Work-related musculoskeletal disorders (WMSDs) are often caused by repetitive lifting, making them a significant concern in occupational health. Although wearable assist devices have become the norm for mitigating the risk of back pain, most spinal assist devices still possess a partially rigid structure that impacts the user’s comfort and flexibility. This paper addresses this issue by presenting a smart textile-actuated spine assistance robotic exosuit (SARE), which can conform to the back seamlessly without impeding the user’s movement and is incredibly lightweight. The SARE can assist the human erector spinae to complete any action with virtually infinite degrees of freedom. To detect the strain on the spine and to control the smart textile automatically, a soft knitting sensor which utilizes fluid pressure as sensing element is used. The new device is validated experimentally with human subjects where it reduces peak electromyography (EMG) signals of lumbar erector spinae by 32%±15% in loaded and 22%±8.2% in unloaded conditions. Moreover, the integrated EMG decreased by 24.2%±13.6% under loaded condition and 23.6%±9% under unloaded condition. In summary, the artificial muscle wearable device represents an anatomical solution to reduce the risk of muscle strain, metabolic energy cost and back pain associated with repetitive lifting tasks. © 2024, CC BY-NC-ND.

AP-2α gene deregulation is associated with renal cell carcinoma patient survival

Background: Renal cell carcinoma (RCC), one of the most fatal urologic tumors, accounts for approximately 3% of all adult cancers and exhibits a high metastatic index at diagnosis and a high rate of relapse. Radical or partial nephrectomy is a curative option for nonmetastatic RCCs. Targeted therapy has been shown to improve the survival of patients with metastatic RCCs. However, the underlying cellular and molecular events associated with RCC pathogenesis are not well known. Methods: To investigate the clinical role of the transcription factor activator protein (AP)-2α in RCC, methylated CpG island recovery assays and microarray analysis were employed. COBRA and RT‒qPCR assays were performed to assess AP-2α expression in RCC. Results: A negative correlation was noted between AP-2α mRNA expression levels and methylation status. Multivariate analyses showed that AP-2α mRNA was a major risk factor not only for overall and disease-free survival in RCC but also for disease-free survival in clear cell RCC. Conclusions: Our results indicated that AP-2α expression was deregulated in RCC and associated with overall patient survival and disease-free survival. Such findings suggest that AP-2α might play an important role in the pathogenesis of RCC. © The Author(s) 2024.

Investigating neural activity is critical to advancing neuroscience and informing clinically-relevant neuromodulation, but it is often burdened by the presence of recording artifacts resulting from electrical stimulation. We developed and evaluated a generalizable method for removing the electrical artifacts elicited by various neurostimulation waveforms. This investigation of a novel variety of stimulation waveforms was facilitated by obtaining and labelling the neural activity of 15 loose cell-attached patch clamp recordings of retinal ganglion cells. These labelled recordings provided 5785 peristimulus spikes for which the pipeline exhibited a true positive rate of 88.7%, false discovery rate of 2.9%, and d-prime of 3.11, far superseding interpolation (of which bore a d-prime of 0.32). We additionally utilized synthesized spike trains to demonstrate recovery of low-amplitude stimulus-embedded spikes with negligible waveform distortion (low root mean square error). This technique could better enable a rich variety of neurostimulation investigations, and aid in the development of clinical neurostimulation strategies.

We propose an in vitro intestinal tube-shaped device with a secreted mucin layer that enables us to achieve stable and continuous co-culturing with bacteria. Our tube-shaped device is made of collagen hydrogel and has 3D crypt-like structures created by electrolytically generated microbubbles. Bacterial suspension can be perfused via glass/silicone tubes attached at both ends of the tube device. Supply of culture medium diffused from the outside of the tube ensures a stable culture of Caco-2 and HT29 cells on the inside of the tube. We confirmed that HT29 cells mainly secreted a mucin layer in the apical portion of cryptic structures that spatially separates the intestinal cells and bacteria as seen in vivo environments. As a result, the co-culture of intestinal cells and lactobacillus in vitro was demonstrated in the 3D crypt-like structures in our intestinal tube-shaped device.

Quantitative evaluation of adhesion severity around subscapularis and its relationship with shoulder range of motion in frozen shoulder and rotator cuff disorder: an observational study using dynamic ultrasonography

Background: This study aimed to evaluate the severity of adhesion between muscles in the shoulder joint using dynamic ultrasonography and to confirm whether adhesions cause range of motion (ROM) restrictions. Methods: Twenty-four shoulders from 15 frozen shoulder patients and 24 shoulders from 18 rotator cuff disorder patients were enrolled. We obtained ultrasound video sequences of the subscapularis (SSC) and deltoid muscles during shoulder external rotation. The mean stretching velocities of the deltoid and SSC were subsequently analyzed using a personal computer. If adhesions occurred between both muscles, the deltoid was stretched more vigorously, and we calculated mean stretching velocity of the deltoid / SSC as adhesion severity. The coracohumeral ligament thickness was measured using the same images. Shoulder ROM was measured by using a universal goniometer. Results: The intraclass correlation coefficients (1.1) and (2.1) of the adhesion severity measurements were 0.85 and 0.91, respectively. Multiple linear regression analysis revealed that the adhesion severity is a significant predictor for external rotation ROM in the rotator cuff disorder group (R² = 0.44, F = 10.1, P < .01, t = −2.9), while coracohumeral ligament thickness predicts ROM in the frozen shoulder group (R² = 0.28, F = 5.5, P = .01, t = −3.0). Conclusion: The proposed method is reliable. Muscle adhesion causes ROM restriction of the shoulder joint. The primary cause of shoulder ROM restriction differed between the diagnostic groups. © 2024 The Author(s)

Melanin-Containing Cellulose-Based MIP Film for the Controlled Release of Methotrexate

The aim of this study is to control the release of methotrexate (MTX) from a cellulose-based molecularly imprinted polymer (MIP) film containing melanin using near-infrared (NIR) laser irradiation. Methacrylic acid (MAA) as a functional monomer for the recognition of MTX was grafted to carboxymethyl cellulose through free radical polymerization. Templating molecule MTX was then attached via hydrogen bonding to MAA. Melanin was then added to the MIP film. The MIP film selectively recognized MTX over folic acid (with a similar structure to MTX). After 808 nm NIR laser irradiation at 0.7 W/cm2 for 10 min, the surface temperature of the MIP film rose above 60 °C and the MTX release and permeation rates increased rapidly. After release the MTX from the MIP film, more uniformly sized pores were observed than seen in the non-molecularly imprinted film. These results indicate that the MIP film is suitable for transdermal drug delivery systems. © 2024, The Authors. All rights reserved.

Nickel oxide (NiO_(x)) has significant cost and stability advantages over poly[bis (4-phenyl)(2,4,6-trimethyl phenyl)amine](PTAA) for inverted p-i-n perovskite solar cells (PSCs),but the poor NiO_(x)/perovskite contact stemming from some reactive species at the interface led to suboptimal device *** solve this problem,we take a multiple donor molecule approach,using 3,3’-(4,8-bis(hexylthio)benzo[1,2-b:4,5-b’]dithiophene-2,6-diyl)bis(10-(6-bromohexyl)-10H-phenoxazine)(BDT-POZ) as an example,to modify the NiO_(x)/perovskite *** primary goal was to reduce the under-coordinated Ni^(≥3+) cations via electron transfer from the donor molecules to NiO_(x),thus mitigating the detrimental reactions between perovskite and NiO_(x).Equally importantly,the hole extraction at the interface was greatly enhanced after the organic donor modification,since the hydrophobic species atop NiO_(x) not only enabled pinhole-free crystallization of the perovskite but also properly tuned the interfacial energy level ***,the PSCs with NiO_(x)/BDT-POZ HTL achieved a high power conversion efficiency (PCE) up to 20.16%,which compared excellently with that of the non-modified devices (17.83%).This work provides a new strategy to tackle the exacting issues that have so far impeded the development of NiO_(x) based PSCs.

The prevalence of oral squamous cell carcinoma (OSCC) is increasing worldwide mainly due to poor oral hygiene and unrestricted lifestyle. Advanced-stage OSCC is associated with poor prognosis and a 5-year survival rate of only 30%–50%. The present study was designed to investigate the anticancer effect and mode of action of Glycyrrhiza-derived semilicoisoflavone B (SFB) in 5-fluorourasil (5FU)-resistant human OSCC cell lines. The study findings revealed that SFB significantly reduces OSCC cell viability and colony formation ability by arresting cell cycle at the G2/M and S phases and reducing the expressions of key cell cycle regulators including cyclin A, cyclin B, CDC2, and CDK2. The compound caused a significant induction in the percentage of nuclear condensation and apoptotic cells in OSCC. Regarding pro-apoptotic mode of action, SFB was found to increase Fas-associated death domain and death receptor 5 expressions and reduce decoy receptor 2 expression, indicating involvement of extrinsic pathway. Moreover, SFB was found to increase pro-apoptotic Bim expression and reduce anti-apoptotic Bcl-2 and Bcl-xL expressions, indicating involvement of intrinsic pathway. Moreover, SFB-mediated induction in cleaved caspases 3, 8, and 9 and cleaved poly(ADP-ribose) polymerase confirmed the induction of caspase-mediated apoptotic pathways. Regarding upstream signaling pathway, SFB was found to reduce extracellular signal regulated kinase 1/2 (ERK) phosphorylation to execute its pro-apoptotic activity. The Human Apoptotic Array findings revealed that SFB suppresses claspin expression, which in turn caused reduced phosphorylation of ATR, checkpoint kinase 1 (Chk1), Wee1, and CDC25C, indicating disruption of ATR-Chk1 signaling pathway by SFB. Taken together, these findings indicate that SFB acts as a potent anticancer compound against 5FU-resistant OSCC by modulating mitogen-activated protein kinase (MAPK) and ATR-Chk1 signaling pathways. © 2024 Wiley Periodicals LLC.

Bimetallic sulfide has attracted much attentions as efficient electroactive materials of battery supercapacitor hybrid (BSH), because of numerous redox states, high conductivity and good energy efficiency. Developing composites having excellent chemical and electrochemical properties is necessary to enhance the energy storage features of bimetallic sulfides. In this study, CuCo2S4 and Ni(OH)2 composites (CuCo2S4@Ni(OH)2) are designed on Ni foam via the facial hydrothermal process and chemical bath deposition as the electroactive material of BSH at first time. Effects of Ni(OH)2 amounts on CuCo2S4 on electrochemical and physical properties of CuCo2S4@Ni(OH)2 are investigated. The physical mixed CuCo2S4 and Ni(OH)2 composite is also fabricated to verify synergistic effects of CuCo2S4@Ni(OH)2. A higher specific capacitance (CF) of 1583.9 F/g along with the capacity of 220.0 mAh/g are got for optimal CuCo2S4@Ni(OH)2 electrode, respectively comparing to 831.7 and 1063.3 F/g of CuCoS4 and Ni(OH)2 electrodes. A BSH is fabricated by the CuCo2S4@Ni(OH)2 electrode and the activated carbon electrode. The potential window of 1.6 V and maximum energy density of 32.1 Wh/kg at 1.1 kW/kg are achieved. The CF retention of 78% and Coulombic efficiency higher than 97% in 8000 charge/discharge cycles are also obtained for this BSH. © 2023

Possible solutions for oxygenation support in critically ill patients with COVID-19

Purpose: Due to the large number of patients with respiratory deficiency during the COVID-19 pandemic, several governments and their respective health care services have been studying ways to complement the care provided by offering immediate solutions. In view of this, the aim of this study was to carry out a systematic review of the advantages and disadvantages of possible solutions in oxygenation support. Methods: This systematic review used the PRISMA-P methodology and sought to list alternatives in oxygenation support that are being applied and studied worldwide. A bibliographic search was conducted in the MEDLINE and Cochrane Central databases, using the keywords SARS-CoV-2, COVID19, or coronavirus;combined with extracorporeal membrane oxygenation (ECMO), mechanical ventilation, mechanical ventilation support, low-cost, anesthesia, anesthesia machine, and ventilation therapy. The records were also found in the gray literature. Results: The search found 85 publications of which 41 articles were considered after excluding duplicate articles, reading the title and summary, and reading the articles in full. The oxygenation supports identified in these publications were the following: ECMO, shared mechanical ventilator, fast or low-cost production equipment, high-flow nasal cannula (HFNC), non-invasive ventilation, and use of anesthesia equipment as a mechanical ventilator. Conclusion: This study demonstrated the importance of a trained clinical team in the application of technologies. The alternatives found for support oxygenation require a more robust clinical evaluation to demonstrate their efficacy and safety for the COVID-19 patient. © 2021, Sociedade Brasileira de Engenharia Biomedica.
