We assessed the characteristics of receptor response for the purpose of developing a vibration stimulator and stimulation methods aiming to improve standing balance. Vibration detection thresholds and the degree to which the site of stimulation matches the site of perception were examined in 22 feet of 11 healthy adults (21.0±1.8 years). Stimulation was applied at a frequency of 225Hz using an original vibration stimulator comprising a frequency oscillator and piezoelectric element. Threshold was determined by averaging measurements obtained by ascending and descending methods. In the ascending method, the applied voltage was measured at the point at which vibration could be perceived, and in the descending method, the applied voltage was measured at the point at which vibration could no longer be detected. The concordance between the site of stimulation and the site of perception was determined by asking subjects to indicate the site at which vibration was perceived using the ascending method. Vibration thresholds were 6.6±3.6V at the base of the fifth metatarsal bone, 5.5±3.3V at the navicular bone, 7.8±3.8V at the lateral malleolus, 6.5±3.1V at the medial malleolus, and 11.9±4.5V at the head of the fibula. Hepatic stellate cells (HSCs) play a crucial role in the development of liver fibrosis which is characterized by massive tissue scarring, elevated hepatic sinusoidal pressure, and portal hypertension. However, while a multitude of chemical factors have been investigated in the pathogenesis of liver fibrosis, physical factors such as elevated hydrostatic pressure and shear stress caused by blood flow in sinusoids remain unclear. In this study, we developed a three-dimensional (3D) cell culture microfluidic platform that mimics the physical environments of hepatic sinusoids to investigate the effects of elevated hydrostatic pressure on HSCs phenotypes.

Background Epidermal growth factor receptor (EGFR) is upregulated in prostate cancer (PCa). However, suppression of EGFR did not improve the patient outcome, possibly due to the activation of PI3K/Akt signaling in PCa. Compounds able to suppress both PI3K/Akt and EGFR signaling may be effective for treating advanced PCa. Purpose We examined if caffeic acid phenethyl ester (CAPE) simultaneously suppresses the EGFR and Akt signaling, migration and tumor growth in PCa cells. Methods Wound healing assay, transwell migration assay and xenograft mice model were used to determine the effects of CAPE on migration and proliferation of PCa cells. Western blot, immunoprecipitation, and immunohistochemistry staining were performed to determine the effects of CAPE on EGFR and Akt signaling. Results CAPE treatment decreased the gene expression of HRAS, RAF1, AKT2, GSK3A , and EGF and the protein expression of phospho-EGFR (Y845, Y1069, Y1148, Y1173), phospho-FAK, Akt, and ERK1/2 in PCa cells. CAPE treatment inhibited the EGF-induced migration of PCa cells. Combined treatment of CAPE with EGFR inhibitor gefitinib showed additive inhibition on migration and proliferation of PCa cells. Injection of CAPE (15 mg/kg/3 days) for 14 days suppressed the tumor growth of prostate xenografts in nude mice as well as suppressed the levels of Ki67, phospho-EGFR Y845, MMP-9, phospho-Akt S473, phospho-Akt T308, Ras, and Raf-1 in prostate xenografts. Conclusions Our study suggested that CAPE can simultaneously suppress the EGFR and Akt signaling in PCa cells and is a potential therapeutic agent for advanced PCa.

During coagulation, blood changes from liquid to gel, and the clot viscoelastic properties continue modifying until fibrinolysis. Ultrasound (US) was used in this work to trace the time-changing shear modulus, μ, during plasma clotting, based on the vibration signature induced by an impulsive acoustic radiation force (iARF) on a sphere embedded in a plasma sample along its clotting process induced based on the aPTT (activated partial thromboplastin time) assay. An US push system transmitted bursts of US pulses (2.03 MHz, 1000 cycles and 1.25 Hz of pulse repetition frequency) to generate an iARF on the glass sphere (4.0 mm in diameter and density of 2500 kg/m3), which displacement was monitored by a 5.0 MHz US pulse-echo probing system. The relative delays between the rf-echo signals from the sphere yielded the signal representing the sphere damped oscillation that was fitted with a theoretical-model displacement function whose unknowns are the iARF amplitude and the medium μ and shear viscosity, η. The method was repeated five times and the time-changing mean μ was determined along 2100 s of the clotting process. During the first 15 s μ remained at 20 Pa and then varied rapidly until 60 s, at a rate of 1.9 Pa/s. Between 60 and 800 s, μ varied at a rate of 0.2 Pa/s. From 800 to 2100 s μ remained stable at about 280 Pa. The interval of fast changing μ corresponds to the fibrin formation period and comprehends the clotting time of 25s typically obtained with the aPTT assay for control plasma. © 2017 IEEE.

Acoustic angiography is a contrast imaging approach in which the superharmonics associated with low frequency excitation of microbubbles are received and processed over a broad bandwidth at a much higher frequency. As demonstrated by Dayton and colleagues, this technique facilitates the visualization of the microvasculature with a resolution typical of micro computed tomography and with superior contrast-to tissue-ratios than conventional nonlinear processing. To date dual frequency contrast imaging has been performed with a 2-4MHz annulus coaligned with 25-30MHz single element focused transducer operated by a Vevo770 (Visualsonics) scanner. This technology has limitations (mechanical scanning, single focus, limited focal depth) which hinder its clinical use but which could be alleviated using dual-frequency arrays (DFA). In this presentation we first describe the performance and applications of mechanical acoustic angiography and early results from the development of dual frequency array devices for acoustic angiography. The logical extension to array based designs is then explored. We built prototype dual frequency devices in both a vertical and horizonal topology. The horizonal topography consisted of a central 256 element high frequency array (HF, 20 MHz) flanked by two single element low frequency transducers(LF, 2 MHz) that extended for the length of the HF array. The vertical topography consisted of a 32-element LF transmitter array positioned behind a 256-element HF receiver array. Both arrays were built from piezoelectric composites. The HF backing material, between the two piezoelectric layers in the vertical design is weakly and highly attenuating at 2 MHz and 20 MHz, respectively. Two matching layers on the LF array and 4 matching layers on the HF array were used to improve transmission throughout the whole array structure. A tomographic clinical cardiac SPECT system consisting of many static CZT-based pinhole gamma cameras is evaluated for dedicated, pendant 3D molecular breast imaging. The reconstructed spatial resolution and linearity of the spherically symmetric FOV were evaluated with a multi-point source array. A series of anthropomorphic phantom imaging measurements using 0.4 and 0.9 mm diameter lesions was performed;lesion-to-background ratios ranged from [13:1] to [3:1] to simulate various biological uptake;three breasts used were 470-1730 mL in volume. Basic contrast, CNR and coefficient of variation were measured with the anthropomorphic torso+organ background. Decreasing acquisition times from 300-30 sec were a surrogate for lower injected doses. All data was reconstructed using MLEM with various iterations on the commercial workstation. Results indicate that the system largely behaves in the expected way: better overall resolution, linearity, sensitivity, and uniformity closer to the detectors but within the FOV. Other key observations include: (1) higher lesion: background ratios yielded higher image contrast and CNR;(2) lesion contrast and CNR appeared independent of acquisition time;(3) breast size had less of an effect on lesion metrics, but yielded decreased out-of-field (e.g. cardiac and hepatic) contributions with larger breasts. These results indicate that lower injected doses could be utilized for 3D MBI. While additional optimization would be beneficial for breast imaging, this high sensitivity SPECT system could be used for dynamic 3D MBI for lesion diagnostics or therapeutic monitoring.

Work Organization Model Applied on Smallholder Dairy Farms in Brazil

This research identified difficulties faced by dairy farmers in family farming, describing this diagnosis through ergonomic demand items （EDIs）. The objective was to elaborate an organization model of work related to dairy farming （WRDF）. As a methodology, a descriptive exploratory study was carried out in 14 family dairy farms that were visited and had their activities monitored. As main results, 11 EDIs were identified. The ones grouped in the infrastructure factor showed the highest resolution priority. The WRDF model was composed of three categories： management, public policies management and infrastructure, which were further subdivided into technical and social aspects. From this perspective, it is believed that the implementation of the model can bring organizational improvement, once adjustments are made in the procedures and working conditions. This study sheds light on the conditions that need to be resolved with higher priority so that the farmers will remain in the dairy business.
