Abstract: Agastache rugosa contains phenolic compounds and flavonoids, and has been extensively used as a traditional herbal medicine. The major components in Agastache rugosa are rosmarinic acid, tilianin, and acacetin, for which several analytical techniques have been reported. However, these substances have yet to be simultaneously quantified in human plasma. In this study, we aimed to simultaneously determine the three active components of Agastacherugosa in human plasma by developing a reliable quantitative analytical method using ultra-high-performance liquid chromatography–tandem mass spectrometry (UHPLC-MS/MS). Chromatographic separation of the plasma samples was achieved using an ACQUITY UPLC® BEH C18 column with a gradient mobile phase of water and acetonitrile containing 0.1% formic acid. Mass spectrometric detection was performed using a triple quadrupole tandem mass spectrometer in negative electrospray ionization (ESI−) and multiple reaction monitoring (MRM) modes. The developed quantitative method was validated for the three active components. All three analytes exhibited a linear response over the ranges of 0.5–50 ng/mL for rosmarinic acid, 0.1–20 ng/mL for acacetin, and 0.5–20 ng/mL for tilianin with a weighting factor of 1/x (where x is the concentration). At three quality control (QC) concentration levels (low, medium, and high), including the lower limit of quantitation (LLOQ), acceptable accuracy (±15%) was achieved in the intra- and interday validations. The concentration of rosmarinic acid was highest in plasma. Tilianin and acacetin appeared and were eliminated earlier in the plasma than rosmarinic acid. This study provides a successfully validated method that can be used in further clinical applications of Agastache rugosa extracts. © 2023, The Authors. All rights reserved.

The advancement of alloys and steel in engineering highlights the critical need for robust methods to ensure structural integrity. While traditional Non-Destructive Testing and Evaluation (NDT/E) techniques provide valuable insights, they often fail to detect pre-existing stress conditions that contribute to fatigue cracks. Recent approaches have addressed this limitation by analyzing stress and strain at the grain level using advanced measurement systems—often based on diffraction techniques—integrated with Finite Element Method (FEM) and Digital Image Correlation (DIC) platforms. However, the application of these techniques for in-service inspections remains limited by challenges in accessibility, cost, and scalability. To overcome these issues, an integrated 2D surface system combining optical microscopy with DIC and grain-level analysis within the FEM framework is proposed. This method enables efficient visualization of grain-level deformation and stress distribution, thereby providing deeper insights into the microstructural behavior of polycrystalline materials under external loads. The proposed methodology is theoretically outlined and validated through simulations and experimental analyses, demonstrating its potential as a practical solution for in-service inspections. Ultimately, this approach is expected to advance inspection techniques by offering an accessible and scalable tool for monitoring microstructural behavior and identifying potential failure mechanisms through surface analysis.

The study determined the effect of incorporating Momordica charantia leaf powder (MCLP) into corn-starch 3D food-printing ink as a functional ingredient. The effects of the particle size (75, 131, and 200 μm) and quantity of MCLP on 3D printing performance, structural, textural, and rheological properties of corn starch gel were evaluated with different concentrations (5, 10, and 15% (w/w)) of corn starch. The viscoelastic properties of food inks were determined considering their behavior during extrusion and self-recovery after printing. Scanning electron microscope was used to characterize the microstructure. Based on the results, a high starch content (15%) with 5% MCLP was more favorable for 3D food printing. In addition, 3D printing performance, textural and rheological properties of formulated ink was mainly governed by the particle size of MCLP. The food ink with a 5% mass fraction of 200 μm MCLP had the highest printing precision and the best masticatory properties. © 2023, The Authors. All rights reserved.

Over hundreds of years, cats have been domesticated and selectively bred, resulting in numerous pedigreed breeds expedited by recent cat shows and breeding associations. Concerns have been raised about the limited breeding options and the genetic implications of inbreeding, indicating challenges in maintaining genetic diversity and accurate identification in purebred cats. In this study, genetic variability and structure were examined in 5 Thai domestic cat breeds using 15 microsatellite markers and mitochondrial DNA (mtDNA) D-loop sequencing. In total, 184 samples representing the Wichien Maat (WCM), Suphalak (SL), Khao-Manee (KM), Korat (KR), and Konja (KJ) breeds were analyzed. High genetic diversity (H_o and H_e > 0.5) was observed in all breeds, and mtDNA analysis revealed two primary haplogroups (A and B) that were shared among all domestic cat breeds in Thailand and globally. However, minor differences were observed between Thai domestic cat breeds based on clustering analyses, in which a distinct genetic structure was observed in the WCM breed. This suggests that allele fixation for distinctive morphological traits has occurred in Thai domestic cat breeds that emerged in isolated regions with shared racial origins. Analysis of relationships among individuals within the breed revealed high identification efficiency in Thai domestic cat breeds (P_(ID)sibs < 10⁻⁴). Additionally, diverse and effective individual identification can be ensured by optimizing marker efficiency by using only nine loci. This comprehensive genetic characterization provides valuable insights into conservation strategies and breeding practices for Thai domestic cat breeds. © The Author(s) 2024.

Mung bean seeds decreased their viability very fast during storage, especially in tropical area without proper packaging. The purpose of this study is to observethe decrease in viability of mung bean seeds during storage and to build a mathematical model for shelf life prediction. Dried mung bean seeds were vacuum packed and stored in Accelerated Shelf-life Testing chamber at five different temperatures of 30° C, 35° C, 40° C, 45° C, 50° C with approximately 80% in relative humidity, to facilitate rapid deterioration with different rate of physical and chemical change in the seed. The results show that there was an increase in moisture content and the germination decreased in different rates. Changes in values for some parametersof the seeds affected the viabilty of the seeds during rapid deterioration. The value of constant (k)for viability decreasing was calculated and used to build the mathematical model for predicting the shelf life of mung bean seeds during storage at any temperature. Using the developed model, it was observed that the shelf life of the Sriti variety of mung bean seeds in vacuum packaging could maintain 90% of germination level for 421 days or 14.3 months if stored at 20 °C, and for 173.37 days or 5.78 months if stored at 25 °C. © Published under licence by IOP Publishing Ltd.

Hydroxyapatite(HA)is a representative substance that induces bone *** research team extracted nanohydroxyapatite(EH)from natural resources,especially equine bones,and developed it as a molecular biological ***(PEI)was used to coat the EH to develop a gene *** verify that PEI is well coated in the EH,we first observed the morphology and dispersity of PEI-coated EH(pEH)by electron *** pEH particles were well distributed,while only the EH particles were not distributed and ***,the existence of nitrogen elements of PEI on the surface of the pEH was confirmed by EDS,calcium concentration measurement and fourier transform infrared spectroscopy(FT-IR).Additionally,the pEH was confirmed to have a more positive charge than the 25 kD PEI by comparing the zeta *** a result of pGL3 transfection,pEH was better able to transport genes to cells than 25 kD *** verification as a gene carrier for pEH,we induced osteogenic differentiation of DPSCs by loading the BMP-2 gene in pEH(BMP-2/pEH)and delivering it to the *** a result,it was confirmed that osteogenic differentiation was promoted by showing that the expression of osteopontin(OPN),osteocalcin(OCN),and runt-related transcription factor 2(RUNX2)was significantly increased in the group treated with BMP-2/*** conclusion,we have not only developed a novel nonviral gene carrier that is better performing and less toxic than 25 kD PEI by modifying natural HA(the agricultural byproduct)but also proved that bone differentiation can be effectively promoted by delivering BMP-2 with pEH to stem cells.

Bacillus species are Gram-positive bacteria that are rod-shaped, endospore-forming, and aerobic or facultatively anaerobic. With over 300 recognized species, Bacillus subtilis stands out as a well-studied model organism. The genus's various species exhibit a wide range of physiological capabilities, allowing them to thrive in diverse environmental conditions. Each cell produces a single endospore, which is highly resistant to heat, cold, radiation, desiccation, and disinfectants. Among Bacillus strains, those capable of producing spores, biofilms, and carotenoids demonstrate significant resilience to UV light. This review examines the genes involved in spore formation, biofilm development, and carotenoid synthesis, emphasizing their roles in UV radiation survival. We explore the interconnections between these processes and their combined contribution to UV resistance, focusing on the underlying genetic mechanisms. These insights will benefit researchers studying the genetic basis of UV radiation resistance in Bacillus species. IMPORTANCE: Bacteria employ adaptive strategies in extreme environments through rapid changes in gene expression, altering their phenotype for survival. Bacillus species, for example, defend against UV radiation by making spores, creating biofilms, and producing pigments. During sporulation, sigma factors (σ, σ, σ, and σ) regulate gene expression to adapt to environmental shifts. It has been found that the spores of some species may contain pigments that strongly absorb UV radiation, playing a crucial role in spore UV resistance. UV light penetrates biofilm matrices minimally, mainly affecting surface cells, which produce compounds like mycosporine-like amino acids and carotenoids to shield against UV damage.

This paper describes a differential CMOS bandpass amplifier dedicated for ECG acquisition. The proposed amplifier is constituted by a current-feedback instrumentation amplifier made from only 4 transistors in conjunction with capacitive-resistive feedback network. The amplifier circuit was designed using 0.35 micron CMOS and verified by post-layout simulation. It shows that the proposed amplifier provides 40 dB and 0 dB mid-band and dc amplifications, respectively. The amplifier bandwidth acquires 0.1 to 279 Hz. The input referred noise and input impedance of 2 μV rms and 295 MΩ (at 50 Hz) are obtained. Monte-Carlo mismatch simulation confirms the amplifier's common-mode rejection ratio of 77 dB. Operated from a 2 V dc supply, the ECG amplifier consumes 672 nW.
