To investigate the genetic basis of type 2 diabetes (T2D) to high resolution, the GoT2D and T2D-GENES consortia catalogued variation from whole-genome sequencing of 2,657 European individuals and exome sequencing of 12,940 individuals of multiple ancestries. Over 27M SNPs, indels, and structural variants were identified, including 99% of low-frequency (minor allele frequency [MAF] 0.1-5%) non-coding variants in the whole-genome sequenced individuals and 99.7% of low-frequency coding variants in the whole-exome sequenced individuals. Each variant was tested for association with T2D in the sequenced individuals, and, to increase power, most were tested in larger numbers of individuals (>80% of low-frequency coding variants in ~82 K Europeans via the exome chip, and ~90% of low-frequency non-coding variants in ~44 K Europeans via genotype imputation). The variants, genotypes, and association statistics from these analyses provide the largest reference to date of human genetic information relevant to T2D, for use in activities such as T2D-focused genotype imputation, functional characterization of variants or genes, and other novel analyses to detect associations between sequence variation and T2D.

There are many university initiatives that focus on technology-based solutions to address the needs of marginalized communities. The technology-based solutions are intended to be economically and socially sustainable. These endeavors are usually well-meaning, creatively designed, and enthusiastically deployed, but do not achieve the sustainable impact envisioned at the outset of the projects. To addresses these shortcomings, at The Pennsylvania State University we are applying three key tenets of systems thinking to our humanitarian engineering and social entrepreneurial ventures: 1) employing regulation via feedback to ensure that the system is actually working;2) defining systems by their interactions and their parts;and 3) understanding that systems exhibit multi-finality. The concept of multi-finality refers to (designing) a system where the individual actors (inputs), the subsystems, and their interactions, all meet their own goals while the system as a whole also meets its goals. In this paper, we lay the framework for the application of specific systems thinking concepts to increase the probability of success of global development ventures. We provide simple yet compelling examples from two different ventures to illustrate the power of systems thinking to train innovative problem-solvers and increase the probability of success of technology-based social entrepreneurial ventures in Africa. © American Society for Engineering Education, 2010.

Emotionally aware agents used to support human-computer interactions form the basis for our hybrid mediated agent architecture. This paper expands on our framework for an emotionally aware interface architecture that couples with the human user where data is mediated between agent processes for adaptive controlled assistive technologies. We present a hybrid emotionally aware mediated agent architecture that couples human and artificial operators. Agent behaviors in the context of emotion are derived from interactions from human users and interactions with its environment. Through the use of a multi-agent system composed of emotionally enhanced sets of agents, we investigate how emotion plays a role in inter-agent communication, cooperation, goal achievement and perception. The behaviors of each agent are derived from its perception of the environment, its goals, knowledge gained from other agents and based on its own current emotional state, which is predicated on fuzzy sets. The state of each interacting agent is determined by its level of frustration combined with its interaction with other agents. The set of actions an agent may perform including that of its own ability to sense and understand the environment is limited by its current level of emotional context. In this work we focus on the analysis of the interaction of agents and review the grouping effect that arises from the inter-agent communications combined with the intra-agent emotional status as they perform the task required by the environment. The environment is based on previous work performed on navigational map learning by context chaining and abstraction.

It is time to recognize that educational technology cannot be considered a panacea for educational reform. technology, when properly used, can be a very effective tool for improving and enhancing instruction and learning experiences in the content areas. However, incorporation of electronic tools, such as computers and videodiscs, into classrooms must be accompanied by systemic change in the educational process. Successful utilization of technology depends on how we provide teachers with the necessary environment for training, tools for instruction, and technology evaluation skills. This paper examines some of the misuses of technology and then explores the appropriate use of technology in the context of current educational reform. The focus is on the integration of technology in the educational process that provides a learning environment allowing discovery and creativity through the use of computer visualizations. The teacher is seen as the essential element, requiring a solid foundation in the general applications of technology, and the ability to evaluate and select systems that are effective and efficient for specific applications.

The principal stages in a rational strategy for the investigation of reaction mechanisms are described, in which the key stage is the formulation of hypotheses at the initial stage of the study. computer programs are examined in which hypotheses are formulated about the mechanisms of complex reactions with participation of the user. Empirical and formal-logical algorithms and programs as well as various aspects of their application are discussed. The programs are compared with those for computer-aided organic synthesis design, which are similar in their structures and algorithms.
