This work concerns a program (EFlow) that has been developed to assist experts in clustering flow cytometry multivariate data. EFlow aims to reduce subjectivity but keeping the expert's control over the analysis. In the present paper we will focus on the use of the iterative optimization algorithm. This algorithm is a hill-climbing unsupervised procedure that in general guarantees good local but not global optimization. A comparison between clustering processes achieved by an expert in flow cytometry and by EFlow was performed in a data related to bone marrow cells stained for the surface expression of molecules (B220, IgM and CD23) that characterize the B cell lineage. This data set is considered to present a medium difficult and had 63780 cells with 7 channels or dimensions. Two types of clustering processes were compared, one with four clusters and another with five clusters. The results obtained by EFlow were considered satisfactory by the expert as they were significantly in agreement with his classification. In some clusters, EFlow classified correctly up to 100% of the cells. At the present time few commercial cytometry software offer equivalent clustering analysis. © 2012 IEEE.

The human brain is a system consisting of various interconnected neural networks, with functional specialization coexisting with functional integration occurring both; temporally and spatially at many levels. The current study ranked and compared fast and slow participants in processing information by assessing latency and amplitude of early and late Event-Related Potential (ERP) components, including P200, N200, Premotor Potential (PMP) and P300. In addition, the Reaction Time (RT) of participants was compared and related to the respective ERP components. For this purpose, twenty right-handed and healthy individuals were subjected to a classical ERP "Oddball" paradigm. Principal Component Analysis (PCA) and Discriminant Function analyses (DFA) used PRE components and the Reaction Time (RT) to classify individuals. Our results indicate that latencies of P200 (O2 electrode), N200 (O2), PMP (C3) and P300 (Pz) components are significantly reduced in the group of fast responding participants. In addition, the P200 amplitude is significantly increased in the group of fast responding participants. Based on these findings, we suggest that the ERP is able to detect even minimal impairments, in the processing of somatosensory information and cognitive and motor stages. Hence, the study of ERP might also be capable of assessing sensorimotor dysfunctions in healthy old-aged people and in neuropsychiatric patients (suffering from dementia, Parkinson's disease, and other neurological disorders).

Temperature-induced changes in soft tissues analyzed by spectral methods and transient elastography: A comparative study

Some biological tissues, such as skeletal muscle, have regular or quasi-periodic structures. Periodicity of these structures can be investigated by spectral methods based on ultrasonic backscattered signals. These methods estimate the Mean Scatterer Spacing (MSS). In this work we analyzed the MSS dependence with temperature in bovine skeletal muscle tissue samples. Through spectral analysis we found that the relative variation of the MSS, in the temperature range 20 - 48°C was of ~ 2% /°C. For the same sample and under the same experimental conditions, we analyzed the propagation velocity of shear waves by transient elastography. It was determined that the relative variation of shear elastic modulus was ~ 3% /°C in the same temperature range. The repeatability of the results was tested in three tissue samples, with a set of 6 measurements on each. The relative variation is therefore similar for both parameters. Thus, this study shows that both techniques can be used as a method of noninvasive thermometry for small temperature range. In addition we emphasize the conceptual differences between both methods. As temperature changes, MSS estimation varies due to two physical phenomena. An apparent variation in mean spacing is produced because the sound speed is temperature-dependent, while tissue expansion produces a real change. In practice it is difficult to discriminate the weight of each component in the MSS variation. On the other side, transient elastography is less sensitive to thermal expansion. Thus it measures the actual shear wave speed change with temperature. This differentiation between both methods could be used to construct useful thermoelastic models for soft tissues in the future.

Many different factors can temporarily or permanently impair movement and impairs cortical organization, e.g. hand immobilization. Such changes have been widely studied using electroencephalography. Within this context, we have investigated the immobilization effects through the theta band coherence analysis, in order to find out whether the immobilization period causes any changes in the inter and intra-hemispheric coherence within the cerebral cortex, as well as to observe whether the theta band provides any information about the neural mechanisms involved during the motor act. We analyzed the cortical changes that occurred after 48 hours of hand immobilization. The theta band coherence was study through electroencephalography in 30 healthy subjects, divided into two groups (control and experimental). Within both groups, the subjects executed a task involving flexion and extension of the index finger, before and after 48 hours. The experimental group, however, was actually submitted to hand immobilization. We were able to observe an increase in the coupling within the experimental group in the frontal, parietal and temporal regions, and a decrease in the motor area. In order to execute manual tasks after some time of movement restriction, greater coherence is present in areas related to attention, movement preparation and sensorimotor integration processes. These results may contribute to a detailed assessment of involved neurophysiological mechanism in motor act execution.

The objective of this work was to identify the existence of biases on the reporting of Homeopathy efficacy in Brazilian newspapers. To this end, the online archives of two large newspapers one in Rio de Janeiro city and one in So Paulo city were searched for all news or reports concerning Homeopathy in the period January 2009December 2010. The identified items were then classified as favorable, unfavorable or neutral in what concerns the efficacy of Homeopathic therapy. Paid advertisements and passing references were excluded. Thirty-one items were identified, and the results indicated an overall preponderance of favorable, as compared to unfavorable, reports (14 favorable, 7 unfavorable). In conclusion, newspaper reporting of homeopathic efficacy is not compatible with the most update scientific evidence on the topic. © 2011 IEEE.

The present work aims at studying the effect of heating patterns generated by therapeutic ultrasound (US) insonation on tissue mimicking device called Phantom. With this purpose, it was constructed a cylindrical calorimeter (80 mm height and 42 mm diameter) which was filled with muscle mimic phantom, for acquisition of temperature an array of thermocouples was placed at 2 and 6 cm depth. Ultrasound physiotherapy equipment was used at two therapeutic frequency (1 and 3 MHz), three intensities, and continuous irradiation mode and applied for 300s static mode. The experimental temperatures elevation profiles in Phantom were compared to a 2D bio-heat equation model, permitting a critical evaluation of such experimental data. The acoustic heating model was based on the US absorption equation, assuming linear propagation and neglecting stationary wave and multiplie reverberation effects. The comparison between experimental and mathematical simulation for the central thermocouple showed that, for far field the correlation between the experimental and simulated values are higher than for the near field. © 2011 IEEE.

This work aims at identifying and evaluating the associations between the pharmaceutical assistance activities in hospital pharmacies services (HPS). The Multiple Correspondence Analysis (MCA) was applied to a sample of 238 hospitals and 30 variables from the survey Diagnosis of Hospital Pharmacy in Brazil. The first dimension of the MCA explained 90.6% of the data variability and allowed discriminate the HPS according to the presence or not of activities. It is noticeable an association between the presence of activities and better categories with large hospitals. © 2011 IEEE.

The present article proposes a methodology for simulating breast ultrasound (BUS) images for assessing segmentation algorithms. The main characteristics of the simulated BUS images are: (i) building an echogenicity map containing common tissues found in real breasts as well as their geometries and (ii) simulating the physical process of image acquisition. The validation procedure is based on measuring the correlation between the theoretical Rayleigh probability density function (PDF) with the one obtained from simulated speckle textures. The results pointed out that the PDF derived from simulated images are strongly correlated with the theoretical PDF with a coefficient of correlation of 0.999. © 2011 IEEE.

Periodicity of structures can be investigated with backscattered ultrasound, usually by applying spectral methods and estimating the Mean Scatterer Spacing (MSS). Biological tissues, such as skeletal muscle, or liver or trabecular bone, can present periodic or quasi-periodic structures. In this work it is studied the behavior of MSS with temperature and frequency for in vitro samples of bovine muscular tissues. MSS was obtained in the range of 37C to 43C, and between 1 MHz to 10.0 MHz Results show that, for the studied sample, MSS decreased with temperature for the muscle. It seems to be in agreement with the expected physiologic behavior of muscular fibers. Further complementary experiments are being envisaged. © 2011 IEEE.
