Microsphere-Enabled micropillar array for Whispering Gallery Mode Virus Detection

Rapid detection of pathogens and analytes at the point of care offers an opportunity for prompt patient management and public health control. This paper reports an open microfluidic platform coupled with active whispering gallery mode (WGM) microsphere resonators for the rapid detection of influenza viruses. The WGM microsphere resonators, precoated with influenza A polyclonal antibodies, are mechanically trapped in the open micropillar array, where the evaporation-driven flow continuously transports a small volume (similar to mu L) of sample to the resonators without auxiliaries. Selective chemical modification of the pillar array changes surface wettability and flow pattern, which enhances the detection sensitivity of the WGM resonator-based virus sensor. The optofluidic sensing platform is able to specifically detect influenza A viruses within 15 min using a few microliters of sample and displays a linear response to different virus concentrations.

A MEMS-Based Electrochemical Angular Accelerometer Based on Planar Electrodes With micropillar array Structure

MEMS-based electrochemical angular accelerometers based on planar electrodes with micropillar array structure were reported in this article, which converted external angular acceleration into an electrical signal based on the liquid inertial mass and the varied concentrations of reactive ions. Theoretical analysis and numerical simulations of the angular accelerometer were carried out, where the key structures of the planar electrode were optimized. Based on microfabrication, electrochemical angular accelerometers based on planar electrodes with micropillar array structure were fabricated and characterized, and the test results were consistent with the numerical simulation. When the area of the electrodes was increased by a factor of 1.67, the sensitivity level of the devices based on planar electrodes with micropillar array structure was about 1.60-1.78 times higher in the frequency range of 0.83-10 Hz than that of the devices without micropillar array, and the sensitivity level at 10 Hz was improved by more than 70%, which is essential for expanding the bandwidth and controlling the noise level of electrochemical angular accelerometers. In summary, the MEMS-based electrochemical angular accelerometers reported in this article can provide a new perspective for performance optimization based on integrated planar electrodes, demonstrating the potential of MEMS-based electrochemical angular accelerometers for applications in seismology.

Hydrogel micropillar array for Temperature Sensing in Fluid

Localized temperature sensing and control on a micrometer-scale have diverse applications in biological systems. We present a micrometer-sized hydrogel pillar array as potential temperature probes and actuators by exploiting sensitive temperature dependence of their volume change. Soft lithography-based molding processes were presented to fabricate poly N-isopropyl acrylamide (p-NIPAAm)-based hydrogel pillar array on a glass substrate. Au nanorods (Au-NDs) as light-induced heating elements were embedded within the hydrogel pillars, and near-infrared (NIR) light was used to modulate temperature in a local area. First, static responses of the micropillar array were characterized as a function of its temperature. It was shown that the hydrogel had a sensitive volume transition near its low-critical solution temperature (LCST). Furthermore, we showed that LCST could be readily adjusted by utilizing copolymerizing with acrylamide (AAm). To demonstrate the feasibility of spatiotemporal temperature mapping and modulation using the presented pillar array, pulsed NIR light was illuminated on a local area of the hydrogel pillar array, and its responses were recorded. Dynamic temperature change in water was mapped based on the abrupt volume change characteristics of the hydrogel pillar, and its potential actuation using NIR light was successfully demonstrated. Considering that the structure can be arrayed in a 2-D pixel format with high-spatial resolution and high-sensitive temperature characteristics, the presented method and the device structure can have diverse applications to change and sense local temperatures in liquid. This is particularly useful in biological systems, where their physiological temperature can be modulated and mapped with high spatial resolution.

Photoresponse Enhancement in Ge MSM Photodetector With Ge micropillar array

Improving detection efficiency of Ge photodetector near the absorptance edge is crucial for fiber-optic telecommunication. Here, we report the unique photodetection properties of germanium (Ge) metal-semiconductor-metal (MSM) photodetector with Ge micropillar array. The responsivity and detectivity of the device at 1550 and 1990 nm are studied. When compared with the Ge photodetector without micropillar, the responsivity and detectivity of micropillar Ge device are improved by 2.76 and 4.21 times at 1550 nm, respectively. This phenomenon is mainly associated with the enhanced absorption efficiency by geometrical light trapping effect and guided modes supported in the micropillar array. Particularly, the values of responsivity and detectivity in the device with Ge micropillar array are enhanced over 29.9 and 37.1 times at 1990 nm than the results in the device without micropillar, which are quite larger than the values at 1550 nm. This feature is related to that the suitable position of absorption enhancement caused by the micropillar array, which can result in the efficiently separation and suppressed recombination of photogenerated carriers under bias voltage. This work provides a feasible approach to broaden the detection wavelength of Ge-based photodetectors, enhance the absorption near the absorption edge, and improve the detection performance.

Glucose levels serve as a fundamental indicator of cell health, reflecting crucial aspects of cellular metabolism and energy production. While effective, traditional methods such as spectrophotometry and chromatography have limitations, such as labour-intensive sample collection, reliance on bulky equipment, extensive sample preparation, and prolonged experimental durations. To address these issues, we introduce a micropillar-based microfluidic electrochemical device (MED) for real-time monitoring of glucose levels in diverse cell culture systems, including human induced pluripotent stem cells (hiPSCs) and murine fibroblast cells (GP + E86). This biosensor demonstrates a linear range of 0.025-1.50 mM and a high sensitivity of 4.71 +/- 0.13 mu A. mM- 1, and a low limit of detection of 19.10 +/- 0.50 mu M. The MED not only delivered fast glucose measurements with accuracy and reliability comparable to ultra-high-performance liquid chromatography (UHPLC) but was also specifically evaluated on GP + E86 murine fibroblast cells at varying seeding densities (1:5 and 1:10 ratios), across different culturing times to accurately monitor dynamic metabolic shifts associated with various growth phases. Furthermore, the MED effectively detected significant changes in glucose consumption in hiPSCs cell cultures contaminated with Escherichia coli (E. coli), highlighting its potential for early contamination detection. Integrating non-invasive, continuous monitoring platforms enhances the reliability of experimental outcomes by enabling cell health monitoring without disrupting the cell culture process. This approach enables real-time monitoring of cell cultures ensuring accurate detection of metabolic changes and early detection of media contamination.

The Shape Effect of Acoustic micropillar array Chips in Flexible Label-Free Separation of Cancer Cells

The precise isolation of circulating tumor cells (CTCs) from blood samples is a potent tool for cancer diagnosis and clinical prognosis. However, CTCs are present in extremely low quantities in the bloodstream, posing a significant challenge to their isolation. In this study, we propose a non-contact acoustic micropillar array (AMPA) chip based on acoustic streaming for the flexible, label-free capture of cancer cells. Three shapes of micropillar array chips (circular, rhombus, and square) were fabricated. The acoustic streaming characteristics generated by the vibration of microstructures of different shapes are studied in depth by combining simulation and experiment. The critical parameters (voltage and flow rate) of the device were systematically investigated using microparticle experiments to optimize capture performance. Subsequently, the capture efficiencies of the three micropillar structures were experimentally evaluated using mouse whole blood samples containing cancer cells. The experimental results revealed that the rhombus microstructure was selected as the optimal shape, demonstrating high capture efficiency (93%) and cell activity (96%). Moreover, the reversibility of the acoustic streaming was harnessed for the flexible release and capture of cancer cells, facilitating optical detection and analysis. This work holds promise for applications in monitoring cancer metastasis, bio-detection, and beyond.

An optofluidic sensor for rapid detection of gasphase analytes from a flowing micro-volume fluid sample towards wearable colorimetric sensing is reported. The sensor is comprised of a metal-insulator-metal (MIM) thin-film structure integrated with a quartz micropillar array for improved delivery of vapor analytes onto the sensing surface. For our proof-ofprinciple demonstration, we utilized a poly(methyl methacrylate) (PMMA) spacer and evaluated its response to ethanol vapors that were delivered in close proximity to the sensing material via the micropillar array. It is demonstrated that the micropillar array reduces the sensor response time from minutes to seconds compared with experiments using a conventional chamber. The sensor's concentration-dependent response also confirms its potential for continuous, compact, and quantitative colorimetric analysis of volatile analytes in low-volume samples.

Numerical Investigation of a Microfluidic Biosensor Based on I-Shaped micropillar array Electrodes

micropillar array electrodes offer several advantages, such as enhanced mass transport, lower detection limits, and the potential for miniaturization, making them instrumental in the design and fabrication of electrochemical biosensors. The performance of these biosensors is influenced by electrode geometry, including parameters like shape and height, which affect surface area and overall sensitivity. In this study, we designed a microfluidic electrochemical biosensor featuring micropillar array electrodes, modeled in COMSOL Multiphysics. We compared the response currents of I-shaped and cylindrical micropillar array electrodes. The working electrode (WE), consisting of 100 micropillars with a height of 300 mu m, exhibited a sensitivity of 1.61 mu Acm2mM-1 in cyclic voltammetry, highlighting its effectiveness for analyte detection.

Adhesion Enhancement of micropillar array by Combining the Adhesive Design from Gecko and Tree Frog

It has long been demonstrated the gecko-inspired micropillar array with T-shape tips possesses the best adhesion performance of a given material. The further enhancement of the adhesion performances of T-shape micropillars can offer redundant adhesion to compensate for the inevitable improper contacts. Here, the array of T-shape polydimethylsiloxane (PDMS) micropillars is incorporated with gradient dispersed calcium carbonate nanoparticles in the micropillar stalk, termed as T-shape gradient micropillars (TG), possessing the modulus gradient with stiff tip and soft root. The gradient modulus in TG facilitates the contact formation and regulates the stress at the detaching interface, resulting in a 4.6 times adhesion and 2.4 times friction as compared with the pure PDMS T-shape micropillar arrays. The study here provides a new design strategy for the super-strong structured dry adhesives.
