7,12-dimethylbenz(a)anthracene induced breast cancer in Sprague Dawley rats proved by ultrasound imaging

Many studies have shown that some of cancer animal models have been established to study various aspects of cancer biology, including breast cancer. Substantial evidence has shown that administration of 7,12-dimethylbenz(a)anthracene (DMBA) in animal models can mimic human breast cancer in many ways. This preliminary research aimed to develop rat breast cancer models that will be used for a further study on phytotherapy as the alternative medicine for breast cancer. Four young Sprague Dawley female rats, aged 60 days, received a single subcutaneous injection of 25 mg DMBA mixed with 0.5 mL sunflower oil and 0.5 mL sodium chloride 0.9%. Control group received 0.5 ml of sunflower oil and 0.5 ml of saline only. Tumor detection by palpation and rats’ body weight measurement were performed every week after DMBA induction. At 10-weeks after DMBA induction, the tumor development was examined by ultrasound imaging. No significant differences were observed in the body weight between DMBA-induced rats and non-induced rats during 8-weeks after induction. Starting from week ninth, the body weight of the DMBA group significantly decreased (p=0.02) compared to the counterpart. A solid mass on the breast area was firstly detected at week seventh in one rat. Three weeks later the solid masses were detected in all DMBA-induced rats that proved by the ultrasound images showing hypoechoic lesions with irregular margin and calcification indicating malignancies. DMBA induced malignant lesions in female rats that demonstrated by ultrasound imaging.

Blood chemistry profiles of DMBA-induced mammary tumor in female sprague dawley rats

Although aerobic glycolysis (Warburg effect) in cancers had been accepted as a significant characteristic of cancer, changes in metabolic state in each stage of tumorigenesis remain to be explored. In this preliminary study, we analyzed the changes of several blood chemistry parameters intending to describe the general metabolic state of the early and advanced stages of tumorigenesis in rat mammary tumor models. The mammary tumor was induced in female Sprague Dawley (SD) rats by subcutaneous injection of 7, 12-dimethylbenz[a]anthracene (DMBA). The rats were palpated on the breast every week after DMBA induction to detect tumor development. Blood chemistry examinations including blood glucose, triglyceride, cholesterol, HDL-cholesterol, urea, and creatinine, were performed at 8-weeks and 12-weeks after DMBA induction. Rats were fasted 12-14 hours before retro-orbital blood collection. The spectrophotometer measured all the parameters. The palpable mammary tumor was firstly detected at 7-weeks on 1 of 4 DMBA induced rats. At three weeks later all rats had palpable tumors that also confirmed by ultrasound imaging. The blood chemistry measurements revealed that DMBA-induced rats had a significantly higher level of triglyceride (p=0.001) and creatinine (p=0.018); and glucose (p=0.007) at 8-weeks and 10-weeks after DMBA induction compared to the non-induced rats, respectively while not in the other parameters. The stages of tumor development may have a different profile of blood chemistry parameters. Further research should be performed to elucidate whether the profile could describe the metabolic reprogramming during tumorigenesis.

The study aimed to evaluate the impact of occupational noise on hearing loss among healthcare workers using audiometry. A longitudinal study was conducted with a six-month follow-up period in a hospital with 21 participants, divided into high-noise-exposure (HNE) and low-noise-exposure (LNE) groups. Mean noise levels were higher in the HNE group (70.4 ± 4.5 dBA), and hearing loss was measured using pure-tone audiometry at baseline and follow-up. Analysis of students' self-regulated learning in terms of gender using blended learning-based laboratory inquiry teaching materials

The Covid-19 pandemic has affected the way students learn from face-to-face to online learning. This condition is a challenge for educators to provide online teaching materials. Teaching materials developed to facilitate students to learn independently. The characteristics of the teaching materials developed give students the freedom to learn and facilitate the development of their abilities and skills. This study aimed to analyze the effectiveness of blended learning-based laboratory inquiry teaching materials in improving students' self-regulated learning in terms of gender. This type of research is a quasi-experimental research. The test subjects in this study were class XI in one of the public high schools in Mataram City, which consisted of two classes, namely the experimental class and the control class, which consisted of 67 students. There are 34 male students and 33 female students. The data collection technique used a self-regulated learning questionnaire instrument. The instrument refers to seven indicators of self-regulated learning. Data analysis was carried out using N-gain and T-test. The results showed that blended learning-based laboratory inquiry teaching materials could increase students' self-regulated learning. Based on the results of the T test, there was a significant difference between the control class and the experimental class. The third self-regulated learning indicator, namely goal setting and planning, got the highest increase compared to other indicators. The second indicator, namely organization and information, obtained the lowest increase compared to other indicators. Self-regulated learning of male students is higher than female students. With the availability of online class media in the blended learning learning model that can be accessed anytime and anywhere by students, it can increase the readiness of students themselves before face-to-face learning in class takes place. Background: A dual-function phantom designed to quantify the apparent diffusion coefficient (ADC) in different fat contents (FCs) and glass bead densities (GBDs) to simulate the human tissues has not been documented yet. We propose a dual-function phantom to quantify the FC and to measure the ADC at different FCs and different GBDs. Methods: A fat-containing diffusion phantom comprised by 30 glass-bead-containing fat-water emulsions consisting of six different FCs (0, 10, 20, 30, 40, and 50%) multiplied by five different GBDs (0, 0.1, 0.25, 0.5, and 1.0 g/50 mL). The FC and ADC were measured by the “iterative decomposition of water and fat with echo asymmetry and least squares estimation-IQ,” IDEAL-IQ, and single-shot echo-planar diffusion-weighted imaging, SS-EP-DWI, sequences, respectively. Linear regression analysis was used to evaluate the relationship among the fat fraction (FF) measured by IDEAL-IQ, GBD, and ADC. Results: The ADC was significantly, negatively, and linearly associated with the FF (the linear slope ranged from -0.005 to -0.017, R² = 0.925 to 0.986, all p < 0.001). The slope of the linear relationship between the ADC and the FF, however, varied among different GBDs (the higher the GBD, the lower the slope). ADCs among emulsions across different GBDs and FFs were overlapped. Emulsions with low GBDs plus high FFs shared a same lower ADC range with those with median or high GBDs plus median or lower FFs. Conclusions: A novel dual-function phantom simulating the human tissues allowed to quantify the influence of FC and GBD on ADC. Relevance statement: The study developed an innovative dual-function MRI phantom to explore the impact of FC on ADC variation that can affect clinical results. The results revealed the superimposed effect on FF and GBD density on ADC measurements. The objectives of this research are to analyze the difference of learning result between the students with the guided inquiry learning model with virtual learning media and those with the guided inquiry learning model with real learning media viewed from their cooperation skills and the interaction of effect between the virtual and real learning media and the cooperation skills on the students’ learning result. The subjects of the research were the students in Grade VIII of the school. The subjects were determined randomly. The data of the research were collected through test of learning result and questionnaire of cooperation skills and analyzed by using the descriptive quantitative technique of analysis. The results of the research are as follows. There was a difference of learning result viewed from the cooperation skills. The students using the guided inquiry model with virtual learning media had a better learning result than those using the guided inquiry model with real learning media. In addition, there was not any interaction of effect between the guided inquiry learning model with virtual and real learning media and the cooperation skills.

Effect of exogenous insulin injection on hematological status in rats (Rattus norvegicus L.)

Long-term exogenous insulin injection can cause hematopoietic disorders that can affect hematological status. This study aimed to examine the effect of exogenous insulin injection on hematological status in rats (Rattus norvegicus L.). This study uses a completely randomized design with 3 groups and not similar replications. The groups were P0 : control, P1 and P2 : rats were injected with exogenous insulin at a dose of 2 IU/Kg body weight for 14 days and 28 days subcutaneously in the abdomen. Hematological status was observed at the end of treatment, namely erythrocytes, hemoglobin concentration, hematocrit value, erythrocyte index, total and differential leukocytes, number of platelets. Data were analyzed using one-way ANOVA and LSD post hoc test. The results show that insulin injection at dose 2 IU/Kg did not have a significant effect on erythrocytes, hemoglobin concentration, hematocrit value, mean corpuscular volume, mean corpuscular hemoglobin concentration, number of leukocytes, number of neutrophils, number of lymphocytes, and number of platelets (p > 0.05), but had a significant effect on mean corpuscular hemoglobin and number of monocytes (p < 0.05). It can be concluded that exogenous insulin injection for 14 days can cause disturbances in hematological status in the form of a decrease in mean corpuscular hemoglobin and an increase in the number of monocytes.

Development of antibacterial chitosan film modified by various Dioscorea sp. starches enriched with garlic essential oil: Optimal formulation by response surface methodology

Food packaging based on chitosan film modified with starches and enriched with garlic essential oil has been successfully developed optimally. Three types of starch from Dioscorea sp. i.e., Dioscorea alata, Dioscorea esculenta and Dioscorea hispida were used in this study, and their optimum formulations were successfully performed, respectively. Box-behnken simulation by response surface methodology, designed a study with three independent variables; chitosan (1-2% (w/v)), starches (1-2% (w/v)) and garlic essential oil (0.1-0.5% (v/v)), with the dependent variable were inhibitor zone of Salmonella typhi and Staphylococcus aureus. Both parameters have significant effect (p<0.05) on antibacterial activity of the films. An in-depth analysis of the effect of different types of starch on the optimal formulation was observed. Herein, optimal formulations are proposed for smart packaging fabrication in the food industry.

The role of vitamin D3 supplementation to transforming growth factor β1 and tumor necrosis factor α levels in steroid resistant nephrotic syndrome

Nephrotic syndrome, the most common kidney disease in children, has been associated with transforming growth factor β1 (TGF-β1) and tumor necrosis factor a (TNF-α). This study aimed to assess the role of vitamin D3 supplementation to TGF-β1 and TNF-α levels in steroid-resistant nephrotic syndrome (SNRS). This research was an experimental study involving 32 subjects divided into two groups. Each group was given a placebo and vitamin D3 for four weeks. Peripheral blood was taken before and after treatment. In the placebo group, there were significant decrease in 25(OH)D, increase in TGF-β1, and increase in TNF-α after treatment (p < 0.05). In the vitamin D3 supplementation group, there were significant increase in 25(OH)D, decrease in TGF-β1, and decrease in TNF-α (p < 0.05). The increase in 25(OH)D had correlation with the decrease in TGF-β1 (r = -0.593; p = 0.000) and the decrease in TNF-α (r = -0.782; p = 0.000). 0ur data indicated that vitamin D3 supplementation could increase 25(OH)D, decrease TGF- β1 and TNF-α. The decrease in TGF-β1, and TNF-α was negatively correlated with the increase in 25(OH)D in SRNS.

Preliminary analysis and the need for physics learning in SMAN 1 Koto Baru

This article describes a preliminary study on students’ characteristics and their needs in physics learning at SMAN 1 Koto Baru. This study was conducted to evaluate the characteristics of the students and to analyze the need of teacher responses assessment in physics learning activities. The sort of research to be used is research and development. This research uses descriptive statistical methods. Items developed in the form of E-Book based on Project Based Learning (PJBL) models sponsored by the science Laboratory Kit, Physics content class X. The model of growth uses the Plomp model. Researchers used interview cards, instructor questionnaires and student questionnaires to collect initial data. Initial data collection was carried out at SMAN 1 Koto Baru. Based on the findings of the survey observations, the results of the analysis on students’ characteristics shows the percentage of attitudes at 66.67%, initial ability at 62.50%, learning styles at 70%, learning interest at 58.3%, and the motivation at a percentage of 68.75%. Analysis of the needs reflected in the performance analysis at 82.14%, the standard graduation analysis is based on the analysis of attitudes at 80%. The acquisition of knowledge at 54.16% and the skill at 46.43%. E-Book can encourage students to participate actively in the materials discussed. The science Laboratory Kit will help teachers understand. Such work will produce true, realistic and efficient products. Therefore, the learning activities becomes meaningful, and can improve students' competence.
