and 0.3% acetic acid (v/v) at a flow rate of 0.8 mL/min. Multiple-reaction monitoring (MRM) scanning was employed with switching electrospray ion source polarity between positive and negative modes in a single run. All calibration curves showed good linearity (r(2) > 0.9915) within the test ranges. The intra-day and inter-day precision for 11 analytes were less than 3.7% and 4.2%, respectively. The recoveries were between 95.3% and 106.7%. Fifty-three batches of commercial samples from different sources were analyzed using the developed method. hierarchical clustering analysis (HCA) and content analysis were performed to differentiate and classify the samples. The results demonstrated that steroidal saponins were the major components in both Liriopes species, and a low content of flavonoids could become an important symbol of distinction between Ophiopogonis and Liriopes. The developed method could successfully differentiate between Ophiopogonis and Liriopes. (C) 2014 Elsevier Ltd. All rights reserved.
