The design of CRISPR-Cas9 guide RNAs requires the analysis of all potential target sites to determine the risk of unintentional edits for any given guide. A number of methods have been developed to evaluate this risk but the size of the target genome is a challenging factor. Many existing methods are limited by their need for an unfeasible amount of time or memory. In this paper, we introduce Crackling++, an update to our previous Crackling method, in which we have reduced processing time by imposing stricter matching criteria. We conducted a benchmark to compare our performance to other tools and show that Crackling++ is the fastest off-targeting scoring method and that it can scale to the largest genomes. © 2024 IEEE.

Dynamic flexible job shop scheduling is an important combinatorial optimization problem that has rich real-world applications such as product processing in manufacturing. Genetic programming has been successfully used to learn scheduling heuristics for dynamic flexible job shop scheduling. Intuitively, users prefer small and effective scheduling heuristics that can not only generate promising schedules but also are computationally efficient and easy to be understood. However, a scheduling heuristic with better effectiveness tends to have a larger size, and the effectiveness of rules and rule size are potentially conflicting objectives. With the traditional dominance relation-based multiobjective algorithms, there is a search bias toward rule size, since rule size is much easier to optimized than effectiveness, and larger rules are easily abandoned, resulting in the loss of effectiveness. To address this issue, this article develops a novel multiobjective genetic programming algorithm that takes size and effectiveness of scheduling heuristics for optimization via multitask learning mechanism. Specifically, we construct two tasks for the multiobjective optimization with biased objectives using different search mechanisms for each task. The focus of the proposed algorithm is to improve the effectiveness of learned small rules by knowledge sharing between constructed tasks which is implemented with the crossover operator. The results show that our proposed algorithm performs significantly better, i.e., with smaller and more effective scheduling heuristics, than the state-of-the-art algorithms in the examined scenarios. By analyzing the population diversity, we find that the proposed algorithm has a good balance between exploration and exploitation during the evolutionary process. © 2024 IEEE.

Neural wave functions accomplished unprecedented accuracies in approximating the ground state of many-electron systems, though at a high computational cost. Recent works proposed amortizing the cost by learning generalized wave functions across different structures and compounds instead of solving each problem independently. Enforcing the permutation antisymmetry of electrons in such generalized neural wave functions remained challenging as existing methods require discrete orbital selection via non-learnable hand-crafted algorithms. This work tackles the problem by defining overparametrized, fully learnable neural wave functions suitable for generalization across molecules. We achieve this by relying on Pfaffians rather than Slater determinants. The Pfaffian allows us to enforce the antisymmetry on arbitrary electronic systems without any constraint on electronic spin configurations or molecular structure. Our empirical evaluation finds that a single neural Pfaffian calculates the ground state and ionization energies with chemical accuracy across various systems. On the TinyMol dataset, we outperform the 'gold-standard' CCSD(T) CBS reference energies by 1.9 mEh and reduce energy errors compared to previous generalized neural wave functions by up to an order of magnitude. © 2024 Neural information processing systems foundation. Problems related to the quality of games, whether on the initial release or after updates, can lead to player dissatisfaction, media attention, and potential financial setbacks. These issues can stem from software bugs, performance bottlenecks, or security vulnerabilities. Despite these challenges, game developers often rely on manual playtesting, highlighting the need for more robust and automated processes in game development. This research explores the application of Large Language Models (LLMs) to automate the creation of unit tests in game development, focusing on strongly typed programming languages such as C++ and C#, which are widely used in the industry. The study focuses on fine-tuning Code Llama, an advanced code generation model, to address common scenarios in game development, including game engines and specific APIs or backends. Although the prototyping and evaluations primarily took place within the Unity game engine, the proposed methods can be adapted to other internal or publicly available solutions. The evaluation results demonstrate these methods' effectiveness in improving existing unit test suites or automatically generating new tests based on natural language descriptions of class contexts and targeted methods. © 2024 The Authors.

Effectiveness of a touch-based hand therapeutic application for hand dexterity in children with handwriting difficulties-a pilot study

Handwriting is an important skill for children during their academic years. It is the coordination of perceptual-motor and cognitive abilities. Some children have difficulties in handwriting, which is an indication of issues in their fine motor skills. Nowadays, technology-based instructions are available to develop fine motor skills in children with handwriting disabilities. This paper describes the details of a newly developed software application namely ‘HanDex’ for children with handwriting difficulties and the experiments conducted to test its effectiveness. The application comprises six hand therapeutic activities which were designed based on the oscillation theory of handwriting. Initially, in the first phase, the experiments were conducted in nine primary school children from various schools, aged 8–12 years. Then, in the second phase, the software training was given to six upper primary school children (10–12 years) with handwriting issues. The results of using this application were promising. So one more study was conducted in another set of nine children (8–11 years) with handwriting difficulties. The participants were assessed by their special educators, class teachers and parents. After the pretest, they received a total of 18 handwriting practice sessions using this new application. At the end of the training sessions, a post test was conducted, to measure the progress in handwriting. The handwriting rubrics with eight handwriting parameters such as letter formation, placement, letter sizing, spacing of letters, legibility, speed, neatness, and spacing of words were used for handwriting assessment. The findings from the study suggested that touch-based hand therapeutic activities were beneficial for children with writing difficulties to improve their handwriting performance and tripod grip. © The Author(s), under exclusive licence to Springer science+Business Media, LLC, part of Springer Nature 2024.

Despite the various advances in medical technology, heart disease continues to rank among the leading causes of mortality in the world, killing millions each year. There is hope that the risks involved with heart disease can be reversed in time and the outcomes of patients improved with earlier prediction and diagnosis. However, the use of conventional methods of diagnostic techniques tends to be very resource-intensive and inaccessible to other more disadvantaged areas. The present work defines a model for machine learning in predicting heart disease and illustrates a more efficient and accessible solution. There is a risk for heart disease that is determined using a set of various clinical features. The algorithm used in the prediction of heart disease is XG Boost that are applied to the dataset. The method provides a reliable tool for the early detection of heart disease, thus helping healthcare providers to improve preventive care and treatment strategies. The results of this study will be helpful in advancing research to identify other avenues for prediction of heart disease. © 2024 IEEE.

Feature Selection(FS)is an important data management technique that aims to minimize redundant information in a *** work proposes DENGO,an improved version of the Northern Goshawk Optimization(NGO),to address the FS *** NGO is an efficient swarm-based algorithm that takes its inspiration from the predatory actions of the northern *** order to overcome the disadvantages that NGO is prone to local optimum trap,slow convergence speed and low convergence accuracy,two strategies are introduced in the original NGO to boost the effectiveness of ***,a learning strategy is proposed where search members learn by learning from the information gaps of other members of the population to enhance the algorithm's global search ability while improving the population ***,a hybrid differential strategy is proposed to improve the capability of the algorithm to escape from the trap of the local optimum by perturbing the individuals to improve convergence accuracy and *** prove the effectiveness of the suggested DENGO,it is measured against eleven advanced algorithms on the CEC2015 and CEC2017 benchmark functions,and the obtained results demonstrate that the DENGO has a stronger global exploration capability with higher convergence performance and ***,the proposed DENGO is used for FS,and the 29 benchmark datasets from the UCL database prove that the DENGO-based FS method equipped with higher classification accuracy and stability compared with eight other popular FS methods,and therefore,DENGO is considered to be one of the most prospective FS ***'s code can be obtained at https://***/matlabcentral/fileexchange/158811-project1.

Cloud computing is a technology that allows the utilisation of a vast network of computers that are distributed and run in parallel with one another. The management of the multimedia files presents challenges and the significant of which is the prevalence of attackers and users with malicious intent. This article presents the conceptual foundation for a four-tiered data security paradigm that can be implemented in cloud computing. The proposed framework employs a DNA-computing-based password or secret key with a length of 1024 bits for the purpose of encrypting the user private multimedia file or correspondence. The DNABMS algorithm is applied in the cryptographic generation of digital identities as well as pseudorandom number generators. The results of the testing that was done on the model have validated both its reliability and its efficacy, demonstrating that the model that was developed is both reliable and effective. Experiments have demonstrated that the developed method is superior to other well-known existing strategies in terms of both effectiveness and efficiency. © 2023 IEEE.

Logs as semi-structured text are rich in semantic information, making their comprehensive understanding crucial for automated log analysis. With the recent success of pre-trained language models in natural language processing, many studies have leveraged these models to understand logs. Despite their successes, existing pre-trained language models still suffer from three weaknesses. Firstly, these models fail to understand domain-specific terminology, especially abbreviations. Secondly, these models struggle to adequately capture the complete log context information. Thirdly, these models have difficulty in obtaining universal representations of different styles of the same logs. To address these challenges, we introduce KnowLog, a knowledge-enhanced pre-trained language model for log understanding. Specifically, to solve the previous two challenges, we exploit abbreviations and natural language descriptions of logs from public documentation as local and global knowledge, respectively, and leverage this knowledge by designing novel pre-training tasks for enhancing the model. To solve the last challenge, we design a contrastive learning-based pre-training task to obtain universal representations. We evaluate KnowLog by fine-tuning it on six different log understanding tasks. Extensive experiments demonstrate that KnowLog significantly enhances log understanding and achieves state-of-the-art results compared to existing pre-trained language models without knowledge enhancement. Moreover, we conduct additional experiments in transfer learning and low-resource scenarios, showcasing the substantial advantages of KnowLog.
