Presenilins(PSs)are the catalytic core of γ-secretase ***,the mechanism of FAD-associated PS mutations in AD pathogenesis still remains *** we review the general biology and mechanism of γ-secretase and focus on the catalytic components–presenilins and their biological functions and contributions to the AD *** functions of presenilins are divided into γ-secretase dependent and γ-secretase independent *** γ-secretase dependent functions of presenilins are exemplified by the sequential cleavages in the processing of APP and Notch;the γ-secretase independent functions of presenilins include stabilizing β-catenin in Wnt signaling pathway,regulating calcium homeostasis and their interaction with synaptic transmission.

This Podcast features an interview with Carla Rothlin and Sourav Ghosh, senior authors of a Research Article that appears in the 12 August 2014 issue of Science Signaling, about a potential target for the treatment of glioblastoma. Glioblastomas are brain tumors, and most are characterized by increased activity of the epidermal growth factor receptor (EGFR). Drugs that inhibit EGFR signaling have shown limited success in treating these tumors, suggesting that additional oncogenic signaling pathways need to be identified in order to effectively treat patients. Kusne et al. found that atypical protein kinase C (aPKC) was activated in glioblastoma cells by EGFR signaling and by the proinflammatory cytokine tumor necrosis factor–α (TNF-α), which is present in the tumor microenvironment. Inhibiting aPKC reduced the growth of glioblastomas in mice, suggesting that targeting aPKC may be an effective strategy for treating glioblastoma.

Pilot Data Assessing the Functional Integrity of the Default Network in Adult ADHD with fMRI and sLORETA

Intrinsic functional connectivity within the default mode network (DMN) of the brain has gained growing interest in attention deficit/hyperactivity disorder (ADHD). The DMN is proposed to support such core functions as theory of mind, self-related activities such as autobiographical self, stimulus independent thought, self-projection, self-reference, and introspective processes as well as central features of self-regulation, task compliance, and executive functions. Based on prior data showing that sLORETA and fMRI localize DMN regions with complementary accuracy, we hypothesized that this combination of methods could provide important information about the functional integrity of DMN connectivity with special attention between parietal and medial prefrontal regions. This study was conducted with 6 adults with ADHD and 7 age-similar, nonclinical controls. The present study recorded brain activity using both EEG and fMRI during rest and a Stroop paradigm. For this study only the eyes-opened rest data were analyzed using the sLORETA and fMRI psychophysiological interaction model respectively. sLORETA functional connectivity differences were assessed with Pearson's correlation measures and Fisher's Z test to examine the strength of the obtained coefficients between groups within the DMN. Differences in functional integrity of the DMN were found for most frequencies except theta, which did not show any difference between groups. Of interest, the ADHD group showed greater intrinsic functional connectivity in the alpha frequency range between medial prefrontal and left parietal regions. Functional connectivity within the DMN in both groups was comparable between both imaging modalities, with sLORETA providing evidence of the frequency-specific meaning of the associations. This study furthers the requisite for utilizing sLORETA in combination with fMRI to obtain knowledge of the connectivity in the context of EEG frequency-specific parameters. The hippocampus plays an important role in the encoding and retrieval of spatial and non-spatial memories. Much is known about the anatomical, physiological and molecular characteristics as well as the connectivity and synaptic properties of various cell types in the hippocampal circuits [1], but how these detailed properties of individual neurons give rise to the encoding and retrieval of memories remains unclear. Computational models play an instrumental role in providing clues on how these processes may take place. Here, we present three computational models of the region CA1 of the hippocampus at various levels of detail. Issues such as retrieval of memories as a function of cue loading, presentation frequency and learning paradigm, memory capacity, recall performance, and theta phase precession in the presence of dopamine neuromodulation and various types of inhibitory interneurons are addressed. The models lead to a number of experimentally testable predictions that may lead to a better understanding of the biophysical computations in the hippocampus.
