To combat the rapidly escalating COVID-19 epidemic in Italy,China’s third expert team against the COVID-19 pandemic in Italy departed on March 25,2020.I joined the expert team on March 25–April 8,*** the chairman of Italian Red Cross,Francesco Rocca,said he appreciated the Chinese experts who,after their arduous work on the front lines of China for over a month,chose to move ahead by flying to Italy to offer help(1).Medical experts of Italian National Institute of infectious diseases said that Chinese experts have gained a great deal of experience that Italy urgently needs,and that due to the considerable success China has had in fighting the epidemic,they wanted to work with Chinese experts on an international level in Italy(2).

Background:Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)vaccine can induce a potent cellular and humoral immune response to protect against SARS-CoV-2 ***,it was unknown whether SARS-CoV-2 vaccination can induce effective natural killer(NK)cell response in people living with human immunodeficiency virus(PLWH)and healthy ***:Forty-seven PLWH and thirty healthy controls(HCs)inoculated with SARS-CoV-2 inactivated vaccine were enrolled from Beijing Youan Hospital in this *** effect of SARS-CoV-2 vaccine on NK cell frequency,phenotype,and function in PLWH and HCs was evaluated by flow cytometry,and the response of NK cells to SARS-CoV-2 Omicron Spike(SARS-2-OS)protein stimulation was also ***:SARS-CoV-2 vaccine inoculation elicited activation and degranulation of NK cells in PLWH,which peaked at 2 weeks and then decreased to a minimum at 12 weeks after the third dose of ***,in vitro stimulation of the corresponding peripheral blood monocular cells from PLWH with SARS-2-OS protein did not upregulate the expression of the aforementioned ***,the frequencies of NK cells expressing the activation markers CD25 and CD69 in PLWH were significantly lower than those in HCs at 0,4 and 12 weeks,but the percentage of CD16^(+)NK cells in PLWH was significantly higher than that in HCs at 2,4 and 12 weeks after the third dose of ***,the frequency of CD16^(+)NK cells was significantly negatively correlated with the proportion of CD107a^(+)NK cells in PLWH at each time point after the third ***,this phenomenon was also observed in HCs at 0,2,and 4 weeks after the third ***,regardless of whether NK cells were stimulated with SARS-2-OS or not,we did not observe any differences in the expression of NK cell degranulation markers between PLWH and ***:SARS-CoV-2 vaccine elicited activation and degranulation of NK cells,indicating that the inoculation of

Premature Stop Codon at Residue 101 within HIV-1 Rev Does Not Influence Viral Replication of Clade BC but Severely Reduces Viral Fitness of Clade B

HIV-1 Rev is an accessory protein that plays a key role in nuclear exportation,stabilization,and translation of the viral *** of HIV-1 clade BC often shows a truncation of 16 AAs due to a premature stop codon at residue *** stop codon presents the highest frequency in clade BC and the lowest frequency in clade *** order to discover the potential biological effect of this truncation on Rev activity and virus replication of clade BC,we constructed Rev expression vectors of clade BC with or without 16 AAs within C-terminal separately,and replaced the stop codon by Q in a CRF07_BC infectious *** found that 16 AAs truncation had no effect on expression and activity of Rev in clade ***,the mutation from the stop codon to Q had no effect on virus replication of clade ***,to investigate the effect of this truncation on Rev activity and replication capacity of clade B,Rev expression vectors of clade B carrying or lacking 16 AAs in C-terminal were constructed respectively,and residue Q at position 101 within Rev was substituted by the stop codon in a clade B infectious *** was found that 16 AAs truncation significantly down-regulated Rev expression and impaired clade B Rev ***,a Q-to-stop codon substitution within Rev significantly reduced viral replication fitness of clade *** results indicate that the premature stop codon at residue 101 within Rev exerts diverse impact on viral replication among different HIV-1 clades.

Background:Antiretroviral therapy(ART)was often associated with dyslipidemia among human immunodeficiency virus(HIV)/acquired immunodeficiency syndrome(aids)*** study aimed to assess treatment-naïve adult male patients with HIV/aids who initiated ART with either co-formulated bictegravir,emtricitabine,and tenofovir alafenamide(BIC/FTC/TAF)or lamivudine,efavirenz,and tenofovir disoproxil fumarate(3TC+EFV+TDF),monitoring at weeks 4,12,24,and ***:A case-control retrospective study was *** newly diagnosed HIV-infected individuals attending the sexual transmission disease(std)/aids clinic of Beijing Youan Hospital,Capital Medical University,from January to December *** patients were divided into BIC/FTC/TAF group or 3TC+EFV+TDF ***-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C),triglyceride(TG),and total cholesterol(TC)at different time points over 48 weeks between two groups were compared.A multivariate Cox regression model was used to identify relevant influencing factors for the population at high risk of increased ***:A total of 870 participants,with 510 in BIC/FTC/TAF group and 360 in 3TC+EFV+TDF *** were no statistically significant differences in median age,baseline CD4/CD8 ratio,median body mass index(BMI)between the two *** both two groups,levels of TG,TC,and LDL-C were higher at 4 weeks,12 weeks,and 24 weeks of treatment(all P<0.05),and there were no statistically significant differences at 48 weeks compared to those at baseline(all P>0.05).In addition,the differences in average changes of the level of TG,TC,HDL-C,and LDL-C from weeks 4,12,24,and 48 to baseline between two groups were not statistically significant(all P>0.05).Multivariate Cox proportional risk model analysis showed that initiating ART with HIV RNA≥10^(5)copies/mL(compared with<10^(5)copies/mL)was associated with an increased risk of elevated LDL-C(hazard ratio=1.26,95%confidence interval:1.07-1.48,

Human immunodeficiency virus/acquired immunodeficiency syndrome prevalence,incidence,and mortality in China,1990 to 2017:a secondary analysis of the Global Burden of Disease Study 2017 data

Background:Despite almost two decades of well-funded and comprehensive response efforts by the Chinese Government,human immunodeficiency virus/acquired immunodeficiency syndrome(HIV/aids)remains a major problem in ***,few studies have recently examined long-term trends in HIV/aids prevalence,incidence,and mortality at the national *** study aimed to determine the prevalence,incidence,and mortality trends for HIV/aids over the past 28 years in ***:We conducted a descriptive,epidemiological,secondary analysis of the Global Burden of Diseases,Injuries,and Risk Factors Study 2017 *** evaluate trends in prevalence,incidence,and mortality over the study period from 1990 to 2017,we calculated values for annual percentage change(APC)and corresponding 95%confidence intervals(CIs)using joinpoint regression ***:A significant increase in HIV/aids prevalence was observed for 1990 to 2009(APC:10.7;95%CI:10.4,11.0;P<0.001),and then remained stable for 2009 to 2017(APC:0.7;95%CI:-0.3,1.7;P=0.1).A significant increase in HIV incidence was also observed for 1990 to 2005(APC:13.0;95%CI:12.6,13.4;P<0.001),and then a significant decrease was detected for 2005 to 2017(APC:-6.5;95%CI:-7.0,-6.1;P<0.001).A significant increase in aids-related mortality rate was detected for 1990 to 2004(APC:10.3;95%CI:9.3,11.3;P<0.001),followed by a period of stability for 2004 to 2013(APC:1.3;95%CI:-0.7,3.3;P=0.2),and then another significant increase for 2013 to 2017(APC:15.3;95%CI:8.7,22.2;P<0.001).Conclusions:Although prevalence has stabilized and incidence has declined,aids-related mortality has risen sharply in recent *** findings suggest more must be done to bring people into treatment earlier,retain them in treatment more effectively,actively seek to reenter them in treatment if they dropout,and improve the quality of treatment and care regimens.
