The purpose of this research is to know the growth rate and biomass production of Scenedesmus obliquus in palm oil mill effluent (POME) medium taken from aerobic pond and to know the lipid concentration produced by S. obliquus. In this research, S. obliquus was cultured in various POME concentration, those were 20, 60, 80, 90, 95, and 100% v/v, while the concentration of inoculum was 30% v/v, and fatty acid content was analysed by gas chromatography. The results revealed that growth rate was decrease with the increasing concentration of POME (0.3545, 0.1792, 0.1566, 0.1268, 0.1158, and 0.1008/day), dry biomass weight was increase with the increasing concentration of POME (0.40, 1.20, 1.30, 1.77, 2.28, and 2.45 g/L), and lipid content also tended to increase with the increasing concentration of POME (13.10%, 36.59%, 36.65%, 50.37%, 60.40%, and 64.98%). Oleic acid and behenic acid were the dominant fatty acid content in S. obliquus lipid cultured in 100% concentration POME media. It is concluded that aerobic-pond POME can be utilized as growth medium for S. obliquus with a high lipid content. © Published under licence by IOP Publishing Ltd.

Solid-state fermentation with variation of inoculum volume in mold to increase the content of alpinetin in rice bran oil

Alpinetin (7-hydroxy-5-methoxyflavanone) is a flavonoid with anti-bacterial, anti-inflammatory, and other important therapeutic effects, so it has significant potential benefits. Rice husks or bran from rice are known to contain antioxidants, one of which is flavonoids. The addition of mold in the fermentation process can be used to enrich the lipid content of the fermented bran. Therefore, Aspergillus terreus chose as a mold. In this study, the fermentation process varied in the volume of mold inoculum with variables 1, 3, 5, 7, and 9 mL in the fermentation medium, respectively. Inoculum volume variables are related to the metabolic products of the mold used, Aspergillus terreus, where the metabolic products in the form of desaturase and elongase enzymes catalyze the formation of unsaturated fatty acids. This unsaturated fatty acid content was tested qualitatively using LC-MS/MS-TQF. The results showed that the volume of inoculum with a variation of 7 mL had the highest lipid yield of 19.94% and with the detection of alpinetin content in RT 13.04 with an area (%) of 31.03%.

The effect of additional carbon source and solvent ratio on the enrichment of alpinetin level in rice bran oil

Rice bran is a by-product of the rice milling process. Rice bran contains a variety of antioxidant compounds that are beneficial for the health of the body, such as vitamin E, oryzanol, and flavonoid compounds. One of the compounds classified in flavonoids is alpinetin which has antioxidant activity that can protect cells from free radicals in the body. Alpinetin is a flavonoid bioactive compound known to have various benefits, such as anti-inflammatory, anticancer, and antioxidant with low systemic toxicity. Enrichment of the content of bioactive compounds and fatty acids in rice bran oil can be carried out by a fermentation process using Aspergillus terreus. This research examined the effect of carbon source concentration and solvent ratio methanol: chloroform. This research used the solid-state fermentation method and modified the Bligh-Dyer extraction process. The extracted rice bran oil then tested using a Liquid Chromatography-Mass Spectrometry (LCMS) qualitatively. This research found that the addition of 10 g/L glucose produced the optimum lipid yield of 7.695%, and the highest alpinetin content was produced with the addition of glucose as much as 8g/L with alpinetin content of 27.11%. The ratio of methanol: chloroform =2:3.1 v/v produced the highest lipid yield, 9.54%, and the initial content of alpinetin compounds in rice bran extract increased from 0.02% to 3.43%.

Production of bioinsecticide from durian peel waste and Dioscorea hispida tuber extract by ultrasonic-assisted extraction

A new armyworm (Spodoptera frugiperda) is rapidly spreading and causing damage to corn crops in Indonesia, thus prompting the need to control the pest with an effective, yet environmentally friendly pest control method, such as bioinsecticide. Natural raw materials like durian peel waste and Dioscorea hispida tuber can be processed to produce a natural bioinsecticide. To discover the optimum processing condition for bioinsecticide production, research is conducted to find the effective solvent material and concentration in ultrasonic-assisted extraction methods. Durian peel waste is extracted using ethanol 30% and methanol 70% as solvents with the ratio variation of 1:0, 0:1, 1:1, 1:2, 2:1. Meanwhile, Dioscorea hispida tubers are extracted with various ethanol concentrations as solvents (ethanol 50%, 70%, 80%, absolute, and distilled water). The result shows that the most effective solvent for durian peel waste extraction is a 2:1 ratio of ethanol and methanol, with the optimum yield of 12.58%. The result also shows that Dioscorea hispida tuber is best extracted with distilled water, yielding 34.70%. The efficacy test concludes that the best solvent variation for pest elimination is a 0:1 ratio of ethanol and methanol durian peel waste and ethanol 80% for Dioscorea hispida tuber. The LC-MS/MS test results identified that alkaloid, flavonoid, and fatty acid compounds potentially caused larvae mortality.

Optimization of ultrasonic extraction parameters for enhanced total flavonoid content from Jengkol peel

Jengkol, a commonly consumed food ingredient, is frequently utilized without its outer layer. However, the outer peel of the jengkol fruit harbors a multitude of secondary metabolites that hold potential as bioinsecticides. Among these compounds, flavonoids stand out as toxic to insects. The quantification of flavonoid content within jengkol peel extracts is denoted as TFC (Total Flavonoid Content). Employing ultrasonic wave extraction, the conversion of jengkol peel into bioinsecticides can be facilitated. This research employs ultrasonic wave extraction, utilizing an ethanol solvent, with a frequency of 53 kHz and a temperature of 40℃. Through systematic variation of sonication duration and solvent concentration, this investigation scrutinizes the impact of these variables on TFC values and extracts yields. The assessment of TFC is conducted using UV-Vis spectrophotometry and referencing a quercetin standard solution. Optimal TFC output, specifically 1.643±0.026 mg QE/g of dried jengkol peel extract, is achieved at a 60-minute extraction period using 70% ethanol solvent. Liquid Chromatography-Mass Spectrometry (LCMS) analysis identifies various compounds with bioinsecticidal potential, including phenolic acids, fatty acids, flavonoids, phytoalexins, and coumarins, exhibiting the highest TFC levels. Employing ANOVA analysis followed by a post hoc LSD (Least Significance Different) test, it is evident with 95% confidence that both extraction time and solvent concentration significantly influence flavonoid content within jengkol peel extracts.

Investigating the influence of methanol:chloroform:water solvent ratios on the extraction process of tocotrienols from rice bran oil

Tocotrienols, belonging to the vitamin E family, exhibit neuroprotective, antioxidant, anticancer, and cholesterol-lowering attributes. A significant reservoir of tocotrienols is found within rice bran. Notably, rice bran oil stands as one of the most abundant vegetable oils in terms of tocotrienol content. This study aims to amplify tocotrienol levels in rice bran oil by fermenting rice bran with the fungus Aspergillus terreus, and by modulating the solvent ratio of methanol:chloroform:water during the extraction process. The research involves a comprehensive approach that encompasses solid-state fermentation of rice bran, succeeded by rice bran oil extraction utilizing the Bligh-Dyer method. This extracted oil is subsequently analyzed using UV spectrophotometry and GC/MS to ascertain tocotrienol levels within the rice bran oil. Findings from the study highlight an optimum solvent ratio of methanol:chloroform:water at 1:1.5:0.9 (v/v/v). Through this optimal configuration, the tocotrienol concentration escalates from 2541.44 ppm to 3642.79 ppm. Furthermore, the introduction of fermentation leads to a rise in tocotrienol levels within the extracted rice bran oil, ascending from 2541.44 ppm to 3257.66 ppm. The resultant rice bran oil extract also presents an array of antioxidant compounds, including n-hexadecanoic acid, benzoic acid, and chlorogenic acid. This collective insight underscores the potential for enhancing tocotrienol levels in rice bran oil through controlled fermentation and strategic solvent ratio adjustments, thereby enriching its potential health benefits.

The public widely uses Paracetamol (PCT) as an analgesic drug to reduce pain and fever symptoms in the human body. The COVID-19 pandemic has led to an increase in PCT consumption, resulting in its presence in water bodies and establishing it as a common aquatic environmental pollutant. Conventional methods for PCT removal rely on chemical treatments, which necessitate harsh reaction conditions and incur high operational costs. This study aims to investigate the potential for PCT degradation within the anode environment of the Microbial Fuel Cell (MFC) system, using a consortium of bacteria isolated from sewage sludge. The initial PCT concentration and the pH of the anode environment were varied. The results showed that PCT degradation occurred within 72 hours. The highest PCT removal rate was achieved at elevated concentrations, specifically 28.54 ± 18.84% for an initial PCT concentration of 31.45 mg/L. Furthermore, the pH levels of the anode environment were observed to influence PCT degradation, with the highest degradation rate recorded at 48.69 ± 0.86% at a pH of 8.2. In this study, the highest average power density reached was 1174.42 mW/m² with an external resistance of 1000 Ω at an initial PCT concentration of 31.45 mg/L. Microbial community analysis was conducted, revealing that Burkholderia sp. dominated the reactor containing PCT. These findings offer valuable insights for the development of more efficient technologies for the removal of PCT from pharmaceutical wastewater using natural microbial communities. © 2023 The Authors

In this work, a new thermostable and organic solvent-tolerant lipase produced from Rhizopus sp. strain PKC12B2 was characterized to evaluate its activity and stability toward pH, temperature, organic solvents, and surfactants. The catalytic efficiency of the produced enzyme was also analyzed through enzyme kinetics study. Produced lipase exhibits remarkable stability at pH 8.0 to pH 10.0 and shows optimum activity at pH 9.0, demonstrating alkali lipase properties. Lipase has an optimum temperature of 45 °C and possesses significant thermal stability in the temperature range between 45 and 55 °C. Exceptional enzyme stability was also demonstrated in the presence of both polar solvents, such as glycerol, isopropanol and acetone, as well as non-polar solvents, such as cyclohexane and heptane up to 30 % (v/v) solvent concentration, where the enzyme retained more than 80 % activity after 1-h pre-incubation. For methanol, ethanol, and n-octanol, the relative activity of lipase was highly stable in the presence of up to 20 % (v/v) solvent concentration. Na⁺, Zn²⁺, and Fe³⁺ ions significantly stimulate the lipase activity at a final concentration of 10 mM. Lipase also demonstrated stimulatory effects towards a broad concentration range (0.2–1 %) of surfactants Tween 80 and Triton X-100. The Linearized Michaelis-Menten model in the form of Lineweaver-Burk (R² = 0.995) and Hanes-Woolf (R² = 0.996) models was found to successfully display the enzyme kinetics, in which the former generated Km and V_max of 1.965 mM and 4.51 mM/min respectively, while the latter generated Km and V_max of 1.295 mM and 3.389 mM/min, respectively. These new enzyme attributes demonstrate its great potential in biotechnology applications, such as for detergent formulations, biodiesel production and organic synthesis industries. © 2024 The Authors

Molecular docking study of Keji Beling leave compound as protein 1J4X inhibitor to cervical cancer cell

VHR (Vaccinia H1-related phosphatase) is a protein that is involved in cervical cancer. 1J4X protein as a protein for cervical cancer cells is a structure of the VHR (Vaccinia H-1 related phosphate) receptor protein whose inhibition can reduce proliferation of cervical cancer cells. With current treatments which are expensive and cause side effects, alternative treatments are needed that are safe and do not cause side effects. Keji beling leaves are known to contain flavonoid compounds which are natural phenolic compounds that have the potential as anticancer. At present the design and development of new drugs can begin to be done in silico with biocomputing studies to analyze interactions known as molecular docking. With this method, the prediction of binding macromolecules (receptors) in the form of protein targets and small molecules (ligands) in the form of compound candidates can be carried out efficiently using software that utilizes docking algorithms and scoring functions. Molecular docking test results give the best docking score for quercetin which is -6.7, followed by n-hexadecanoic acid of -5.1 and finally 9-dodecenoic acid, methyl ester, (E) - worth -4.2 of 1J4X protein. Quercetin is a compound with the best inhibition potential against VHR which is indicated by the lowest docking score.

Molecular docking bioactive compound from Strobilanthes crispus to decrease cholesterol levels

Hypercholesterolemia is a disease caused by an increase in the concentration of cholesterol in the blood. One non-pharmacological treatment using herbal plants such as Strobilanthes crispus. This plant has flavonoid and phytosterols content that has been proven to reduce cholesterol levels in the blood. In this study, molecular docking was conducted to screen for the bioactive compounds that correspond to the interaction of the protein. Quercetin and β-sitosterol compounds are then docked using Autoodock Tools-1.5.6 to see the results of their affinity score for the HMG-CoA reductase enzyme. The affinity results obtained from quercetin, β-sitosterol and reference compounds (simvastatin) are -7.8 kcal/mol; -7.1 kcal/mol; and -6.1 kcal/mol. This shows that quercetin has the best affinity score for the inhibition of HMG-CoA reductase enzyme when compared with β-sitosterol and reference compounds.
