Two new 16-hydroxybriaranes, 11-dehydroxybriavioid B (1) and 12-epi-briastecholide L (2), have been isolated from an algal-containing octocoral identified as Briareum stechei. The structures of metabolites 1 and 2 were corroborated by extensive 2D-NMR studies and comparison with literature data. Briarane 2 was found to reduce the release of inducible nitric oxide synthase (iNOS) to 87.92 %, at a concentration of 10 μM. © 2023 Phytochemical Society of Europe

Phytotherapeutic potential of Campomanesia xanthocarpa (Mart.) O. Berg: antitumor effects in vitro and in silico, with emphasis on SK-MEL-28 melanoma cells-a study on leaf and fruit infusions

The study investigated the efficacy of Campomanesia xanthocarpa infusions on human melanoma cells (SK-MEL-28). The phytochemical profile revealed 18 phenolic compounds in the leaf infusion and 9 in the fruit infusion. After 24 h of treatment, the infusions demonstrated antineoplastic effects, reducing cell viability at all tested concentrations for the leaf infusion. For the fruit infusion, a significant reduction in cell viability was observed specifically at the 800 μg/mL concentration. Fluorescence microscopy and mitochondrial membrane potential results indicated that the leaf infusion was more effective in reducing cell viability and mitochondrial function in SK-MEL-28 cells, possibly due to its greater variety of phenolic compounds compared to the fruit infusion. The leaf infusion also induced higher production of intracellular reactive oxygen species compared to the fruit infusion. Protein sulfhydryl levels were reduced for the leaf infusion. Epigallocatechin gallate, Isoquercitrin, Rutin, Kaempferol-3-O-rutinoside, Chlorogenic acid, and Ellagic acid were identified as the main compounds with activity against SK-MEL-28 cells. Molecular docking analysis underscored factors such as affinity, cavity size, binding mode, and contact residues with specific compounds chosen for their favorable properties in targeting BRAF, CDK4, CDK6, MEK1, and MEK2. The variability in binding affinities may directly influence the compounds' ability to inhibit different signaling pathways related to cancer cell growth and proliferation. The results suggest that phenolic compounds from C. xanthocarpa extracts have therapeutic potential and could contribute to melanoma therapies.

chemical investigations on the ethyl acetate extract of the octocoral Capnella imbricata collected from the waters of Taiwan, which led to the isolation of three capnellenes (1–3), including one new metabolite, 9(12)-capnellene-8β,10α,15-triol (1), along with two known metabolites, 9(12)-capnellene-5α,8β,10α-triol (2), and 9(12)-capnellene-8β,10α-diol (3). The structures of the isolated metabolites were elucidated through extensive spectroscopic analyses, while the relative stereochemistry of 3 was confirmed by a single-crystal X-ray diffraction analysis for the first time in this study. Compounds 1–3 were evaluated in terms of their anti-inflammatory activity by examining their inhibitory effects on the generation of superoxide anions and the release of elastase by human neutrophils. © 2022 Elsevier B.V.

We evaluated the concentration of copper (Cu), zinc (Zn), mercury (Hg), nickel (Ni), chromium (Cr), vanadium (V), and cobalt (Co) in the mangrove tissues (leaf and root) and adjusted sediments, as well as seawater samples in the north coast of the Persian Gulf from three counties (Mahshahr, Bord Khun, Asaluyeh). Assaluyeh and Mahshahr were selected because of their proximity to the oil and petrochemical industries and Bord Khun was considered for being away from pollution sources. The metal detected in highest concentration in mangrove leaves and roots was Cu (Asaluyeh: 1.507 μg/g and 1.198 μg/g, Bord Khun: 1.812 and 1.756 μg/g, and Mahshahr: 0.880 and 1.617 μg/g, for leaves and roots, respectively). For sediments, Cr (among the metals) had the highest concentration (1.325 μg/g) in Asaluyeh, whereas in Bord Khun and Mahshahr Ni was predominant: 2.542 and 2.237 μg/g, respectively. For seawater, Cu was the most abundant metal and Hg was the one with the lowest concentration. The bio-concentration factor for Zn and Cu revealed that mangroves in the study areas have suitable potential for phytoremediation of these two metals. Translocation factors from the roots to leaves also indicated higher transportability for Zn and Cu through the plant tissues, compared to the other metals. Ecological indexes quantified the contamination of sediments and revealed a low level of heavy metal accumulation in sediments of the north of the Persian Gulf. Human risk hazards appeared low in all the areas under study. Gas and petrochemical activities can be considered as a potential source of the discharge of heavy metals into the aquatic environment. © 2021 Elsevier B.V.

Dendritic cell (DC) maturation and antigen presentation are key factors for successful vaccine-based cancer immunotherapy. This study developed manganese-based layered double hydroxide (Mn-LDH) nanoparticles as a self-adjuvanted vaccine carrier that not only promoted DC maturation through synergistically depleting endogenous glutathione (GSH) and activating STING signaling pathway, but also facilitated the delivery of model antigen ovalbumin (OVA) into lymph nodes and subsequent antigen presentation in DCs. Significant therapeutic-prophylactic efficacy of the OVA-loaded Mn-LDH (OVA/Mn-LDH) nanovaccine was determined by the tumor growth inhibition in the mice bearing B16-OVA tumor. Our results showed that the OVA/Mn-LDH nanoparticles could be a potent delivery system for cancer vaccine development without the need of adjuvant. Therefore, the combination of GSH exhaustion and STING pathway activation might be an advisable approach for promoting DC maturation and antigen presentation, finally improving cancer vaccine efficacy.

Helicobacter pylori ( H. pylori ) infection is a critical initiating factor in numerous gastrointestinal diseases. Here, we present yogurt-inspired hybrid membrane vesicles (hMVs) engineered by fusing bacterial outer membrane vesicles (OMVs) with modified milk fat globule membrane vesicles (MMVs) to prevent and treat H. pylori infection. The hMVs function as binding site competitors, reducing bacterial adhesion while regulating epithelial integrity and immune responses. This dual mechanism synergistically inhibits H. pylori infection and promotes gastric epithelial repair. In vitro , hMVs demonstrated remarkable efficacy in accelerating scratch-wound healing, enhancing lipid raft formation, increasing tight junction protein ZO-1 expression, and promoting macrophage phagocytosis of apoptotic epithelial cells. In mouse models for both H. pylori prevention and treatment, oral administration of hMVs effectively reduced H. pylori loads by 86% and 66%, respectively. Additionally, this treatment mitigated inflammatory factor secretion, decreased neutrophil recruitment, and preserved the abundance of beneficial probiotics in the stomach.

Application of deep learning to predict the low serum albumin in new hemodialysis patients

BACKGROUND:Serum albumin level is a crucial nutritional indicator for patients on dialysis. Approximately one-third of patients on hemodialysis (HD) have protein malnutrition. Therefore, the serum albumin level of patients on HD is strongly correlated with mortality. METHODS:In study, the data sets were obtained from the longitudinal electronic health records of the largest HD center in Taiwan from July 2011 to December 2015, included 1,567 new patients on HD who met the inclusion criteria. Multivariate logistic regression was performed to evaluate the association of clinical factors with low serum albumin, and the grasshopper optimization algorithm (GOA) was used for feature selection. The quantile g-computation method was used to calculate the weight ratio of each factor. Machine learning and deep learning (DL) methods were used to predict the low serum albumin. The area under the curve (AUC) and accuracy were calculated to determine the model performance. RESULTS:Age, gender, hypertension, hemoglobin, iron, ferritin, sodium, potassium, calcium, creatinine, alkaline phosphatase, and triglyceride levels were significantly associated with low serum albumin. The AUC and accuracy of the GOA quantile g-computation weight model combined with the Bi-LSTM method were 98% and 95%, respectively. CONCLUSION:The GOA method was able to rapidly identify the optimal combination of factors associated with serum albumin in patients on HD, and the quantile g-computation with DL methods could determine the most effective GOA quantile g-computation weight prediction model. The serum albumin status of patients on HD can be predicted by the proposed model and accordingly provide patients with better a prognostic care and treatment.
