目的探讨全反式维甲酸(all-trans retinoic ac id,ATRA)联合三羟异黄酮(Gen iste in)抑制肺癌细胞的增殖和促进其凋亡的作用。方法采用不同剂量ATRA、Gen iste in及两者联合处理人肺腺癌细胞株A549,观察A549细胞生长情况并运用MTT法检测其生长抑制情况,流式细胞仪进行细胞周期分析及凋亡率检测。结果A549肺癌细胞经ATRA、Gen iste in及两者联合处理后,细胞增殖能力明显下降,更多的细胞被阻滞于G2/M期,细胞凋亡明显增加,其中以两者联合用药最为显著。结论ATRA联合Gen iste in能协同抑制肺癌A549细胞的恶性增殖并促进其凋亡。

目的利用大肠杆菌双杂交系统筛选脑组织中与大鼠AMP激活的蛋白激酶α2(AMP-activated protein kinaseα2,AMPKα2)相互作用的蛋白质,以期进一步探讨AMPKα2在脑内的生物学功能及调节机制。方法以重组质粒pBT-AMPKα2作为诱饵,应用大肠杆菌双杂交系统筛选大鼠胎脑cDNA文库,并对结果进行生物信息学分析。结果筛选出与AMPKα2结合的蛋白7个,包括聚合泛素、热休克蛋白8(heat shock protein8,HSP8)、磷酸果糖激酶、细胞色素C氧化酶亚基Ⅰ(cytochrome C oxidase subunitⅠ,COXⅠ)、人类白细胞抗原B关联性转录物3(HLA-B-associatedtranscript3,BAT3)异构体1、蛋白酪氨酸磷酸酶受体型D(protein tyrosine phosphatase receptor type D,Ptprd)、岛-脑蛋白1(islet-brain1,IB1)。结论寻找到了脑内与AMPKα2相互作用的7种蛋白:聚合泛素、HSP8、磷酸果糖激酶、COXⅠ、BAT3异构体1、Ptprd、IB1。

The overall cellular gene expression is well realized by the coordinated regulation between nuclear DNA and mitochondria DNA. Coordinated regulation between two genomes plays a key role not only in mitochondria biogenesis and protein synthesis but also more importantly in the regulation of gene expression of mitochondria respiratory chain subunits in order to mediate the mitochondrial respiratory function. In the present review, we focus on the mechanism of coordinated regulation between two genomes.
