Purpose: Pancreatic ductal adenocarcinoma, one of the most lethal malignancies, compromises the performance of traditional therapeutic regimens in the clinic because of stromal resistance to systemic drug delivery and poor prognosis caused by tumor metastasis. In recent years, tumor immunotherapy has achieved exciting therapeutic effects in a variety of tumors, such as melanoma, breast cancer and non-small cell lung cancer, but immunotherapy strategies for pancreatic cancer are still being explored. However, due to the unique tumor microenvironment of pancreatic cancer, the effect of immunotherapy is limited. Therefore, a biocompatible therapeutic paradigm that can effectively inhibit pancreatic tumor growth while simultaneously eliminating tumor metastasis is urgently ***: Supramolecular TP5-ICG nanofibrils(TP5-ICG NFs) are fabricated through coassembly of clinically approved immunomodulatory thymopentin(TP5) and near-infrared indocyanine green(ICG). The orthotopic pancreatic tumor-bearing C57BL/6 mice were intratumorally injected with TP5-ICG NFs, free ICG and PBS and then all the tumors received localized photothermal therapy using the laser irradiation. Therapy effects on tumor volumes were monitored over three weeks using multiple imaging modalities, including magnetic resonance imaging(MRI) and bioluminescence imaging(BLI). Flow cytometry for DC cells and T cells of splenocytes and histology analysis were performed to confirm the immunomodulation ***: TP5-ICG NFs showed improved photophysical capabilities for immunomodulation efficacy, fluorescence imaging, and photothermal conversion in vitro. In vivo BLI and MRI both indicated that TP5-ICG NFs can effectively inhibit tumor growth and distant metastasis. Flow cytometry of mouse splenocytes confirmed the proliferation and differentiation of antitumor immune cells after injection with TP5-ICG NFs. 建立了一种从Cohn血浆分离副产物组分IV中纯化高纯度转铁蛋白(Tf)标准物质的技术,对Cohn组分IV进行复溶、离心和过滤,采用Capto DEAE阴离子交换色谱进行初分离,再通过穿透式Octyl-Sepharose 4 Fast Flow疏水色谱进一步精制;对纯化产品进行了结构、纯度、活性等表征,采用十二烷基磺酸钠-聚丙烯酰胺凝胶电泳、基质辅助激光解吸电离飞行时间质谱、高效液相色谱及酶联免疫吸附法对Tf标准物质进行了分析.结果表明,Tf产品纯度大于99%,收率为79%;圆二色光谱分析显示Tf的二级结构未发生改变,铁离子结合能力实验表明Tf可结合2个铁离子,保持活性.
