Single-cell electrophoretic assays offer proteoform detection specificity, but target multiplexing can be further enhanced by transitioning from fluorescence to mass spectrometry imaging for readout. We report for the first time multiplexed ion beam imaging (MIBI) of isoelectrically separated proteoforms from single cells (scIEF). MIBI of scIEF enables the detection of proteoforms differing by a single charge unit. Additionally, we demonstrate that MIBI's ability to do multiple depth image acquisition-comparable to z-stack confocal imaging-enables us to disregard surface artefacts that would normally confound quantitation. © 2020 CBMS-0001

Optical attenuators extend dynamic range but alter angular response of planar ultraviolet-C dosimeters

A challenge for sensors used in ultraviolet-C (UV-C) decontamination protocols of N95 respirators is validation that the entire N95 surface receives the minimum acceptable dose. Photochromic indicators (PCIs) can accurately measure UV-C dose on nonplanar surfaces, but often saturate below doses required to decontaminate porous, multilayered textiles such as N95s. Here, we investigate the use of optical attenuators to extend PCI dynamic range while maintaining a near-ideal angular response - critical for accurate measurements when UV-C is uncollimated. Through an analytical model, we show that tuning attenuator refractive index, attenuation coefficient, and thickness can extend dynamic range, but compromises ideal angular response unless the attenuator is an ideal diffuser. To demonstrate this tradeoff empirically, we pair PCIs with model specular (floated borosilicate) and diffuse (polytetrafluoroethylene) attenuators, characterize the angular response, and evaluate on-N95 UV-C dose measurement accuracy of each PCI-attenuator stack in a UV-C decontamination system. While both borosilicate and polytetrafluoroethylene increase PCI dynamic range >4×, both attenuators introduce angle-dependent transmittance, which causes location-dependent underestimation of UV-C dose. The PCI-borosilicate and PCI-polytetrafluoroethylene stacks underreport true on-N95 dose by 1) 14.7% and 3.6%, respectively, on a surface near-normal to the array of source lamps, and 2) 40.8% and 19.8%, respectively, on a steeply sloped location. Overall, we demonstrate that while planar optical attenuators can increase PCI dynamic range, verification of near-ideal angular response is critical for accurate UVC dose measurement. © 2021, CC BY-NC-SA.

Histone acetylation is an important epigenetic modification that governs gene expression, chromatin changes in stress response, and cell fate transition. FRET biosensors have been developed for various epigenetic events to enable spatiotemporal tracking of sub-cellular signaling events. Previously reported histone H3 acetylation biosensor recognizing two acetyl residues lacked specificity. In this study, using a single bromodomain of the BRD4, we have developed a genetically encoded H3K9ac biosensor. We systematically investigated different combinations of the BET family protein as binding domains and performed site-saturated mutagenesis to optimize the biosensor, achieving a dynamic FRET change up to 30% under TSA treatment. With the application of the optimized H3K9ac biosensor, we revealed different basal active chromatin architectures in invasive tumor cells compared to benign tumor cells. Furthermore, we found that H3K9ac level increased dramatically when cancer cells passed through microchannels, which models the physical constraints and mechanical microenvironmental conditions that cancer cells encounter when passing through narrow spaces within the body. This result highlights the chromatin plasticity in response to external mechanical stresses. In summary, our H3K9ac biosensor provides a versatile tool for mechanistic investigation of cell fate transition in cancer and mechanotransduction.

The isothermal amplification coupled with CRISPR detection has attracted tremendous interest for the sensitive, specific, and qualitative detection of nucleic acids. We report here a Microfluidics Enabled Digital Isothermal CRISPR-cas13a Assay (MEDICA) which leverages the droplet microfluidics technology to enable recombinase polymerase amplification (RPA) and Cas13a trans-cleavage reporting in individual droplet partitions for absolute quantification. The optimal one-pot MEDICA achieved single nucleic acid molecule detection within 30 min. We applied MEDICA to quantify HPV 16 and 18 viral loads in clinical samples, indicating complete accordance with quantitative polymerase chain reaction (qPCR) results. © 2021 MicroTAS 2021 - 25th International Conference on Miniaturized Systems for Chemistry and Life Sciences. Biomechanics plays an integral role in cancer development and progression. Accurately quantifying the mechanical properties of cells would enable a more comprehensive understanding of cancer, complementing conventional biochemical and genetic analyses. Here, we introduce a microfluidic platform for measuring single cell viscoelasticity at multiple frequencies in a high-throughput and label-free manner. Using the Coulter principal and a unique microfluidic channel geometry, our device records resistance pulses in order to estimate a cell's viscoelastic phenotype. MRI can provide localized assessment of lung function for monitoring people with lung disease. Hyperpolarized 129Xe MRI directly images pulmonary gas distribution but requires specialized hardware. Conventional 1H MRI acquisitions can also provide functional maps using free-breathing approaches. The purpose of this study is to evaluate regional ventilation derived from 3D ultrashort echo-time (UTE) 1H MRI using Motion-Compensated Low-Rank constrained reconstruction (MoCoLoR), by comparing against 129Xe MRI and pulmonary function testing as reference-standard. The study is retrospective in design. The study included 57 participants (25.4 ± 15.8 years, 35 males and 22 females): 12 healthy volunteers, 20 pediatric, and 25 adult people with cystic fibrosis (CF) scanned between January 2022 and February 2023. Field strength/sequence: 3T;129Xe: 2D multislice spoiled gradient-recalled sequence;UTE 1H: variable-density 3D radial sequence. K-means-based 129Xe ventilation defect percent (VDP), forced expiratory volume in 1 s (FEV1), and lung clearance index (LCI) were evaluated against UTE 1H VDP from a modified k-means method. The correspondence of ventilation defect maps from 129Xe and UTE 1H was also evaluated. Statistical tests included the Pearson correlation coefficient (r) and t tests, with p 129Xe and UTE 1H VDP were significantly correlated (r = 0.64, p = (Formula presented.)). Bland–Altman analysis showed a bias of −0.05 (p = (Formula presented.)) and limits of agreement of (0.07, −0.17). The Dice spatial accuracy of the UTE-based ventilation defect regions using 129Xe as reference was 0.64 ± 0.05. Hydrogen sulfide (H2S), an antioxidant and cell signaling molecule, plays a critical role in redox reactions in the biological system, and its imbalances in blood levels have been implicated in various diseases. Here, we demonstrate a novel chemosensor-based "turn-on" electrochemiluminescence (ECL) system for the selective and sensitive detection of H2S in a type 1 diabetes rat model. Our luminogenic chemosensor (Probe) is a redox-stable cyclometalating iridium (III) complex with emission-quenching 2,4-dinitrophenyl ether (DNP) that shows a significant enhancement in emission intensity upon cleavage of DNP by selective reaction towards H2S. To achieve robust and reliable ECL emission of Probe for biological analysis, we developed a facile deproteinization protocol based on organic solvent-induced protein precipitation/syringe filtration that eliminates uncontrolled protein adsorption on the electrode surface from protein-rich biological samples. Our deproteinization-assisted ECL chemosensor system showed effectiveness in detecting disease-associated H2S levels in biological media with a linear response in the range of 0-250 µM (R2: 0.992), the limit of detection of 7.2 µM, and average signal-to-noise ratio of ~25. It enables reliable in vitro monitoring of H2S level changes in blood samples of a type 1 diabetes rat model. This strategy to integrate chemosensor-based ECL and facile deproteinization protocol provides a sensitive and rapid disease-monitoring platform for detecting small-molecule biomarkers that cannot be detected by conventional ECL sandwich immunoassays. © 2023, The Authors.
