Cancer remains a significant global health challenge, with the Cell Division Cycle 7 (CDC7) protein emerging as a potential therapeutic target due to its critical role in tumor proliferation, survival, and resistance. However, a comprehensive analysis of CDC7 across multiple cancers is lacking, and existing therapeutic options have come with limited clinical success. The aim of this is to integrate a comprehensive pan-cancer analysis of CDC7 with the identification of novel marine-derived inhibitors, bridging the understanding of CDC7's role as a prognostic biomarker and therapeutic target across diverse cancer types. In this study, we conducted a pan-cancer analysis of CDC7 across 33 tumor types using publicly available datasets to evaluate its expression, genetic alterations, immune interactions, survival, and prognostic significance. Additionally, a marine-derived compound library of 31,492 molecules was screened to identify potential CDC7 inhibitors using chemoinformatics and machine learning. The top candidates underwent rigorous evaluations, including molecular docking, pharmacokinetics, toxicity, Density Functional Theory (DFT) calculations, and Molecular Dynamics (MD) simulations. The findings revealed that CDC7 is overexpressed in several cancers and is associated with poor survival outcomes and unfavorable prognosis. Enrichment analysis linked CDC7 to critical DNA replication pathways, while its role in modulating tumor-immune interactions highlighted its potential as a target for immunotherapy. Among all tested compounds, Tetrahydroaltersolanol D (CMNPD21999) exhibited the strongest binding affinity and stability, along with better drug-likeness and zero toxicity. These attributes highlight its potential as a promising drug candidate for CDC7 inhibition and future cancer treatment development. Furthermore, additional in vitro and in vivo studies are required to confirm the effectiveness of this drug candidate against the CDC7 protein. © 2025

Navigating crowds: Reconsidering visitor recreational experience and crowding preference for a forest park in Taiwan

As forest recreational activities gain popularity, the potential for crowding and activity-based conflicts to diminish sustainable recreational quality becomes a critical concern. Consequently, it is vital to understand the factors shaping recreational preferences and conflict perceptions in natural settings. This study selected Xitou Nature Education Area, Taiwan's most frequented forest park, as a case study to investigate complex relationships among visitor demographics, crowding perceptions, and recreational behaviors. Using a questionnaire and choice experiment, the study explored perceptions of crowding and recreational conflicts within Xitou, alongside visitors' willingness to travel further to avoid crowded conditions. The analysis showed that visitor preferences differed significantly across socioeconomic groups, with age playing a substantial role in shaping motivations and behaviors. While overall perceptions of crowding were moderate, visitors aged 65 and older demonstrated notable tolerance for crowds. Despite the park's high visitation, the incidence of recreational conflict was relatively low, with primary disturbances attributed to visitors resting in various locations, park staff, and group game participants. Contrary to expectations, visitors did not exhibit strong willingness to pay to avoid crowding. Instead, results surprisingly suggested a slight preference for increased social interaction within the park, challenging the general assumption that crowding is always viewed negatively. Drawing from visitor perceptions of overcrowding, we propose enhanced regulations and stricter monitoring for designated resting and picnicking areas to elevate the forest recreation experience. Crucially, this study highlights that effective park management must acknowledge and cater to diverse visitor motivations, whether they seek physical activity, social engagement, or solitude. © 2025

The utilization of graphitic carbon nitride (C3N4) to enhance the oxidative polymerization of hydroquinone (HQ) within birnessite (δ-MnO2) systems was investigated. C3N4, a Lewis base enriched with nitrogen, acted as a bridge through π−π interactions between metal oxides and polyphenols. These interactions increased the affinity of the reactants, resulting in an enhanced reaction rate of six-fold, accelerated browning, and improved complex stability of C3N4...HQ...MnO2 complex. As a result, a humic-like substance (HS) was rapidly formed with improved surface morphology and larger particle sizes. Electron transfer of HQ...MnO2 promoted by C3N4 played a crucial role in HQ oxidation, expanding atomic distance and composite morphology. Theoretical calculations supported the experimental results, indicating strong affinity and higher binding energies in the C3N4...HQ...MnO2 complex. The complex had a more stable structure with a narrower HOMO-LUMO gap, facilitating HS formation. 13C and 1H NMR spectra confirmed the experimental-theoretical agreement. © 2023, The Authors. We aimed at the ecotoxicity of CNFs (at 10 and 100 mg/kg) in Lumbricus terrestris earthworms naturally infected with Monocystis sp. After 28-d of exposure, treatments did not affect the survival rate. However, we observed a significant loss of body biomass, and Monocystis sp. infection in seminal vesicles was potentiated by exposure to CNFs. Earthworms exposed to CNFs showed a redox imbalance in the seminal vesicle, muscle, and intestine and an alteration in nitric oxide production in these organs. In muscles, we also noticed a significant reduction in AChE activity in earthworms exposed to CNFs. The histopathological analyses revealed the treatments' significant effect on the structures of the different evaluated tissues. When taken together and after applying IBRv2 and PCA, we noticed that the response of earthworms to CNFs at 100 mg/kg showed a more significant deviation from the unexposed group. Thus, we demonstrate that CNFs increase the parasite load of Monocystis sp. of adult L. terrestris earthworms and induce biochemical changes and histopathological changes, especially at 100 mg/kg. Our results point to the additional impact these nanomaterials can have on the health of earthworms, signaling the need for greater attention to their disposal and ecotoxicological effects on soil organisms. © 2023, The Authors. Thus, we aimed at the ecotoxicity of CNFs (at 10 and 100 mg/kg) in Lumbricus terrestris earthworms naturally infected with Monocystis sp. After 28 days of exposure, treatments did not affect the survival rate. However, we observed a significant loss of body biomass, and Monocystis sp. infection in seminal vesicles was potentiated by exposure to CNFs. Although we did not notice a concentration-response for several of the biomarkers, when taken together and after the application of IBRv2 and principal component analysis (PCA), we noticed that the response of earthworms to CNFs at 100 mg/kg showed a more significant deviation from the unexposed group. This was mainly determined by inhibiting antioxidant activity in the seminal vesicle, biochemical biomarkers assessed in muscle and intestine, and histomorphometric muscle biomarkers from earthworms exposed to CNFs at 100 mg/kg. Thus, we demonstrate that CNFs increase the parasite load of Monocystis sp. of adult L. terrestris earthworms and induce biochemical changes and histopathological changes, especially at 100 mg/kg. Our results point to the additional impact these nanomaterials can have on the health of earthworms, signaling the need for greater attention to their disposal and ecotoxicological effects on soil organisms. © 2023, The Authors. This study aims to investigate the distribution and ecological risk assessment of microplastics (MPs) in peatland areas located in Long An province, Vietnam's Mekong Delta. In general, polyvinyl chloride (60.7%), polyethylene (25.8%), and polypropylene (11.9%) were the most abundant polymers determined in the thirty sediment samples. The hazard index (HI) remarked a level of III for MPs contamination in Tan Thanh and Thanh Hoa districts. The pollution load index (PLI) and potential ecological risk index (RI) indicated that the contamination risk of MPs polymer types in the studied sites is relatively high. According to PLI values, MPs levels of peatlands in Tan Thanh and Thanh Hoa are high and moderate, respectively, while the peatland sediments in Duc Hue district are less contaminated. Furthermore, ecological risk indexes in the peatland areas were relatively high, with PLIoverall (level III);HIoverall (level V), and RIoverall (extreme danger). Hence, this study proposed a SWOT framework for challenges of MPs pollution in order to manage peatlands appropriately and minimize ecological risks. As a result, several practical strategies and appropriate approaches have been recommended to reduce microplastics towards a circular economy. These findings provide the initial quantitative assessment insights into hazard levels and ecological impacts of MPs in Vietnam's Mekong Delta peatlands. © 2022 Elsevier Inc.

Both the proportion and expenditure of dining out and takeaway in Taiwan have been increasing year by year. This study investigates the factors influencing the catering service providers (CSPs) procurement of domestically produced or imported food materials. The empirical results showed that the procurement cost, quality, safety, and availability of ingredients were the major considerations for CSPs to purchase ingredients. Different types of CSPs had different cognitions, attitudes, and purchasing behaviors toward domestically produced and imported ingredients. Bento establishments, self-service cafeterias, and noodle shops adopted higher proportions of domestically produced ingredients (DPIs), and they also had more confidence in these ingredients. On the contrary, table service restaurants, all-you-can-eat restaurants, breakfast eateries, coffee shops, and fast food restaurants adopted higher proportions of imported ingredients. The results may be related to the food items purchased and catering services provided by different CSPs. Therefore, CSPs should be included in the scope and promotion of food and agriculture education so as to increase their emphasis on ingredients for health, increase their tendency to purchase DPIs, promote the disclosure of information on food producers and ingredients, and enhance the communications among food producers, buyers, CSPs, and dining-out consumers.

Fermentation of seagrape seaweed ( Caulerpa lentillifera ) with Lactiplantibacillus plantarum and supplemented with coffee cherry pulp enhances microbial viability, bioactive compound content, and antioxidant activity while reducing cytotoxicity and improving sensory acceptance. The co-fermented formulation containing 50 g/L seagrape, 25 g/L coffee cherry pulp, L. plantarum TISTR 2070, and 30 g/L sucrose exhibited the highest total phenolic content and antioxidant capacity, with DPPH, FRAP, and ABTS values significantly exceeding those of the unfermented control ( p ≤ 0.05). Cold storage at 4°C demonstrated that the fermented beverage retained its antioxidant properties and maintained microbial viability with minor changes in sugar content and pH. Additionally, cytotoxicity testing on human colon cells demonstrated that fermentation markedly reduced cytotoxicity, with the CC 50 increasing from 16,020 ± 445.6 µg/mL (unfermented SG) to 25,860 ± 336.6 µg/mL in the co-fermented sample ( p ≤ 0.05), indicating improved safety for human consumption. Sensory evaluation confirmed that the fermented beverage was more accepted than the unfermented counterpart, with improvements in taste, mouthfeel, and overall preference. Multivariate analysis by PCA further revealed strong correlations between microbial activity, antioxidant capacity, and sensory attributes. LC-QTOF-MS analysis confirmed the release and biotransformation of diverse bioactive metabolites during fermentation, contributing to the enhanced functional properties of the final product. With its enhanced bioactivity, stability, reduced cytotoxicity, and superior sensory attributes, co-fermentation of seagrape and coffee cherry pulp presents a promising strategy for the development of health-promoting functional beverages.

Nature reserve optimization with buffer zones and wildlife corridors for rare species

The creation of protected areas is an important public policy strategy for protecting species, as mentioned in the Aichi targets. This paper formulates spatial integer programming (set covering) models to protect both rare species (arbitrarily defined here as species breeding in 1% or less of the territory) and common species. Spatial constraints are used to form buffer zones around core areas that protect rare species, and cost-efficient corridors linking these buffered cores are then designed. The models are applied on a portion of the State of Oregon and results are evaluated in light of the expected United Nations post-2020 targets for area-based conservation measures. It is estimated that such results aimed at covering all species and enhancing protection of at-risk species seem locally consistent with the global biodiversity objectives for 2030.
