Colorectal cancer (CRC) is a leading cause of mortality worldwide. We conducted a genome-wide association study meta-analysis of 100,204 CRC cases and 154,587 controls of European and east Asian ancestry, identifying 205 independent risk associations, of which 50 were unreported. We performed integrative genomic, transcriptomic and methylomic analyses across large bowel mucosa and other tissues. Transcriptome- and methylome-wide association studies revealed an additional 53 risk associations. We identified 155 high-confidence effector genes functionally linked to CRC risk, many of which had no previously established role in CRC. These have multiple different functions and specifically indicate that variation in normal colorectal homeostasis, proliferation, cell adhesion, migration, immunity and microbial interactions determines CRC risk. Crosstissue analyses indicated that over a third of effector genes most probably act outside the colonic mucosa. Our findings provide insights into colorectal oncogenesis and highlight potential targets across tissues for new CRC treatment and chemoprevention strategies.

Kilonovae are a rare class of astrophysical transients powered by the radioactive decay of nuclei heavier than iron, synthesized in the merger of two compact objects 1–4. Over the first few days, the kilonova evolution is dominated by a large number of radioactive isotopes contributing to the heating rate2, 5. On timescales of weeks to months, its behavior is predicted to differ depending on the ejecta composition and merger remnant6–9. However, late-time observations of known kilonovae are either missing or limited10, 11. Here we report observations of a luminous red transient with a quasi-thermal spectrum, following an unusual gamma-ray burst of long duration. We classify this thermal emission as a kilonova and track its evolution up to two months after the burst. At these late times, the recession of the photospheric radius and the rapidly-decaying bolometric luminosity (Lbol ∝ t−2.7±0.4) support the recombination of lanthanide-rich ejecta as they cool. We develop a concept for a waveguide that exploits spatial control of nonlinear surface-polaritonic waves. Our scheme includes an optical cavity with four-level N-type atoms in a lossless dielectric placed above a negative-index metamaterial layer. We propose exciting a polaritonic Akhmediev breather at a certain position of the interface between the atomic medium and the metamaterial by modifying laser-field intensities and detunings. Furthermore, we propose generating position-dependent polaritonic frequency combs by engineering widths of the electromagnetically induced transparency window commensurate with the surface-polaritonic modulation instability. Therefore, this waveguide acts as a high-speed polaritonic modulator and position-dependent frequency-comb generator, which can be applied to compact photonic chips. Copyright © 2019, The Authors. The ability of an eavesdropper (Eve) to perform an intercept-resend attack on a free-space quantum-key-distribution (QKD) receiver by precisely controlling the incidence angle of an attack laser has been previously demonstrated. However, such an attack could be ineffective in the presence of atmospheric turbulence due to beam wander and spatial mode aberrations induced by the air's varying index of refraction. We experimentally investigate the impact turbulence has on Eve's attack on a free-space polarization-encoding QKD receiver by emulating atmospheric turbulence with a spatial light modulator. Our results identify how well Eve would need to compensate for turbulence to perform a successful attack by either reducing her distance to the receiver or using beam wavefront correction via adaptive optics. Furthermore, we use an entanglement-breaking scheme to find a theoretical limit on the turbulence strength that hinders Eve's attack.

Fundamental bounds on the operation of Fano nonlinear isolators

Nonlinear isolators have attracted significant attention for their ability to break reciprocity and provide isolation without the need of an external bias. A popular approach for the design of such devices is based on Fano resonators, which, due to their sharp frequency response, can lead to very large isolation for moderate input intensities. Here, we show that, independent of their specific implementation, these devices are subject to fundamental bounds on the transmission coefficient in the forward direction versus their quality factor, input power, and nonreciprocal intensity range. Our analysis quantifies a general tradeoff between forward transmission and these metrics, stemming directly from time-reversal symmetry, and that unitary transmission is only possible for vanishing nonreciprocity. Our results also shed light on the operation of resonant nonlinear isolators, reveal their fundamental limitations, and provide indications on how it is possible to design nonlinear isolators with optimal performance.
