We highlight the work of a multi-university collaborative programme, PREMIERE (PREdictive modelling with QuantIfication of UncERtainty for MultiphasE systems), which is at the intersection of multi-physics and machine learning, aiming to enhance predictive capabilities in complex multiphase flow systems across diverse length and time scales. Our contributions encompass a variety of approaches, including the Design of Experiments for nanoparticle synthesis optimisation, Generalised Latent Assimilation models for drop coalescence prediction, Bayesian regularised artificial neural networks, eXtreme Gradient Boosting for microdroplet formation prediction, and a sub-sampling based adversarial neural network for predicting slug flow behaviour in two-phase pipe flows. Additionally, we introduce a generalised latent assimilation technique, Long Short-Term Memory networks for sequence forecasting mixing performance in stirred and static mixers, active learning via Bayesian optimisation to recover coalescence model parameters for high current density electrolysers, Gaussian process regression for drop size distribution predictions for sprays, and acoustic emission signal inversion using gradient boosting machines to characterise particle size distribution in fluidised beds. We also offer perspectives on the development of a shape optimisation framework that leverages the use of a multi-fidelity multiphase emulator. The results presented have applications in chemical synthesis, microfluidics, product manufacturing, and green hydrogen generation. © 2024, The Authors. All rights reserved.

Over the last two decades, the importance of epigenetics for interpretation of diverse genetic and genomic data has become increasingly clear. The need for study of indirect (non-gene) factors determining gene characteristic behavior and organism function, together with analysis of outcomes which are nondeterministic, is now well recognized. Given the increasing availability of large-scale datasets, analysis has inevitably become richer, but also more complex, and the formation of structured hypotheses, together with questions designed to refine experiment, relies heavily on computational tools. In particular, the effort to explore the whole genomic–epigenomic landscape has motivated an interdisciplinary approach towards large-scale multivariable and combinatorial analysis as well as novel database developments. Exploration of heritable changes in phenotype relies not only on newer sophisticated sequencing methods but also on legacy data, revisited for their contribution to understanding of transcriptional regulation and disease. The challenges presented are nontrivial, not least in terms of interpretation across multiple scales from cell to organism, but the field is advancing rapidly. With an early initial focus on cancers, both in development of models and database provision, work is emerging on brain function and neural pathways, while newer targets again are the behavioral sciences, in which interest is now burgeoning. In the following article, key developments and advances are summarized and current methods and tools reviewed.

In 2023, La Niña conditions that generally prevailed in the eastern Pacific Ocean from mid-2020 into early 2023 gave way to a strong El Niño by October. Atmospheric concentrations of Earth’s major greenhouse gases—carbon dioxide, methane, and nitrous oxide—all increased to record-high levels. The annual global average carbon dioxide concentration in the atmosphere rose to 419.3±0.1 ppm, which is 50% greater than the pre-industrial level. The growth from 2022 to 2023 was 2.8 ppm, the fourth highest in the record since the 1960s. The combined short-term effects of El Niño and the long-term effects of increasing levels of heat-trapping gases in the atmosphere contributed to new records for many essential climate variables reported here. The annual global temperature across land and oceans was the highest in records dating as far back as 1850, with the last seven months (June–December) having each been record warm. Over land, the globally averaged temperature was also record high. Dozens of countries reported record or near-record warmth for the year, including China and continental Europe as a whole (warmest on record), India and Russia (second warmest), and Canada (third warmest). Intense and widespread heatwaves were reported around the world. In Vietnam, an all-time national maximum temperature record of 44.2°C was observed at Tuong Duong on 7 May, surpassing the previous record of 43.4°C at Huong Khe on 20 April 2019. In Brazil, the air temperature reached 44.8°C in Araçuaí in Minas Gerais on 20 November, potentially a new national record and 12.8°C above normal. The effect of rising temperatures was apparent in the cryosphere, where snow cover extent by June 2023 was the smallest in the 56-year record for North America and seventh smallest for the Northern Hemisphere overall. CP2K is an open source electronic structure and molecular dynamics software package to perform atomistic simulations of solid-state, liquid, molecular and biological systems. It is especially aimed at massively-parallel and linear-scaling electronic structure methods and state-of-the-art ab-initio molecular dynamics simulations. Excellent performance for electronic structure calculations is achieved using novel algorithms implemented for modern high-performance computing systems. This review revisits the main capabilities of CP2K to perform efficient and accurate electronic structure simulations. The emphasis is put on density functional theory and multiple post-Hartree-Fock methods using the Gaussian and plane wave approach and its augmented all-electron extension. Research software has become a central asset in academic research. It optimizes existing and enables new research methods, implements and embeds research knowledge, and constitutes an essential research product in itself. Research software must be sustainable in order to understand, replicate, reproduce, and build upon existing research or conduct new research effectively. In other words, software must be available, discoverable, usable, and adaptable to new needs, both now and in the future. Research software therefore requires an environment that supports sustainability. Hence, a change is needed in the way research software development and maintenance are currently motivated, incentivized, funded, structurally and infrastructurally supported, and legally treated. Failing to do so will threaten the quality and validity of research. In this paper, we identify challenges for research software sustainability in Germany and beyond, in terms of motivation, selection, research software engineering personnel, funding, infrastructure, and legal aspects. Besides researchers, we specifically address political and academic decision-makers to increase awareness of the importance and needs of sustainable research software practices. In particular, we recommend strategies and measures to create an environment for sustainable research software, with the ultimate goal to ensure that software-driven research is valid, reproducible and sustainable, and that software is recognized as a first class citizen in research. Chronic care manages long-term, progressive conditions, while acute care addresses short-term conditions. Chronic conditions increasingly strain health systems, which are often unprepared for these demands. This study examines the burden of conditions requiring acute versus chronic care, including sequelae. Conditions and sequelae from the Global Burden of Diseases Study 2019 were classified into acute or chronic care categories. Data were analysed by age, sex, and socio-demographic index, presenting total numbers and contributions to burden metrics such as Disability-Adjusted Life Years (DALYs), Years Lived with Disability (YLD), and Years of Life Lost (YLL). Approximately 68% of DALYs were attributed to chronic care, while 27% were due to acute care. Chronic care needs increased with age, representing 86% of YLDs and 71% of YLLs, and accounting for 93% of YLDs from sequelae. These findings highlight that chronic care needs far exceed acute care needs globally, necessitating health systems to adapt accordingly. © 2025. The Author(s).

Cancer - uncontrolled cell proliferation, is caused by the deregulation of key genes that control cellular mechanisms, including division, differentiation, apoptosis, and movement. Here, we introduce LogisticCrypt, a logistic model on crypt structure at specific time points, which focuses on cell division, differentiation and apoptosis rates and on cell competition in crypt space. Comparative analysis of time-intervals for occurrence of the crypt phenomena is performed with respect to deregulation in intestinal cell number between the colon and small intestine crypts, obtained during cancer initiation and when carcinogen influence applies. Results are found to be more variable for the colon crypt, compared with the small intestine crypt, suggesting that the former is more sensitive to alterations in stem cell division rate than the latter. In addition, significantly higher carcinogen impact on crypt dynamics is evident in the colon crypt, compared with the small intestine crypt: cell proliferation time was observed to decrease by factors of 13 and 4.50 respectively, compared to situations when only the progenitor compartment was affected.

Epigenetics has emerged only recently as a fundamentally important area of biological and medical research that has implications for our understanding of human diseases including cancer, autoimmune and neuropsychiatric disorders. Here, we use a computational model developed for colon and small intestine crypt dynamics in order to investigate the effect of a set of potential methylation inhibitors with respect to methylation modifications in intestinal tissues during cancer initiation. Inhibitor effect was marked for abnormal crypt groups, with largest average methylation differences observed ≈0.75% in the colon and ≈0.79% in the small intestine lower when inhibitor was present (during relatively short time-periods of 90 days).

Epigenetic changes correspond to heritable modifications of the chromosome structure, which do not involve alteration of the DNA sequence but do affect gene expression. These mechanisms play an important role in normal cell differentiation, but aberration is associated also with several diseases, including cancer and neural disorders. In consequence, despite intensive studies in recent years, the contribution of modifications remains largely unquantified due to overall system complexity and insufficient data. Computational models can provide powerful auxiliary tools to experimentation, not least as scales from the sub-cellular through cell populations (or to networks of genes) can be spanned. In this paper, the challenges to development, of realistic cross-scale models, are discussed and illustrated with respect to current work.

Technologies for Ambient Assisted Living (AAL) combine new Information and Communication Technologies (ICT) to improve and increase the quality of life of the elderly. The SenseCam visual lifelogging device is now used not only to support memory recall, but also by research groups in other fields in order to investigate human lifestyle. Recent and continuing work in Dublin City University's SCI-SYM centre has been an application and evaluation of a novel approach, namely use of the cross correlation matrix and Maximum Overlap Discrete Wavelet Transform (MODWT) to analyse SenseCam lifelog data streams. By examination of the eigenspectrum, we show that these approaches enable detection of key sources or major events in the time SenseCam recording, with MODWT also providing useful insight on details of major events. In this paper, we analyse the data collected from the EvAAL (Evaluating AAL System Through Competitive Benchmarking) competition. The results confirmed our previous findings [1, 2]. We believe that highlighting key episodes to identify event boundaries can be used to develop automatic classifiers for visual lifelogs, helping to infer different lifestyle characteristics. © 2013 Springer-Verlag Berlin Heidelberg.
