目的:探究当前α-突触核蛋白(α-synuclein,αS)研究领域的研究热点和发展趋势。方法:以1998至2017年Web of Science核心合集数据库中收录的10 991篇αS相关文献为数据来源,利用CiteSpace V进行可视化处理和分析,梳理并揭示αS研究的知识基础、研究热点与研究前沿。结果:αS研究发文量和引文量随年份呈指数增长,美国是αS研究领域的主要研究力量;αS研究的高频共被引文献主要关注αS与路易小体的关联、αS基因突变、αS聚集以及帕金森病Braak分期等问题。αS研究的主要研究热点集中于αS相关疾病研究、体外研究、帕金森病致病机制研究等。αS研究前沿和未来研究趋势将围绕在帕金森病生物标志物、自噬等致病机制、神经保护治疗以及小鼠模型上。国内学者在αS研究领域更多关注帕金森病的致病机制及动物模型研究。结论:目前国际上有关αS的研究正处在快速发展的阶段,本研究分析了αS研究领域的知识基础、研究热点和前沿,揭示了近二十年来该研究领域的动态发展规律,为相关学科研究提供了参考。

Recent increasing restrictions on animals have persuaded researchers to seek in vitro alternatives that could reduce the use of experimental animals and provide a unique methodology for the exploration of specific toxicant mechanisms in *** various in vitro models have been reported,their application in toxicological studies has been limited due to various *** the present study,an improved three-dimensional rat testicular cell co-culture model was established by the addition of KSR to the culture medium instead of *** effects of KSR and FBS were assessed and followed by an assessment of the effects of BPA on somatic cell function and germ cell differentiation in vitro using this improved co-culture *** was found that the addition of KSR could enhance cell proliferation and germ cell meiosis,which confirmed the functional integrity of the co-culture ***,the effects of BPA on the in vitro model demonstrated that the model could successfully mimic in vivo testis physiology *** concluded that the modified model could be used to evaluate various chemicals on rat testis cells and investigate the specific mechanisms of toxicant activity in the testis in future studies.

The purpose of the present research is to further validate the application of the improved 3D rat testicular cell co-culture model to evaluate the effects of various reproductive toxic chemicals on the function of the main somatic cells,as well as on spermatogonial cell differentiation and even spermatogenesis,and to investigate the specific toxicant mechanisms in testes with HZ 1006,a hydroxamate-based HD *** to the characteristics of HZ 1006,the appropriate exposure duration(8,16 and 24 days),dosage(0,3.125,6.25,12.5 and 25 μM) and toxic endpoints suitable for detection were selected in the *** results showed inhibition of cell proliferation,reduced testosterone level and decreased spermatogonial cell meiosis-specific gene expression,as well as decreased Ar protein level and Ar target gene PSA expression,accompanied by inhibition of Hdac6 expression after HZ 1006 exposure in the 3D rat testicular cell co-culture *** findings indicate that the improved 3D rat testicular cell co-culture model we have established has the potential to become a new testicular toxicity test system that can be used to test toxic characteristics and mechanisms of new compounds and has good application prospects,although it is necessary to do more research on the model.
