目的以去乙酰化酶抑制剂丁酸钠(Na Bu)和乙酰转移酶p300作为乙酰化作用因素,探讨人胚肾HEK293细胞中乙酰化对二甲基精氨酸二甲胺水解酶1(DDAH1)表达水平的调节。方法应用real-time PCR方法检测Na Bu刺激或转染野生型及突变型p300表达载体后DDAH1 m RNA表达水平的变化;应用Western blot方法检测Na Bu和p300对DDAH1蛋白表达水平的影响。结果 Real-time PCR结果显示Na Bu以时间依赖方式上调DDAH1 m RNA表达水平,同时转染野生型p300表达载体显著上调DDAH1 m RNA水平,而突变型p300作用不明显;Western blot结果显示Na Bu及野生型p300可提高DDAH1蛋白表达,但突变型p300无此作用,与对DDAH1 m RNA表达的作用一致。结论乙酰化可上调HEK293细胞中DDAH1 m RNA和蛋白表达水平。
