Colorectal cancer (CRC) ranks among the most prevalent tumors, particularly affecting men and women, with mouse models and imaging technologies like colonoscopy, endoluminal (eUBM), and transabdominal (tUBM) ultrasound biomicroscopy playing pivotal roles in exploring new treatment avenues. The evaluation of colon wall thickness (Wt), a significant marker of tumor presence, is feasible through eUBM or tUBM, contrasting with colonoscopy's limitations. This study scrutinizes the consistency of Wt measurements obtained via eUBM and tUBM in mice, with or without colon tumors induced by azoxymethane (AOM) and dextran sulfate sodium salt (DSS). UBM images were captured using both eUBM and tUBM instruments, with subsequent Wt measurements performed using ImageJ software. Comparison between Wt obtained from eUBM and tUBM images used the One-Way ANOVA, for normal distributed data, or the Kruskal-Wallis test otherwise. Local Ethics Committee for Animal Use in Research approved the work (protocol 014/20). In general, the results indicate a substantial median Wt disparity between control and tumor groups measured by both techniques (p<0.05). Notably, discrepancies in Wt findings between the two methods were observed in the control group (p<0.05), despite the excellent resolution of the images in delineating colon layers.

Gas vesicles (GVs), gas-filled protein-encapsulated nanostructures, synthesized by microorganisms such as Halobacterium salinarum NRC-1 (HALO) and Anabaena flos aquae (ANA) have been reported as potential ultrasound contrast agents (UCA) due to their ability to mechanically vibrate with the same frequency, or harmonics, as the one from the incident ultrasound (US) wave. Despite GVs resonating in frequencies above 21 MHz, they collapse irreversibly when the US incident field pressure exceeds the vesicle collapse pressure. There are few reports on the relationship between the pressure of the acoustic wave incident in the GVs and the corresponding intensity of the backscattered wave signal from the GVs and, therefore, this study was designed to investigate how varying the relative incident US power, at 21MHz, reflects on the intensity of the US backscattered signal and the collapse of HALO and ANA GVs. The mean (±SD) of the grey level intensity for the ROIs, in maximum intensity pixel images, with GVs and PBS, were determined for transmitted acoustic relative power of 1, 5, 10, 20%. ANA-GVs presented a lower signal intensity, but greater resistance to the increase in relative US pressure. In conclusion, the best behavior was for the Halo-GVs at 5% power and at 21MHz frequency.

Purpose: Foot and ankle pathologies are among the most prevalent conditions within the human locomotor system. Imaging examinations are crucial for diagnosing, treating, and achieving satisfactory functional outcomes for these conditions. However, conventional imaging exams often fail to capture all the necessary details for precise analysis, necessitating the use of multiple imaging modalities to evaluate bones, ligaments, cartilage, and other structures. Objective: This research aims to explore computational technologies and algorithms for 3D reconstruction and fusion of foot and ankle images, alongside the analysis, segmentation, classification, and evaluation techniques applied to these images. Methods: The study investigates different computational approaches for 3D reconstruction and multimodal image fusion, integrating X-ray and MRI data. Advanced segmentation and classification techniques are applied to enhance the precision of structural assessment. Additionally, we assess the availability and quality of databases for training and validating these models. Results: Our findings highlight a significant gap in research concerning these specific applications, as well as a shortage of suitable databases for 3D reconstruction and fusion of images from X-rays and MRI. Conclusion: The study emphasizes the need for dedicated datasets and further research to develop effective computational solutions for multimodal foot and ankle imaging. © The Author(s), under exclusive licence to The Brazilian Society of biomedical engineering 2025.

This article proposes the creation, implementation and development of an integration plugin for ImageJ that makes calls in MATLAB functions for the segmentation UBM imagens, more specific endoluminal (eUBM) e transabdominal (tUBM), in the laboratory the images are generating with frequencies between 30-40 MHZ. These images allow view colon layer, identify tumors in colons, to calculate tumor volume and 3D visualization. This integration may to improve the time taken to perform the procedure and the user experience to reduce the number of tasks and change windows, making the experience more fluid and with less risk of errors.

Gas vesicles (GVs), nanostructure gas-filled protein-shelled compartments, act as ultrasound contrast agents (UCAs). Recent works report bioengineered GvpC, one of the shell proteins, potentializes interactions with specific molecules in tumor microenvironment. An important target of investigation is Cathepsin B (CTSB), an enzyme produced by several tumors. CTSB acts in the tumoral microenvironment to increase tumor growth and invasion. This work objective relied on bioengineering the GVs to promote the GvpC-CTSB interaction with a subsequent increase of ultrasound (US) signal backscattered. GvpC was engineered to contain a known CTSB cleavage sequence re-added in striped GV (ΔGV). Western Blot (WB) test was used to compare the GVwt (wild type) and GV∗ (GV engendered) before and after 24 hours reaction with CTSB. US images for PBS (negative control), GVtw, ΔGV, GV∗ and CTSB+GV∗, were obtained in 21MHz (5% total power) and regions-of-interest (ROIs) over the final B-mode image were determined. The WB test for GVwt resulted in mean (±SD) GvpC concentrations of 25169 (±8574) and 32873 (±9113) and GV∗ was 32487 (±6867) and 12763 (±2846), at t0 and t24, respectively. The US image gray level intensities for the ROIs related to the samples of PBS, GVwt, ΔGV, GV∗ and CTSB+GV∗ were 55.8 (±17.6), 47.4 (±16.6), 46.1 (±22.8) and 150.2 (±24), respectively. These results reveal an increase in the US signal backscattered from GV∗ after CTSV reaction. This increase is likely related to a direct GvpC degradation by CTSB, without affecting the GV∗ structure. In conclusion, high frequency US B-mode images showed the GV∗ with the potential to act as an UCA sensitive to the CTSB presence in tumoral microenvironment. © 2023 IEEE.

Purpose: Ultrasound contrast agent (UCA), used to enhance the contrast in ultrasonography, consists of microbubbles filled with an inert gas and encapsulated with a protein or a lipidic-stabilize membrane. The UCAs are able to resonate when exposed to ultrasound waves and to generate an echo-acoustic signal with increased intensity. Recently, a technique to produce UCA from microorganisms was developed and it consists of lysing microorganism cells to obtain gas vesicle (GV) after cultivation in the laboratory. GVs occur almost exclusively in aquatic microorganisms, as Microcystis aeruginosa (MYC), and provide buoyancy which permit to float towards the water surface. This work implemented a technique to produce GVs from MYC, at the Ultrasound Laboratory, and inspected their properties of acting as UCAs, analyzed through ultrasonic images acquired at 40 MHz, and their structural characteristics such as morphology, diameter, and length. Methods: After the cultivation of MYC, the buoyant cells with GVs were separated from the culture medium and submitted to cell lysis with chemical treatment. Thereafter, the solution with lysed cells was centrifuged, at 300g during 8 h at 4–8 °C, to isolate the GVs (absorbance 0.6, λ = 500 nm). Results: Transmission electron microscopy inspection revealed the MYC-GVs with elliptical morphology and having a diameter between 100 and 150 nm and length near to 200 nm. B-mode ultrasound images of the solutions proved the GV’s ability to generate an echo signal, thus confirming their performance as UCAs. © 2020, Sociedade Brasileira de Engenharia Biomedica.

Colon cancer has a high incidence, without considering skin cancer, being the third most common cancer in the world. Despite this high incidence and high mortality rate, most of the outcomes could be prevented by using accurate techniques for early cancer detection to enable early treatment. In this context, it is important to conduct preclinical studies, using animal models of colon diseases, to understand the development of the disease and to test new diagnostic approaches including imaging instrumentation. We have previously used a 3-French ultrasound mini-probe to image the layered structure in the mouse colon, to detect polyps, and to visualize their invasion through the colon wall. The present work aimed to measure the colon wall thickness and perform a numeric differentiation between the thickness of normal and inflamed mouse colon, a crucial step to elucidate the disease progression in colorectal cancer mouse model.

During coagulation, blood changes from liquid to gel, and the clot viscoelastic properties continue modifying until fibrinolysis. Ultrasound (US) was used in this work to trace the time-changing shear modulus, μ, during plasma clotting, based on the vibration signature induced by an impulsive acoustic radiation force (iARF) on a sphere embedded in a plasma sample along its clotting process induced based on the aPTT (activated partial thromboplastin time) assay. An US push system transmitted bursts of US pulses (2.03 MHz, 1000 cycles and 1.25 Hz of pulse repetition frequency) to generate an iARF on the glass sphere (4.0 mm in diameter and density of 2500 kg/m3), which displacement was monitored by a 5.0 MHz US pulse-echo probing system. The relative delays between the rf-echo signals from the sphere yielded the signal representing the sphere damped oscillation that was fitted with a theoretical-model displacement function whose unknowns are the iARF amplitude and the medium μ and shear viscosity, η. The method was repeated five times and the time-changing mean μ was determined along 2100 s of the clotting process. During the first 15 s μ remained at 20 Pa and then varied rapidly until 60 s, at a rate of 1.9 Pa/s. Between 60 and 800 s, μ varied at a rate of 0.2 Pa/s. From 800 to 2100 s μ remained stable at about 280 Pa. The interval of fast changing μ corresponds to the fibrin formation period and comprehends the clotting time of 25s typically obtained with the aPTT assay for control plasma. © 2017 IEEE.

Objectives: The 12-item Self-Compassion Scale–Short Form (SCS–SF) is a widely used instrument for the assessment of self-compassion. To date, there have been few examinations of this instrument’s psychometric properties, particularly across nations and languages. Therefore, we used data from the Body Image in Nature Survey (BINS) to assess measurement invariance of the SCS–SF across nations, languages, gender identities, and age groups. Methods: Participants (N = 56,968) from 65 nations completed the SCS–SF in 40 languages. Using these data, we tested various hypothesised models of the SCS–SF in the total sample and, using multi-group confirmatory factor analysis, tested for invariance of the optimal model across national groups, languages, gender identities, and age groups. Results: In the total dataset, we found that an 11-item, 2-factor model (i.e., SCS-11) provided best fit to the data, with the two factors tapping distinct constructs of compassionate and uncompassionate self-responding. The SCS-11 was found to be partially scalar invariant across national groups and languages, and fully scalar invariant across gender identities and age groups. There was wide variation in latent means for the two factors, particularly across national groups and languages. Further analyses showed negligible associations between the two factors and sociodemographic variables, including marital status, financial security, and urbanicity. Conclusions: Our results suggest that it may be possible to derive a stable 2-factor model of the SCS–SF for use in cross-cultural research, but also highlight the likelihood of cross-national and cross-linguistic variations in the way that self-compassion is understood. © The Author(s) 2025.
