Detecting biomolecules especially the disease markers, is an important issue for practical diagnosis and clinical therapies. Herein, we proposed an all-DNA hydrogel-based electrochemical impedance sensing platform for trace detection of miRNA-21 through the synergistic integration of dual amplification circuits: catalytic hairpin assembly (CHA) and clamped hybridization chain reaction (C-HCR). We designed a hairpin DNA with complementary sequences to the target miRNA-21, and the hairpin reactants in CHA were initiated to self-assemble into dsDNA products when added the target. This reaction generates numerous trigger sequences that activate the subsequent C-HCR to form DNA hydrogels on the electrode surface. The insulating DNA hydrogel film significantly enhances impedance responses, allowing us to establish a highly sensitive impedimetric biosensor for miRNA-21 analysis. Our dual signal amplification approach using DNA hydrogels as the amplification element showed higher electrochemical response, broader dynamic range (10-17-10-8 M), and lower detection limit (6.5 aM). In addition, this electrochemical impedimetric biosensor with excellent selectivity, non-destructivity, and label-free feature is suitable for practical analytes, highlighting its great potential in clinical diagnostics. © 2025, The Authors. Stability of complex communities: A perspective from discrete-time dynamics

Understanding the stability of complex communities is a central focus in ecology, many important theoretical advancements have been made to identify drivers of ecological stability. However, previous results often rely on the continuous-time dynamics, assuming that species have overlapping generations. In contrast, numerous real-world communities consist of species with non-overlapping generations, whose quantitative behavior can only be precisely represented by discrete-time dynamics rather than continuous ones. Here, we develop a theoretical framework and propose a metric to quantify the stability of complex communities characterized by non-overlapping generations and diverse interaction types. In stark contrast to existing results for overlapping generations, we find that increasing self-regulation strength first stabilizes and then destabilizes complex communities. This pattern is further confirmed in both exploitative (E. aerogenes, P. aurantiaca, P. chlororaphis, P. citronellolis) and competitive (P. putida, P. veroni, S. marcescens) soil microbial communities. Moreover, we show that communities with diverse interaction types become the most stable, which is corroborated by empirical mouse microbial networks. Furthermore, we reveal that the prevalence of weak interactions can stabilize communities, which is consistent with findings from existing microbial experiments. Our analyses of complex communities with non-overlapping generations provide a more comprehensive understanding of ecological stability and informs practical strategies for ecological restoration and control. This paper presents the results from an international survey that investigated the impacts of the built environment on occupant well-being during the corona virus disease 2019 (COVID-19) pandemic when most professionals were forced to work from home (WFH). The survey was comprised of 81 questions focusing on the respondent’s profiles, residences, home indoor environmental quality, health, and home working experiences. A total of 1460 responses were collected from 35 countries, and 1137 of them were considered complete for the analysis. The results suggest that home spatial layout has a significant impact on occupant well-being during WFH since home-life distractions and noises due to the lack of a personal workspace are likely to prevent productive work. Lack of scenic views, inadequate daylighting, and poor acoustics were also reported to be detrimental to occupant productivity and the general WFH experience. It is also revealed from this survey that temperature, relative humidity, and indoor air quality generally have higher satisfaction ratios compared with the indoor lighting and acoustic conditions, and the home layout. Hence, home design for lighting, acoustics, and layout should also receive greater attention in the future. Copyright © 2021 by ASME.

Second, it develops a flexible framework that utilizes a Large Language Model for OCR error correction and linguistic surface form detection in digitized corpora. Air infiltration has great importance regarding energy, comfort, moisture and dust deposition in naturally ventilated historical buildings like churches. In these buildings, air infiltration is driven by stack effect and wind. When assessing the wind effect, reasonable estimation of wind pressure coefficients is crucial. Local wind pressure coefficients are significantly affected by the shape of the building and turbulence generated by its geometrical features, which – for moderately large churches – typically consist of main building body and window recesses, gable roof, tower, apse, and sacristy. This study conducted wind tunnel and numerical investigations of wind pressure coefficients at a church, with gradual addition of these features. Wind pressure was measured on a small-scale model in a wind tunnel, while computational fluid dynamics (CFD) simulations were carried out for both small-scale and full-scale models. By comparing the experimental and numerical results, the SST k − ω turbulence model showed the best accuracy, yielding CFD results in good agreement with wind tunnel measurements. Most challenging was predicting the strong negative wind pressures occurring on a flat roof. Building structures like tower, apse, and sacristy had significant impact on the wind pressures on the central, main building body. This study adds knowledge on wind pressure effects by commonly occurring building components, with particular reference to churches and similar buildings. © 2024, The Authors. All rights reserved.

In this paper, we study both multi-armed and contextual bandit problems in censored environments. Our goal is to estimate the performance loss due to censorship in the context of classical algorithms designed for uncensored environments. Our main contributions include the introduction of a broad class of censorship models and their analysis in terms of the effective dimension of the problem - a natural measure of its underlying statistical complexity and main driver of the regret bound. In particular, the effective dimension allows us to maintain the structure of the original problem at first order, while embedding it in a bigger space, and thus naturally leads to results analogous to uncensored settings. Our analysis involves a continuous generalization of the Elliptical Potential Inequality, which we believe is of independent interest. We also discover an interesting property of decision- making under censorship: a transient phase during which initial misspecification of censorship is self-corrected at an extra cost; followed by a stationary phase that reflects the inherent slowdown of learning governed by the effective dimension. Our results are useful for applications of sequential decision-making models where the feedback received depends on strategic uncertainty (e.g., agents' willingness to follow a recommendation) and/or random uncertainty (e.g., loss or delay in arrival of information).

Traffic forecasting is an important issue in intelligent traffic systems (ITS). Graph neural networks (GNNs) are effective deep learning models to capture the complex spatio-temporal dependency of traffic data, achieving ideal prediction performance. In this paper, we propose attention-based graph neural ODE (ASTGODE) that explicitly learns the dynamics of the traffic system, which makes the prediction of our machine learning model more explainable. Our model aggregates traffic patterns of different periods and has satisfactory performance on two real-world traffic data sets. The results show that our model achieves the highest accuracy of the root mean square error metric among all the existing GNN models in our experiments. © 2023, CC BY-SA.

如何实现人类社会经济的可持续发展是本世纪的重大挑战之一。近几十年来,中国经济实力大幅提升,国际影响力显著增强。然而,经济社会高速发展可能会给生态环境造成诸多不利影响,其中之一就是人类活动污水排放总量的快速增长。长江经济带作为中国高质量发展的重点区域,科学评估和揭示沿江城市群污水治理成效及影响因素至关重要。本研究核算了2007-2017年间长江流域主要污染物的浓度变化趋势,利用数据驱动型技术对影响污染物通量变化的关键因子及其贡献开展了系统解耦和量化分析,揭示了污染物排放强度的时空异质性,解析了沿江城镇污水系统发展对污染物通量削减的作用效果。研究发现,长江流域水污染治理成效总体向好,长江经济带国内生产总值和沿江人类活动污水排放强度之间的强耦合在2013年之后呈现脱钩趋势,这主要得益于城镇排水和污水厂基础设施建设的快速发展。研究提出,长江流域城镇污水治理如何实现以水载资源再生利用为导引的“绿色零碳模式”转变,对于全面改善长江流域的水环境质量具有关键作用,同时也能为区域生态环境保护与经济高质量发展提供有益启示和借鉴。

Improvement of Waste Sludge Dewaterability by a Novel Co-Conditioning Approach with Polyaluminum Chloride-Sludge Based Carbon-Polyacrylamide

A novel sludge conditioning method incorporating sludge-based carbon (SBC) as a filter aid into the coagulation and flocculation co-conditioning process is proposed to strengthen sludge dewaterability, namely the co-conditioning of polyaluminum chloride (PAC)-SBC-polyacrylamide (PAM). The results show that the co-conditioning method can effectively reduce the specific resistance to filtration (SRF), moisture content (MC) of the filter cake and sludge compressibility coefficient (CC), meanwhile increase the net sludge solids yield (YN) of municipal sludge (MS) and textile dyeing sludge (TDS), resulting in a significant improvement in sludge dewatering performance for two types of sludge. The response of sludge dewatering performance to different conditioning agents and the variation characteristics of sludge EPS in co-conditioning process suggest that the co-conditioning mechanism involves a synergistic effect of coagulation, adsorption, and flocculation, which process includes the aggregation and disintegration of microbial cell flocs caused by PAC, the skeleton-building action and biopolymer adsorption by SBC, and sludge floc agglomeration facilitated by PAM. In addition, SBC also optimizes coagulation by electrostatic neutralization, while strengthening flocculation by bridging biopolymers and PAM. Moreover, co-conditioning exhibits a remarkable ability to eliminate dissolved organic matter from the filtrate, leading to an enhancement in the filtrate quality and a reduction in the downstream burden for sludge filtrate treatment. Consequently, the PAC-SBC-PAM co-conditioning emerges as a promising method for sludge pretreatment. © 2023, The Authors. All rights reserved.
