The liver plays a key role in lipid metabolism as the hub of fatty acid synthesis and lipid circulation through lipoprotein *** an abundance of dietary fat intake,excessive fat accumulates in the blood, causing hyperlipidemia,and in the liver cells as lipid droplets,resulting in hepatic ***,we reported that in SRC-3-/- mice,the serum level of TC,TG and lipoprotein HDL were significantly lower than their wild type littermates under a normal *** diet induced obesity model,SRC-3-/- mice were protected from hyperlipidemia and fatty liver compared to SRC-3+/+ *** investigation revealed that the expression level of PPARα,a master gene governing lipid metabolism,was elevated while its negative regulator COUP-TFⅡwas decreased in livers of SRC-3-/- *** the molecular level,under all-trans retinoic acid treatment,SRC-3 interacted with retinoic receptor a and bound to -628/-448 region of the COUP-TFⅡpromoter to activate its expression ***,these findings suggest,for the first time,a crucial role of SRC-3 in lipid homeostasis.
