Arabidopsis is a widely used model plant to gain basic knowledge on plant physiology and development. Live imaging is an important technique to visualize and quantify elemental processes in plant development. To uncover novel theories underlying plant growth and cell division, accurate cell tracking on live imaging is of utmost importance. The commonly used cell tracking software, TrackMate, adopts tracking-by-detection fashion, which applies Laplacian of Gaussian (LoG) for blob detection, and Linear Assignment Problem (LAP) tracker for tracking. However, they do not perform sufficiently when cells are densely arranged. To alleviate the problems mentioned above, we propose an accurate tracking method based on Genetic algorithm (GA) using knowledge of Arabidopsis root cellular patterns and spatial relationship among volumes. Our method can be described as a coarse-to-fine method, in which we first conducted relatively easy line-level tracking of cell nuclei, then performed complicated nuclear tracking based on known linear arrangement of cell files and their spatial relationship between nuclei. Our method has been evaluated on a long-time live imaging dataset of Arabidopsis root tips, and with minor manual rectification, it accurately tracks nuclei. To the best of our knowledge, this research represents the first successful attempt to address a long-standing problem in the field of time-lapse microscopy in the root meristem by proposing an accurate tracking method for Arabidopsis root nuclei. © 2025, CC BY.

Complex processes often arise from sequences of simpler interactions involving a few particles at a time. These interactions, however, may not be directly accessible to experiments. Here we develop the first efficient method for unravelling the causal structure of the interactions in a multipartite quantum process, under the assumption that the process has bounded information loss and induces causal dependencies whose strength is above a fixed (but otherwise arbitrary) threshold. Our method is based on a quantum algorithm whose complexity scales polynomially in the total number of input/output systems, in the dimension of the systems involved in each interaction, and in the inverse of the chosen threshold for the strength of the causal dependencies. Under additional assumptions, we also provide a second algorithm that has lower complexity and requires only local state preparation and local measurements. Our algorithms can be used to identify processes that can be characterized efficiently with the technique of quantum process tomography. Similarly, they can be used to identify useful communication channels in quantum networks, and to test the internal structure of uncharacterized quantum circuits. Copyright © 2021, The Authors. Fano 3-Folds, Reflexive Polytopes and Brane Brick Models

Reflexive polytopes in n dimensions have attracted much attention both in mathematics and theoretical physics due to their connection to Fano n-folds and mirror symmetry. This work focuses on the 18 regular reflexive polytopes corresponding to smooth Fano 3-folds. For the first time, we show that all 18 regular reflexive polytopes have corresponding 2d (0, 2) gauge theories realized by brane brick models. These 2d gauge theories can be considered as the worldvolume theories of D1-branes probing the toric Calabi-Yau 4-singularities whose toric diagrams are given by the associated regular reflexive polytopes. The generators of the mesonic moduli space of the brane brick models are shown to form a lattice of generators due to the charges under the rank 3 mesonic flavor symmetry. It is shown that the lattice of generators is the exact polar dual reflexive polytope to the corresponding toric diagram of the brane brick model. This duality not only highlights the close relationship between the geometry and 2d gauge theory, but also opens up pathways towards new discoveries in relation to reflexive polytopes and brane brick models. PURPOSE:Pediatric hydrocephalus is the most common cause of surgically treatable neurological disease in children. Controversies exist whether endoscopic third ventriculostomy (ETV) or cerebrospinal fluid (CSF) shunt placement is the most appropriate treatment for pediatric hydrocephalus. This study aimed to compare the risk of re-operation and death between the two procedures. METHODS:We performed a retrospective population-based cohort study and included patients younger than 20-years-old who underwent CSF shunt or ETV for hydrocephalus from the Taiwan National Health Insurance Research Database. RESULTS:A total of 3,555 pediatric patients from 2004 to 2017 were selected, including 2,340 (65.8%) patients that received CSF shunt placement and 1215 (34.2%) patients that underwent ETV. The incidence of all-cause death was 3.31 per 100 person-year for CSF shunt group and 2.52 per 100 person-year for ETV group, with an adjusted hazard ratio (HR) of 0.79 (95% confidence interval [CI] = 0.66-0.94, p = 0.009). The cumulative incidence competing risk for reoperation was 31.2% for the CSF shunt group and 26.4% for the ETV group, with an adjusted subdistribution HR of 0.82 (95% CI = 0.70-0.96, p = 0.015). Subgroup analysis showed that ETV was beneficial for hydrocephalus coexisting with brain or spinal tumor, central nervous system infection, and intracranial hemorrhage. CONCLUSION:Our data indicates ETV is a better operative procedure for pediatric hydrocephalus when advanced surgical techniques and instruments are available.

This paper presents our work to enhance a state-of-the-art level generator (Sketch-to-Level Generator) that generates levels for an Angry-Birds-like game from drawn sketches. To achieve this task, Cycle-Consistent Adversarial Networks (Cycle-GAN) are used. CycleGAN is trained using two datasets: sketch drawings and typical level-structures. The former are taken from Google's Quick, Draw! datasets, and the latter from the winning level generator at the 2017 and 2018 AIBIRDS level generation competitions. The output of the trained CycleGAN is used as the input of Sketch-to-Level Generator. Our results show that the proposed preprocessing technique using CycleGAN allows Sketch-to-Level Generator to more successfully generate levels from arbitrary sketch drawings.

Microservice application developers try to mitigate the impact of partial outages typically by implementing service-to-service interactions that use well-known resiliency patterns, such as Retry, Fail Fast, and Circuit Breaker. However, those resiliency patterns-as well as their available open-source implementations-are often documented informally, leaving it up to application developers to figure out when and how to use those patterns in the context of a particular microservice application. In this paper, we take a first step towards improving on this situation by introducing a model checking-based approach in which we use the PRISM probabilistic model checker to analyze the behavior of the Retry and Circuit Breaker resiliency patterns as continuous-time Markov chains (CTMC). This approach has enabled us to quantify the impact of applying each resiliency pattern on multiple quality attributes, as well as to determine how to best tune their parameters to deal with varying service availability conditions, in the context of a simple client-service interaction scenario.

People with color vision deficiency (CVD) may have difficulty in discriminating colors. To improve their color perception, several compensation methods have been proposed which considered naturalness maintenance and contrast emphasis. All these methods are based on the simulation model of severe CVD and hence it is not clear whether are also effective for people with light CVD. In this paper, we conduct subjective study to evaluate the effectiveness of these methods for people with varying degrees of CVD.

The administration of recombinant cytokines, particularly interleukin-12 (IL-12), holds promising clinical potential for treating various cancers. Sustained intratumoral delivery of IL-12 can restore tumor resident CD8 + effector T cells and induce the priming of antitumor CD8 + effector T cells. However, these CD8 + T cell-dependent anticancer efficacy is usually transient and accompanies the activation of immune suppressive CD4 + Foxp3 + T regulatory cells. The underlying mechanism of T regulatory cell activation in IL-12 therapy is the upregulation of IFNγ dependent indoleamine 2,3-dioxygenase (IDO) expression. Due to this negative feedback, the combinatorial use of drugs should be considered to enhance the efficacy of IL-12-mediated therapy. Herein, we designed a lipid nanoparticle (LNP) system which can successfully deliver an IDO inhibitor indoximod (IND) and IL-12 encoding mRNA. In order to increase the loading efficiency, the IND prodrug was synthesized by conjugating IND with cholesterol by ester linkage. Optimized IND prodrug encapsulating LNP successfully transfected tumor cells and macrophages, resulting in the secretion of IL-12 cytokine. With IL-12 transfection, macrophages upregulated T cell co-stimulation factor and released TNFα cytokine, indicating that the tumor microenvironment could be changed from cold tumor to hot tumor for immunotherapy. Also, by the elevated secretion of IL-12 cytokine, T cells release high levels of IFNγ, which is a central role in IL-12-mediated immunotherapy. This co-delivery system presents a promising strategy to overcome the limitations of single IL-12-mediated therapy by simultaneously promoting antitumor immune responses and inhibiting immunosuppressive mechanisms, thereby enhancing the overall efficacy of cancer immunotherapy.

Detecting manipulated images and videos is an important aspect of digital media forensics. Due to severe discriminative information loss caused by resolution degradation, the performance of most existing methods is significantly reduced on low resolution manipulated images. To address this issue, we propose an Artifacts-Focus Super-Resolution (AFSR) module and a Two-stream Feature Extractor (TFE). The AFSR recovers facial cues and manipulation artifact details using an autoencoder learned with an artifacts focus training loss. The TFE adopts a two-stream feature extractor with key points-based fusion pooling to learn discriminative facial representations. These two complementary modules are jointly trained to recover and capture distinctive manipulation artifacts in low resolution images. Extensive experiments on two benchmarks including FaceForensics++ and DeepfakeTIMIT, evidence the favorable performance of our method against other state-of-the-art methods.
