目的:建立一种用SYBR Green Ⅰ荧光染料检测乙肝病毒的实时聚合酶链反应(PCR)方法。
方法:应用SYBR Green Ⅰ染料掺入双链DNA的原理,并对实时荧光定量PCR(real-time PCR)反应条件进行优化,验证方法的特异性、敏感性和稳定性,定量检测HBV DNA水平。以抗乙肝药物拉米夫定为例证明该方法。
结果:建立了定量检测HBV DNA的方法。
结论:应用SYBR Green Ⅰ对HBV DNA的相对定量具有简便、快速、重复性好、特异性强的特点,适用于抗HBV药物的大样品筛选及评价优化工作,并可发展成为高通量技术。

A reverse-phase HPLC method was developed for the simultaneous separation and determination of five bioactive phenolic acids,yunnaneic acid E,rosmarinic acid,lithospermic acid,salvianolic acid B and salvianolic acid A in eight different samples of Salvia yunnanensis collected in Yunnan *** comparison,the sample of Salvia miltiorrhiza was included.
