本文旨在概述miR-122对肝内外组织脂质代谢的调控作用及具体分子机制。MiR-122为肝脏特异性高表达miRNA,与肝内外组织脂质代谢调控及体内血脂水平密切相关。拮抗miR-122可抑制肝内甾醇反应元件结合蛋白-1(sterol regulatory element binding protein-1,SREBP-1)介导脂质合成并同时促进脂质β氧化和脂肪分解,防止肝脏脂质过度累积及非酒精性脂肪肝病进展。肝源性miR-122亦可经血液循环影响外周脂质代谢。拮抗miR-122可抑制脂肪细胞成脂肥大、改善血脂水平保护心肌细胞及促进骨骼肌纤维甘油三酯合成与储存,抑制体内肥胖症、动脉粥样硬化性心血管疾病及糖尿病的发展。总之,抑制miR-122表达及生物学作用可改善肝内外组织脂质代谢过程,防止脂代谢紊乱疾病的发生发展。
