Peripheral nerve injury (PNI) can result in severe disabilities, profoundly impacting patients' quality of life and potentially endangering their lives. Therefore, understanding the potential molecular mechanisms that facilitate the regeneration of damaged nerves is crucial. Evidence indicates that Schwann cells (SCs) play a pivotal role in repairing peripheral nerve injuries. Previous studies have shown that rna, particularly non-coding rna (ncrna), plays a crucial role in nerve regeneration, including the proliferation and dedifferentiation of SCs. In this review, the individual roles of ncrna in SCs and PNI are analyzed. This review not only enhances the understanding of ncrna's role in nerve injury repair but also provides a significant theoretical foundation and inspiration for the development of new therapeutic strategies. After a PNI, nCrna plays a crucial role in the repair process involving SCs and the injured nerve. In this review, the individual roles of nCrna in SCs and PNI are analyzed. This review not only enhances the understanding of ncrna's role in nerve injury repair but also provides a significant theoretical foundation and inspiration for the development of new therapeutic strategies. image

Brain cancer remains a formidable challenge with limited treatment options. non-coding rnas (ncrnas) have emerged as promising biomarkers due to their dysregulation in tumorigenesis. This review explores the potential of biosensors for early detection of brain cancer by targeting ncrnas. We discuss the classification and functions of ncrnas, emphasizing their involvement in key cancer-related processes. Additionally, we delve into recent advancements in biosensor technology, focusing on their ability to accurately detect specific ncrna biomarkers associated with brain cancer. Our findings underscore the potential of biosensors to revolutionize brain cancer diagnosis, enabling personalized medicine and improving patient outcomes. Future research should focus on refining biosensor technology and expanding their clinical application.

N6-methyladenosine (m6A) and non-coding rna (ncrna) play important roles in the occurrence, development, and prognosis of multiple myeloma (MM). They not only affect stemness, growth, and apoptosis of MM cells but also intervene in MM proliferation, migration, invasion, and even drug resistance. It is also a prognostic factor for poor MM survival. Therefore, in-depth research on the mechanisms of m6A and ncrna in MM significant for diagnosis, treatment, and prognosis of MM.

Dynamic non-coding rna biomarker reveals lung injury and repair induced by polystyrene nanoplastics

Polystyrene nanoplastics (PS-NPs) are omnipresent in the air and can be inhaled by humans. However, their longterm adverse implications and toxicological mechanisms for human respiratory health are unclear. Therefore, this study aims to provide new insights into the pulmonary toxicity of PS-NPs using mice and organoid models. After subacute and subchronic inhalation of PS-NPs, mice showed pronounced lung injury characterized by respiratory rate changes, altered hematology, and histological evidence of tissue damage and oxidative stress. Similarly, repeated PS-NPs exposure also restricted organoid growth and cause oxidative damage. Notably, through Bisquerna analysis for a single-cell dataset and canonical markers verification, it was found that PS-NPs induced the emergence and accumulation of transitional cells, suggesting impaired alveolar epithelial repair processes. Sequencing analyses revealed dynamic alterations in non-coding rna (ncrna) profiles, including circrnas and lncrnas, in response to PS-NPs exposure. Moreover, temporal profiling highlighted distinct sets of ncrnas as early and progression-associated biomarkers of PS-NP-induced lung injury. These biomarkers correlated with aberrant transitional cells, implicating their roles in disrupted cellular differen tiation and repair mechanisms. Overall, this study observed the multifaceted toxicological responses of PS-NPs to the respiratory system, emphasizing the critical involvement of ncrnas in mediating PS-NP-induce transitional cells, which was crucial for elucidating the pathophysiology of nanoplastic-induced lung injury and developing targeted therapeutic strategies.

Programmed cell death (PCD), an essential and inevitable phenomenon, is integral to organismal development. It not only maintains cellular homeostasis but also prevents aberrant cell proliferation, thereby protecting normal growth and development from detrimental factors. And it is governed by a highly complex and sophisticated regulatory network in which non-coding rnas (ncrnas) play pivotal roles. ncrnas refer to rna molecules that do not encode proteins and encompass various types, including long non-coding rna (lncrna), microrna (mirna), and circular rna (circrna). Herein, we investigate the specific signaling mechanisms by which ncrnas regulate stem cells, elucidating their role in modulating PCD process through interactions with specific molecules. We further summarize the impact of above modulating role on stem cell differentiation, proliferation, cycle regulation and diverse disease development and therapy. Additionally, given the emerging trends in the therapeutic application of ncrnas and stem cells, we explore the potential of their combined application for disease treatment. © 2025 The Authors

Systemic sclerosis (SSc) is a chronic autoimmune disease characterized by imbalanced immunity, vasculopathy, and excessive fibrosis. The etiology and pathology of this disease remain to be fully elucidated. Genetic predisposition, along with epigenetic modifications are widely considered to significantly affect its development. Among the components of epigenetics, non-coding rnas (ncrnas), comprising various types such as microrna, long ncrna, circular rna, and others, play a crucial role. These ncrnas perform several functions in the development of SSc and can potentially be employed as new targets for its diagnosis and treatment. This review discusses the roles of ncrnas in the three primary pathological hallmarks-vasculopathy, imbalanced immunity, and excessive fibrosis-of SSc and highlights research progress in the role of rnas in translational medicine against SSc. The review also provides a comprehensive outline of the key function of ncrnas in SSc.

Characterization of a small non-coding rna S612 in Bacillus subtilis

Small regulatory rnas (srnas) are non-coding rna molecules that fine-tune various cellular processes and respond to various environmental stimuli. In Bacillus subtilis, the regulatory mechanisms and specific targets of several srnas remain largely unknown. In this study, we identified and characterized S612 as a self-terminating srna in B. subtilis. The expression of S612 is regulated by external signals, including nutrient availability and salt concentration. Overexpression of S612 induced filamentous cells with extensive cellular elongation and complete inhibition of sporulation, indicating its potential to control cell morphology and spore formation. S612 directly targets and downregulates genes through post-transcriptional base pairing with mrnas, including ylmD, trpE, ycxC, yycS, rapH, and amyE, some of which are involved in cell membrane integrity, cell wall synthesis, and sporulation initiation. Therefore, we propose that S612 is an important post-transcriptional regulator of cell morphology and sporulation. © 2024

non-coding rnas (ncrnas) are involved in several different regulatory pathways including reproduction. In teleost fish, efficacious reproduction is heavily dependent on the completion of the reproductive cycle. The presence of ncrna, however, and their expression dynamics and putative regulatory role in mature and immature gonads have not yet been extensively explored. Therefore, the abundance of ncrnas in mature and immature female sharpsnout seabream (Diplodus puntazzo) was investigated. The sharpsnout seabream is a rudimentary hermaphrodite which, in captivity, displays dysfunctions in the gonad maturation process. Our analyses revealed a gonad specific read length distribution with two main peaks representing mirnas (21-26 nt) and PIWI rna (27-34 nt). Besides, distinct expression patterns for several ncrna biotypes including micrornas (mirnas), PIWI rnas (pirnas), and ribosomal rnas (rrnas) were detected. Identified mirna accounted to 938, corresponding to similar to 13% of obtained transcripts. Among the differential expressed ncrnas, 10 (similar to 7%) were annotated as mirna, out of which 2 were found in higher abundance in immature gonads (miR-125c and miR-24) and 8 (miR-451, miR-7a, miR-122-1, miR190a, miR129, ENSGACT00000029608, ENSGACT00000029489, and ENSGACT00000029667) were found to be higher expressed in mature gonads. Putative mirna targets, including long non-coding rnas (lncrnas) and genes, are proposed. Target genes are involved in several processes of fish oocyte development, such as steroidogenesis, proteolysis, and apoptosis, and may explain hormone regulation. This study demonstrates a gonad maturation biased ncrna profile which in turn may support the role of ncrnas in ovarian physiology and reproductive performance of fish, stressing the specific function of each rna biotype in oocyte development.

Alzheimer's disease (AD) is a neurodegenerative disorder characterized by complex pathophysiological features. Amyloid plaques resulting from extracellular amyloid deposition and neurofibrillary tangles formed by intracellular hyperphosphorylated tau accumulation serve as primary neuropathological criteria for AD diagnosis. The activation of microglia has been closely associated with these pathological manifestations. non-coding rna (ncrna), a versatile molecule involved in various cellular functions such as genetic information storage and transport, as well as catalysis of biochemical reactions, plays a crucial role in microglial activation. This review aims to investigate the regulatory role of ncrnas in protein expression by directly targeting genes, proteins, and interactions. Furthermore, it explores the ability of ncrnas to modulate inflammatory pathways, influence the expression of inflammatory factors, and regulate microglia activation, all of which contribute to neuroinflammation and AD. However, there are still significant controversies surrounding microglial activation and polarization. The categorization into M1 and M2 phenotypes may oversimplify the intricate and multifaceted regulatory processes in microglial response to neuroinflammation. Limited research has been conducted on the role of ncrnas in regulating microglial activation and inducing distinct polarization states in the context of neuroinflammation. Moreover, the regulatory mechanisms through which ncrnas govern microglial function continue to be refined. The current understanding of ncrna regulatory pathways involved in microglial activation remains incomplete and may be influenced by spatial, temporal, and tissue-specific factors. Therefore, further in-depth investigations are warranted. In conclusion, there are ongoing debates and uncertainties regarding the activation and polarization of microglial cells, particularly concerning the categoriza-tion into M1 and M2 phenotypes.
