Human pluripotent stem cells as tools for high-throughput and high-content screening in drug discovery

:A significant bottleneck in drug discovery is the lack of suitable models for sensitive, reliable, and rapid assessment of lead molecules in preclinical stages of drug discovery. Human pluripotent stem cells (hPSCs) derived either from early human blastocysts (human embryonic stem cells) or by reprogramming somatic cells to a pluripotent state (human-induced pluripotent stem cells) can be propagated extensively in vitro while retaining the ability to differentiate into any specialized cell type within the body. In this review, we discuss how these unique features of hPSCs could offer a way of producing relevant in vitro models amenable to high-throughput testing for drug discovery. We summarize recent progress in inducing differentiation of hPSCs to specific cell types, and describe the ongoing efforts in applying hPSCs and their differentiated derivatives in disease modeling, drug discovery, and developmental toxicology. Moreover, we review the applications of high-content imaging assays in detecting the changes in the phenotype of hPSCs and their differentiated progeny. Finally, we highlight challenges that need to be overcome in order for the application of hPSC technology to fully benefit drug discovery.

Most of the best performing link prediction ranking measures evaluate the common neighbourhood of a pair of nodes in a network, in order to assess the likelihood of a new link. On the other hand, the same zero rank value is given to node pairs with no common neighbourhood, which usually are a large number of potentially new links, thus resulting in very low quality overall link ranking in terms of average edit distance to the optimal rank. In this paper we introduce a general technique for improving the quality of the ranking of common neighbours-based measures. The proposed method iteratively applies any given ranking measure to the quasi-common neighbours of the node pair. Experiments held on widely accepted datasets show that QCNAA, a quasi-common neighbourhood measure derived from the well know Adamic-Adar (AA), generates rankings which generally improve the ranking quality, while maintaining the prediction capability of the original AA measure.

This paper presents a Single-path Delay Feedback (SDF) architecture for implementing a Fast Fourier Transform (FFT) processor on FPGA for Multiple Input Multiple Output Orthogonal Frequency Division Multiplexing. An FPGA resource efficient and shared multiplier technique for parallel processing of two and four data streams is demonstrated and utilized for realization within the presented SDF architecture based on a radix-2 butterfly model. The presented approach allows significant utilization efficiency of FPGA hardware-based multiplier elements opposed to a relative 50% less efficiency within conventional radix-2 SDF FPGA implemented techniques. The FPGA implementation demonstrated significant FPGA space resource savings compared to conventional radix-2 SDF methods and has been evaluated with other relative hardware architecture techniques. Additionally, the presented architecture is suitable for implementing and scaling to any FFT size N in correspondence with N = 2 m . Furthermore, the proposed architecture is easily controlled through binary counter control signals. The presented architectures have been designed with Xilinx System Generator, realized and evaluated on a Virtex-5 FPGA XC5VSX240T-2FF1738 device.

Some aspects of marine electric power plants load registration and identification have been presented. Methods of operating data collecting about marine electric power plant loads have been shown based on an example of three vessels - a chemical tanker, a container ship and a tanker. Power plants of these vessels have been described. Examples of collected operating data were presented. Range of marine electric power plant load identification research has been characterized, which presented in the paper results are a part of.

This paper presents a predictive current control strategy with active damping method applied to a current source inverter. This control strategy uses a discrete-time model of the system to predict the future value of the grid current for all possible current vectors generated by the inverter. According to a cost function that minimizes the errors of the predicted grid current and the reference grid current, the current vector at the next sampling time is determined. To suppress the LC resonance in the current source inverter, active damping method using virtual harmonic resistor is adopted. The performance of the proposed control method is compared with the conventional trilogic pulse width modulation control. Simulation results show that the predictive current control with active damping method has an excellent steady-state response as well as an extremely fast dynamic response compared with the conventional method.

Models of quantum systems on curved space-times lack sufficient experimental verification. Some speculative theories suggest that quantum properties, such as entanglement, may exhibit entirely different behavior to purely classical systems. By measuring this effect or lack thereof, we can test the hypotheses behind several such models. For instance, as predicted by Ralph and coworkers [T C Ralph, G J Milburn, and T Downes, Phys. Rev. A, 79(2):22121, 2009;T C Ralph and J Pienaar, New Journal of Physics, 16(8):85008, 2014], a bipartite entangled system could decohere if each particle traversed through a different gravitational field gradient. We propose to study this effect in a ground to space uplink scenario. We extend the above theoretical predictions of Ralph and coworkers and discuss the scientific consequences of detecting/failing to detect the predicted gravitational decoherence. We present a detailed mission design of the European Space Agency's (ESA) Space QUEST (Space - Quantum Entanglement Space Test) mission, and study the feasibility of the mission scheme. Urea selective catalytic reduction(urea-SCR) control is very important for diesel engine emissions. In this paper, a Continuous Stirred Tank Reactor(CSTR) is used to represent the SCR model, then a cascade control system is deduced based on active disturbance rejection control(ADRC) strategy. The cascade control system consists of two ADRC controllers, and every ADRC controller contains a Extended State Observer(ESO) to estimate the total disturbance and a PID feedback control law to handle the ammonia coverage ratio tracking. Finally, the simulation results are given to demonstrate the effectiveness of the proposed control scheme tracking the reference and the robustness to the noise.

The primary objective is to investigate the applicability of spectral methods for designing feedback controllers for a closed loop system with an input shaper with time delays. The shaper is included in the feedback loop in order to pre-compensate low-damped oscillatory modes of a flexible subsystem. However, after inserting a shaper into the feedback loop, the closed loop dynamics becomes infinite dimensional. A robust controller design then requires guaranteeing that all the infinitely many poles of the closed loop system will safely be located in the left half of the complex plane. Besides, the closed loop dynamics need to be sufficiently fast in order to take over and keep filtering properties of the input shaper. For the spectral design of the controller, the recently developed spectral abscissa minimization approach for interconnected time delay systems is applied. The presented methods are tested on a case study simulation example, which is a multi-degree of freedom mechanical system. © 2015, IFAC (International Federation of automatic control) Hosting by Elsevier Ltd. In this paper, we provide a compositional framework for the construction of infinite approximations of interconnected stochastic control systems. Our approach is based on a notion of so-called stochastic simulation functions that are associated with interfaces. The stochastic simulation functions are used to quantify the approximation error while the interfaces are used to lift the controllers synthesized for the approximation to the controllers for the original stochastic system. In the first part of the paper, we analyze interconnected stochastic control systems which consist of several stochastic control subsystems. We derive sufficient conditions that facilitate the compositional construction of stochastic simulation functions together with the associated interfaces. Specifically, we show how to construct a stochastic simulation function with the corresponding interface for the interconnected stochastic control system from the simulation functions and interfaces of the individual stochastic control subsystems. In the second part of the paper, we focus on linear stochastic control systems. We extend a methodology, which is known for the non-probabilistic case, to construct infinite approximations of linear stochastic control systems together with their stochastic simulation functions and the corresponding interfaces. Finally, we illustrate the effectiveness of the proposed results on the interconnection of four linear stochastic control subsystems.

Large-scale interconnected uncertain systems commonly have large state and uncertainty dimensions. Aside from the heavy computational cost of performing robust stability analysis in a centralized manner, privacy requirements in the network can also introduce further issues. In this paper, we utilize IQC analysis for analyzing large-scale interconnected uncertain systems and we evade these issues by describing a decomposition scheme that is based on the interconnection structure of the system. This scheme is based on the so-called chordal decomposition and does not add any conservativeness to the analysis approach. The decomposed problem can be solved using distributed computational algorithms without the need for a centralized computational unit. We further discuss the merits of the proposed analysis approach using a numerical experiment. © IFAC.
