Airway functions are under the control of the vagus nerve which centrally originates from airway-related vagal preganglionic neurons(AVPNs)in the nucleus ambiguus(NA)and the dorsal motor nucleus of the vagus(DMV)in medulla *** AVPNs may be regulated by orexin,a peptide produced by hypothalamic neurons and proved to regulate multiple physiological functions including *** this study,we investigated 1)the expression of orexin receptors in the AVPNs which were retrogradely labeled by fluorescent injected into the trachea wall;2)the effect of orexin on the AVPNs in brainstem slices using patch-clamp techniques;3)the physiological effects of centrally administrated orexin in anesthetized *** found that the AVPNs in the NA were stained for orexin 1 receptors;bath application of orexin-A increased the excitability of AVPNs and enhanced the synaptic inputs of AVPNs,including glutamatergic,GABAergic and glycinergic transmissions;central administration of orexin-A into the cisterna magna of juvenile rats produced a significant increase in airway resistance(RL and Re)and a significant decrease in dynamic compliance(Cdyn).These results indicate that orexin regulates the activities of AVPNs and have a role in the control of airway functions.
