Multiple exosome RNA analysis methods for lung cancer diagnosis through integrated on-chip microfluidic system

Exosomes are now raising focus as a prospective biomarker for cancer diagnostics and prognosis owing to its unique bio-origin and *** take part in cellular communication and receptor mediation and transfer their cargos(e.g.,proteins,m RNA and DNA).Quantitative analysis of tumor-related nucleic acid mutations can be a potential method to cancer diagnosis and prognosis in early *** we present an integrated microfluidic system for exosome on-chip isolation and lung cancer RNA analysis through droplet digital PCR(dd PCR).Gradient dilution experiments show great linearity over a large concentration range with R^(2)=*** the system,four cell lines and two mutation targets were parallelly detected for mutation *** experiments demonstrated mutation heterogeneity and the results were agree with cell *** results proved our integrated microfluidic system as a promising means for early cancer diagnosis and prognosis in the era of liquid biopsy.

For accurately identifying the distribution charac-teristic of Gaussian-like noises in unmanned aerial vehicle(UAV)state estimation,this paper proposes a non-parametric scheme based on curve similarity *** the framework of the pro-posed scheme,a Parzen window(kernel density estimation,KDE)method on sliding window technology is applied for roughly esti-mating the sample probability density,a precise data probability density function(PDF)model is constructed with the least square method on K-fold cross validation,and the testing result based on evaluation method is obtained based on some data characteristic analyses of curve shape,abruptness and *** com-parison simulations with classical methods and UAV flight exper-iment shows that the proposed scheme has higher recognition accuracy than classical methods for some kinds of Gaussian-like data,which provides better reference for the design of Kalman filter(KF)in complex water environment.

Development of high-efficiency electrode materials for the electrochemical CO_(2)reduction reaction(CO_(2)RR)with high current density and selectivity compatible with industry is an important but significant ***,we describe a facile strategy to enhance the selectivity and current density by regulating the local electron density of the cobalt site in a series of stable,conjugated,bimetallic Co/Zn polyphthalocyanine frameworks Co_(x)Zn_(y)PPc with an AB stacking model under alkaline aqueous *** adjusting the ratio of Co and Zn to 3:1,the optimal Co3Zn1PPc exhibits an industry-compatible CO partial current density of 212 mA cm^(−2)at−0.9 V versus reversible hydrogen electrode in a flow cell,which is 1.7 and 9.1 times that of the single metal polyphthalocyanine CoPPc and ZnPPc,***_(3)Zn_(1)PPc shows a high CO Faraday efficiency of more than 90%in a wide operating potential window of−0.3 to−0.9 ***-depth experimental and theoretical analysis revealed that introduction of electron-rich Zn atoms modified the electron density of the active Co center,placing Co in the electronrich region and weakening the bonding strength with the reaction intermediate,thereby improving the CO_(2)RR *** results clarify the interaction mechanism of dual metal sites at the atomic level and provide a new avenue for the design of electrocatalysts with potential in industrial applications.

For improving the classification accuracy of bottlenose dolphin communication signals(whistle)under captive conditions when overlapping echolocation signals(click),a fusion classification method based on machine learning is *** time-frequency distribution features of whistle signals are extracted to train the random forest classifier,and the Mel timefrequency diagram features are used to train the convolution neural network *** this basis,a fusion decision-maker is designed to classify and recognize the aliased whistle *** classification and recognition results of the experimental data collected from the sound signals of captive dolphins showed that the fusion classification method has better classification *** classification accuracy of the aliased whistle signals is more than 94%,which is better than the time-frequency distribution feature classifier and Mel time-frequency graph feature classifier,and can improve the classification ability of the aliased signals.

Information Freshness-Oriented Trajectory Planning and Resource Allocation for UAV-Assisted Vehicular Networks

In this paper,multi-UAV trajectory planning and resource allocation are jointly investigated to improve the information freshness for vehicular networks,where the vehicles collect time-critical traffic information by on-board sensors and upload to the UAVs through their allocated spectrum *** adopt the expected sum age of information(ESAoI)to measure the network-wide information *** is jointly affected by both the UAVs trajectory and the resource allocation,which are coupled with each other and make the analysis of ESAoI *** tackle this challenge,we introduce a joint trajectory planning and resource allocation procedure,where the UAVs firstly fly to their destinations and then hover to allocate resource blocks(RBs)during a *** on this procedure,we formulate a trajectory planning and resource allocation problem for ESAoI *** solve the mixed integer nonlinear programming(MINLP)problem with hybrid decision variables,we propose a TD3 trajectory planning and Round-robin resource allocation(TTPRRA).Specifically,we exploit the exploration and learning ability of the twin delayed deep deterministic policy gradient algorithm(TD3)for UAVs trajectory planning,and utilize Round Robin rule for the optimal resource *** TTP-RRA,the UAVs obtain their flight velocities by sensing the locations and the age of information(AoI)of the vehicles,then allocate the RBs to the vehicles in a descending order of AoI until the remaining RBs are not sufficient to support another successful *** results demonstrate that TTP-RRA outperforms the baseline approaches in terms of ESAoI and average AoI(AAoI).

DNA methylation is one of the earliest known modification pathways, and it regulates gene expression, which in turn influences many biological processes. Direct, label-free detection of DNA methylation with high sensitivity remains a great challenge. Surface-enhanced Raman scattering (SERS), a non-invasive and label-free vibration spectroscopy technique, offers sensitive intrinsic chemical information that makes it an attractive option for DNA analysis. In this study, we employed iodide-modified silver nanoparticles to generate highly consistent SERS signals of DNA at micromolar concentrations in aqueous solutions. This enabled the acquisition of single-base sensitive DNA fingerprint details pertaining to base methylations (such as 5-methylcytosine). As a proof of concept, the SER spectra of a DNA and its methylated counterpart were compared and analyzed, resulting in an obvious identification of DNA methylation. In particular, we first designed two DNA sequences, the sole change being that one of the nucleotides, cytosine, is replaced with a 5-methylcytosine. Further SERS experimental study revealed that methylating a single cytosine in the DNA strand caused a subtle but evident alteration in the SERS spectrum. A new Raman peak emerged at 760 cm-1, the Raman peak at 792 cm-1 moved to 790 cm-1, and there was a noticeable drop in the peak intensity ratio between 1572 cm-1 and 1634 cm-1. This approach may provide a novel and easy-to-use tool for the label-free identification of single-molecule DNA mutations or modifications, furthering the advancement of ultra-sensitive genomic research in the future. © 2024 SPIE. Consistency analysis of GRACE and GRACE-FO data in the study of global mean sea level change

Global mean sea level(GMSL) change is one of the important indicators of global climate change and is a crucial scientific issue of continuing interest. As satellite altimeter data, the Gravity Recovery and Climate Experiment(GRACE) and Argo continue to be updated, especially with the release of GRACE Follow-On(GRACE-FO) data, making it necessary to combine these latest data to estimate sea level change. Determinations on whether the GRACE and GRACE-FO observation systems provide unbiased global observation data have not been effectively evaluated. Therefore, this research mainly investigated the consistency of GRACE and GRACE-FO observation data in studying GMSL change. By comparing the sum of the GMSL calculated by the two gravity satellites and Argo data with the GMSL calculated by satellite altimeters, the discrepancy between GRACE-FO + Argo and satellite altimeter data is about 7.9 ± 2.3 mm, which is likely derived from the inconsistency between GRACE and GRACE-FO data.
