The subtyping of Acute lymphocytic leukemia (ALL) is important for proper treatment strategies and prognosis. Conventional methods for manual blood and bone marrow testing are time-consuming and labor-intensive, while recent flow cytometric immunophenotyping has the limitations such as high cost. Here we develop the deep learning-based light scattering imaging flow cytometry for label-free classification of ALL. The single ALL cells confined in three dimensional (3D) hydrodynamically focused stream are excited by light sheet. Our label-free microfluidic cytometry obtains big-data two dimensional (2D) light scattering patterns from single ALL cells of B/T subtypes. A deep learning framework named Inception V3-SIFT (Scale invariant feature transform)-Scattering Net (ISSC-Net) is developed, which can perform high-precision classification of T-ALL and B-ALL cell line cells with an accuracy of 0.993 +/- 0.003. Our deep learning-based 2D light scattering flow cytometry is promising for automatic and accurate subtyping of un-stained ALL. An extension of quasi-one-dimensional tomography

A method for rapid Limited-angle tomography is suggested that allows the reconstruction of density distribution in hardwood samples with high accuracy and spatial resolution from only a few x-ray projections. The sparsity of the experimental x-ray data is compensated by incorporation of a priori knowledge about generic wood macrostructure, as well as some morphological information obtainable from optical images of the sample surface, in the mathematical framework of the tomographic analysis. Numerical tests of the proposed method confirm its potential as a practical technique for nondestructive testing of trees for scientific and industrial purposes. It can also be useful for the analysis of other types of object with an underlying layered structure. (C) 1998 Optical Society of America.

Computed Tomographic Metal Artifact Reduction for the Detection and Quantitation of Small Features Near Large Metallic Implants: A Comparison of Published Methods

Computed tomographic imaging of tissue surrounding metallic implants is often limited by metal artifacts. This paper compares 3 existing metal artifact reduction techniques that are based on segmentation of metal-affected regions in native images, followed by reprojection of segmented areas into original Radon space, removal of metal trace(s), and renewed reconstruction: Detector row-wise linear interpolation, 2-dimensional interpolation, and combination of row-wise linear interpolation and adaptive filtering. For each method, improvements of CT number accuracy and signal-noise as well as contrast-noise ratios near the prosthesis and in the image periphery over the values found for native images were evaluated in a phantom experiment simulating osteolytic bone lesions of different size and density around a Chrome-Cobalt hip prosthesis stem. Improvement in diagnostic usability was evaluated as lesion detectability by size. Quantitative and qualitative results showed that the linear interpolation and the combination method removed the artifacts most effectively. The mean accuracy error over different regions of interest placed in the direct vicinity of the metal and in the periphery of the object decreased 10-fold with linear interpolation. These methods increased contrast-noise ratio up to 68% of that measured on artifact-free images for the least dense lesion. Qualitatively, the linear interpolation and the combination method improved the lesion delectability and enabled differentiation of different lesion densities. However, in proximity to the stem, some artifacts remained for all methods. We conclude that published algorithms for metal artifact reduction substantially improve image quality for CT imaging of a metallic object and may be adequate for quantitative measurements except for the direct vicinity of the metallic object.

Polarization sensitive optical frequency domain imaging system for endobronchial imaging

A polarization sensitive endoscopic optical frequency domain imaging (PS-OFDI) system with a motorized distal scanning catheter is demonstrated. It employs a passive polarization delay unit to multiplex two orthogonal probing polarization states in depth, and a polarization diverse detection unit to detect interference signal in two orthogonal polarization channels. Per depth location four electro-magnetic field components are measured that can be represented in a complex 2x2 field matrix. A Jones matrix of the sample is derived and the sample birefringence is extracted by eigenvalue decomposition. The condition of balanced detection and the polarization mode dispersion are quantified. A complex field averaging method based on the alignment of randomly pointing field phasors is developed to reduce speckle noise. The variation of the polarization states incident on the tissue due to the circular scanning and catheter sheath birefringence is investigated. With this system we demonstrated imaging of ex vivo chicken muscle, in vivo pig lung and ex vivo human lung specimens. (C)2015 Optical Society of America

OSIS: remote sensing code for estimating aerosol optical properties in urban areas from very high spatial resolution images

The achievement of new satellite or airborne remote sensing instruments enables the more precise study of cities with metric spatial resolutions. For studies such as the radiative characterization of urban features, knowledge of the atmosphere and particularly of aerosols is required to perform first an atmospheric compensation of the remote sensing images. However, to our knowledge, no efficient aerosol characterization technique adapted both to urban areas and to very high spatial resolution images has yet been developed. The goal of this paper is so to present a new code to characterize aerosol optical properties, OSIS, adapted to urban remote sensing images of metric spatial resolution acquired in the visible and near-IR spectral domains. First, a new aerosol characterization method based on the observation of shadow/sun transitions is presented, offering the advantage to avoid the assessment of target reflectances. Its principle and the modeling of the signal used to solve the retrieval equation are then detailed. Finally, a sensitivity study of OSIS from synthetic images simulated by the radiative transfer code AMARTIS v2 is also presented. This study has shown an intrinsic precision of this tool of Delta tau(a) = ***(a) +/- (0.02 + ***(a)) for retrieval of aerosol optical thicknesses. This study shows that OSIS is a powerful tool for aerosol characterization that has a precision similar to satellite products for the aerosol optical thicknesses retrieval and that can be applied to every very high spatial resolution instrument, multispectral or hyperspectral, airborne or satellite. RGB-to-RGBG conversion algorithm with adaptive weighting factors based on edge detection and minimal square error

The sub-pixel arrangement of the RGBG panel and the image with RGB format are different and the algorithm that converts RGB to RGBG is urgently needed to display an image with RGB arrangement on the RGBG panel. However, the information loss is still large although color fringing artifacts are weakened in the published papers that study this conversion. In this paper, an RGB-to-RGBG conversion algorithm with adaptive weighting factors based on edge detection and minimal square error (EDMSE) is proposed. The main points of innovation include the following: (1) the edge detection is first proposed to distinguish image details with serious color fringing artifacts and image details which are prone to be lost in the process of RGB-RGBG conversion;(2) for image details with serious color fringing artifacts, the weighting factor 0.5 is applied to weaken the color fringing artifacts;and (3) for image details that are prone to be lost in the process of RGB-RGBG conversion, a special mechanism to minimize square error is proposed. The experiment shows that the color fringing artifacts are slightly improved by EDMSE, and the values of MSE of the image processed are 19.6% and 7% smaller than those of the image processed by the direct assignment and weighting factor algorithm, respectively. The proposed algorithm is implemented on a field programmable gate array to enable the image display on the RGBG panel. (C) 2018 Optical Society of America

Three-dimensional imaging of stationary and moving targets in turbid underwater environments using a single-photon detector array

Three-dimensional imaging in underwater environments was investigated using a picosecond resolution silicon single-photon avalanche diode (SPAD) detector array fabricated in complementary metal-oxide semiconductor (CMOS) technology. Each detector in the 192 x 128 SPAD array had an individual time-to-digital converter allowing rapid, simultaneous acquisition of data for the entire array using the time-correlated single-photon counting approach. A picosecond pulsed laser diode source operating at a wavelength of 670 nm was used to illuminate the underwater scenes, emitting an average optical power up to 8 mW. Both stationary and moving targets were imaged under a variety of underwater scattering conditions. The acquisition of depth and intensity videos of moving targets was demonstrated in dark laboratory conditions through scattering water, equivalent to having up to 6.7 attenuation lengths between the transceiver and target. Data were analyzed using a pixel-wise approach, as well as an image processing algorithm based on a median filter and polynomial approximation. Intraoperative optical coherence tomography is still not overly pervasive in routine ophthalmic surgery, despite evident clinical benefits. That is because today's spectral-domain optical coherence tomography systems lack flexibility, acquisition speed, and imaging depth. We present to the best of our knowledge the most flexible swept-source optical coherence tomography (SS-OCT) engine coupled to an ophthalmic surgical microscope that operates at MHz A-scan rates. We use a MEMS tunable VCSEL to implement application-specific imaging modes, enabling diagnostic and documentary capture scans, live B-scan visualizations, and real-time 4D-OCT renderings. The technical design and implementation of the SS-OCT engine, as well as the reconstruction and rendering platform, are presented. All imaging modes are evaluated in surgical mock maneuvers using ex vivo bovine and porcine eye models. Synthetic hyperspectral array video database with applications to cross-spectral reconstruction and hyperspectral video coding

In this paper, a synthetic hyperspectral video database is introduced. Since it is impossible to record ground-truth hyperspectral videos, this database offers the possibility to leverage the evaluation of algorithms in diverse appli-cations. For all scenes, depth maps are provided as well to yield the position of a pixel in all spatial dimensions as well as the reflectance in spectral dimension. Two novel algorithms for two different applications are proposed to prove the diversity of applications that can be addressed by this novel database. First, a cross-spectral image reconstruction algorithm is extended to exploit the temporal correlation between two consecutive frames. The evaluation using this hyperspectral database shows an increase in peak signal-to-noise ratio (PSNR) of up to 5.6 dB dependent on the scene. Second, a hyperspectral video coder is introduced, which extends an existing hyperspectral image coder by exploiting temporal correlation. Modern optical sensors and measurement systems usually are a powerful combination of optical elements, active hardware components like actuators or sensing devices as well as a sophisticated control software and data evaluation algorithms. In order to develop and operate such systems, it is necessary to have a flexible, intuitive, and fast underlying software framework that also allows for rapid prototyping of a sensor in a dynamic lab environment. This software must be able to control and communicate with all necessary hardware devices and has to provide all the highly performant evaluation, data, and image processing algorithms required. In this publication, we want to present the open source measurement and data evaluation software suite itom, which has been designed considering the denoted requirements and whose development began in 2011.
