In order to enhance the glutathione peroxidase(GPX) catalytic activity of the selenium-containing single-chain variable fragments(Se-scFv), a novel human scFv was designed on the basis of the structure of human antibody and optimized via bioinformatics methods such as homologous sequence analysis, three-dimensional(3D) model building, binding-site analysis and docking. The DNA sequence of the new human scFv was synthesized and cloned into the expression vector pET22b(+), then the scFv protein was expressed in soluble form in Escherichia coli BL21(DE3) and purified by Ni2+-immobilized metal affinity chromatography(IMAC). The serine residue of scFv in the active site was converted into selenocysteine(Sec) with the chemical modification method, thus, the human Se-scFv with GPX activity was obtained. The GPX activity of the Se-scFv protein was characterized. Compared with other Se-scFv, the new human Se-scFv showed similar efficiency for catalyzing the reduction of hydrogen peroxide by glutathione. It exhibited pH and temperature dependent catalytic activity and a typical ping-pong kinetic mechanism.

Structural damage may weaken the aircraft and lead to loss of control, which is the leading cause of aircraft fatal accidents in the world. Meanwhile, advanced configuration aircraft is more complex than traditional aircraft and the controlling is more difficult. Hence, based on the modeling of structural damaged aircraft, firstly, the aerodynamic characteristics are analyzed according to trim, linearization, stick-fixed response and disturbance simulation. The results show that the damage causes offset of center of gravity and the pitch-roll, pitch-yaw coupling, which would make the emergency more complex. Secondly, the remaining flight performance is calculated by inducing the Remaining Capability Factor, which indicates that the remaining flight performance is reduced due to the damage. The results of above analysis should be considered in the future controller design.

The demand of hydrogen in oil refinery is increasing as market forces and environmental legislation, so hydrogen network management is becoming increasingly important in refineries. Most studies focused on single-objective optimization problem for the hydrogen network, but few account for the multi-objective optimization problem. This paper presents a novel approach for modeling and multi-objective optimization for hydrogen network in refineries. An improved multi-objective optimization model is proposed based on the concept of superstructure. The optimization includes minimization of operating cost and minimization of investment cost of equipment. The proposed methodology for the multi-objective optimization of hydrogen network takes into account flow rate constraints, pressure constraints, purity constraints, impurity constraints, payback period, etc. The method considers all the feasible connections and subjects this to mixed-integer nonlinear programming (MINLP). A deterministic optimization method is applied to solve this multi-objective optimization problem. Finally, a real case study is intro-duced to illustrate the applicability of the approach.

Robust real-time tracking of non-rigid objects is a challenging task. Color is a powerful feature for tracking deformable objects in image sequences with complex backgrounds. Color distribution is applied, as it is robust to partial occlusion, is rotation and scale invariant and computationally efficient. Particle filter has been proven very successful for non-linear and non-Gaussian estimation tracking problems. The article presents the integration of color distributions into particle filtering. A target is tracked with a particle filter by comparing its histogram with the histograms of the sample positions using the Bhattacharyya distance. Additionally, to solve the sample impoverishment (all particles collapse to a single point within a few iterations) in the particle-filter algorithm, a new resampling algorithm is proposed to tackle sample impoverishment. The performance of the proposed filter is evaluated qualitatively on various real-world video sequences. The experimental results show that the improved color-based particle filter algorithm can reduce sample impoverishment effectively and track the moving object very well.
