Influence of different processing methods on anti-trypsin activity in bovine colostrum

Colostrum is important for supplying the bovine neonate with nutrients, bioactive substances, and immunoglobulins to acquire passive immunity. Trypsin inhibitory activity represents a general characteristic of the first bovine colostrum. It is assumed that the anti-trypsin activity serves to protect bioactive molecules in the colostrum from being digested by the calf. The objectives of the study were to establish a test to determine anti-trypsin activity and test the hypothesis that freezing, acidification, and heat treatment alter anti-trypsin activity compared with untreated bovine colostrum. A photometric assay was established to determine anti-trypsin activity. The activity was expressed in milligrams of inhibited trypsin per milliliter of colostrum. Anti-trypsin activity was measured in untreated colostrum, frozen colostrum (−20°C for 24 h), colostrum acidified using 10% formic acid in a way that it was present as 1% in the sample, and colostrum heat treated according to 2 different protocols (60°C for 60 min and 63.5°C for 30 min). In our study, trypsin inhibition in 40 untreated colostrum samples (0.80 mg/mL, SEM 0.03) corresponded to that observed in frozen colostrum samples (0.79 mg/mL, SEM 0.03). In these first analyzed 40 and 59 extra samples (in total 99 samples), the anti-trypsin activity of frozen colostrum (0.85 mg/mL, SEM 0.01) was compared with colostrum subjected to acidification (0.84 mg/mL, SEM 0.01), heat treatment at 60°C for 60 min (0.65 mg/mL, SEM 0.02), and heat treatment at 63.5°C for 30 min (0.61 mg/mL, SEM 0.02). Acidification did not significantly affect trypsin inhibition. Both heat treatment protocols significantly reduced anti-trypsin activity. In the future, when investigating the effects of postharvest storage and processing on bovine colostrum, the influence on anti-trypsin activity should be evaluated in addition to the effects on immunoglobulins and other components. © 2025

Sinbaglustat is a brain-penetrating small molecule that inhibits the non-lysosomal glucocerebrosidase (GBA2) and, with lower potency, glucosylceramide synthase (GCS). Sinbaglustat has passed clinical phase I. Our preclinical study assessed its efficacy in a transgenic mouse model of G_M1 gangliosidosis, lacking a functional β-galactosidase enzyme (Glb1^−/−). Starting at 4 weeks of age, mice were either treated with a nominal dose of 10 or 300 mg/kg/day of sinbaglustat or remained untreated. Wild-type (WT) mice served as control. Body weight, clinical and neurological signs, and motor function was assessed until 17–18 weeks (4 months) and 30 weeks (7 months) of age when mice were euthanized for ex vivo assessments. In comparison to WT, Glb1^−/− mice showed the expected accumulation of G_M1 gangliosidosis-related sphingolipids, neuropathology, and behavioral deficits. Both dosages of sinbaglustat left G_M1 and lyso G_M1 levels in the brain unaffected but delayed the onset of motor impairment and progression of clinical disease in Glb1^−/− mice with the higher dose being more efficacious. Histologically and immunohistochemically, both treatment groups of Glb1^−/− mice displayed reduced neuronal vacuolation. Only the higher dose of sinbaglustat decreased axonal damage and astrogliosis, which was also associated with a decrease of the axonal/neuronal damage marker plasma neurofilament light at 4 months (17–18 weeks). Both doses of sinbaglustat increased the GBA2 substrate glucosylceramide (GluCer) in the brain, while only the high dose reduced GluCer and other glycosphingolipids (GSLs) in the periphery indicating additional inhibition of GCS. We conclude that sinbaglustat had a therapeutic-like effect in the G_M1 gangliosidosis mouse model. © 2024

Platelet autoantibodies have an impact on the platelet count in patients

Background: It is widely accepted that autoantibodies directed against platelet glycoproteins (GPs) are a major pathophysiological mechanism in immune thrombocytopenia, but little clinical data are available demonstrating an association between platelet antibodies and platelet counts. Objectives: We hypothesized that if platelet antibodies are clinically relevant, the number of targeted GPs and antibody concentration should be associated with the extent of thrombocytopenia. Methods: Platelet antibodies were identified in a direct GP-specific test that detects antibodies against GPIIb/IIIa and GPIb/IX. Using laboratory data from 12 335 thrombocytopenic patients with and without GP-specific platelet antibodies, we conducted a large retrospective cohort study. Results: We identified 1469 adults with GP-specific platelet antibodies in our database with complete entries. Compared with 10 866 adults without antibodies, patients with antibodies had significantly lower median platelet counts (54 G/L [IQR, 29-89] vs 85 G/L [IQR, 52-123], P < .0001). Patients with antibodies against 2 GPs had significantly lower platelet counts than patients with antibodies against 1 GP (47 G/L [IQR, 26-81] vs 62 G/L [IQR, 32-99], P < .0001 for GPIIb/IIIa and 58 G/L [IQR, 32-99], P = .0004 for GPIb/IX). Increasing antibody levels correlated with decreasing platelet counts for anti-GPIIb/IIIa (R² = .69;rho −0.84) and anti-GPIb/IX (R² = .57;rho −0.6). Conclusion: The presence of autoantibodies against GPIIb/IIIa or GPIb/IX is associated with lower platelet counts. More GPs targeted by autoantibodies and increasing antibody levels predict lower platelet counts. Platelet antibodies appear to be of clinical relevance. © 2025 The Author(s)

Clinical 'electrical response audiometry' on a general-purpose computer (IBM 1800)

A computer programme permitting the use of a general-purpose computer (IBM 1800) in evoked response audiometry has been developed. A DC coupling of the computer is obtained by digital parallel displacement of the sampling sweeps. EEG amplitudes (RMS) are calculated before and after stimulation. The V potentials in babies are facilitated by this DC coupling. Titanium (Ti) implants are widely used for tooth replacement due to their exceptional mechanical properties and high biocompatibility. However, their inherently inert surface limits osteogenic potential and makes them prone to bacterial colonization, increasing the risk of biofilm formation and implant-related infections. To address these limitations, surface modification of Ti is essential. This study aimed to enhance the surface properties of Ti by coating it with polydopamine (PDA) and further doping it with copper and calcium ions. TPDA was prepared and subsequently used to fabricate TPDA@Cu and TPDA@CuCa samples. Material characterization confirmed that TPDA@CuCa exhibited excellent surface wettability and biocompatibility, with Cu2+ and Ca2+ being continuously and stably released in liquid environments. Additionally, TPDA@CuCa significantly improved protein adsorption, facilitating favorable cellular interactions. In vitro experiments demonstrated that TPDA@CuCa exhibited strong antimicrobial activity against Escherichia coli and Staphylococcus aureus, enhanced osteoblast adhesion, mineralization, and upregulated osteogenic gene expression. This bifunctional surface modification strategy offers a promising approach to enhancing both the osteogenic and antibacterial properties of Ti implants. © 2025, The Authors. All rights reserved.

High-quality electrocardiogram (ECG) signals play a pivotal role in many heart-related healthcare applications. However, various types of real-world noises can contaminate the ECG signal during acquisition, thereby impairing its practical applicability. Thus, it is of great significance to develop an effective method for noise filtering in contaminated ECG signals. Most existing techniques predominantly focus on reducing noise in contaminated ECG signals in time domain, overlooking the potential benefits of time-frequency analysis. In this study, we propose a magnitude-aware deep learning framework with phase retrieval for denoising contaminated ECG signals (ECGDenoiser) based on time-frequency domain analysis. Firstly, the corrupted magnitude spectrum and phase spectrum are obtained by applying the short-time Fourier transform (STFT) to the contaminated ECG signal. Secondly, the ConvNeXtV2 Attention UNet is proposed to separate noise magnitude spectrum from the corrupted spectrum, and a fast Griffin-Lim algorithm is introduced to reconstruct the phase spectrum. Finally, the inverse STFT is adopted to recover the signal through estimated ECG magnitude spectrum and enhanced phase spectrum. The denoising performance of the proposed method is denoted by root mean square error (RMSE) and signal-to-noise ratio (SNR). Experiments conducted on single-channel and multi-channel ECG datasets demonstrate that ECGDenoiser surpasses conventional filtering techniques and deep learning-based methods in addressing seven distinct categories of noise interference. SALPINGITIS AND CERVICITIS IN URO-ARTHRITIS

To compare anamnestic cervicitis and salpingitis as a relative risk for uro-arthritis the medical history of 60 consecutive females with uro-arthritis was compared with that of 311 interviewed controls. Salpingitis was revealed to be a relative risk factor of 3.2 as compared to cervicitis 1.4 in connection with uro-arthritis. HLA-B27 was tested in the 60 patients and in an additional 7 females with uro-arthritis, all of whom had a history of salpingitis. HLA-B27 was positive in 42% (28/67). There were no significant differences between those with or without cervicitis or salpingitis or both. Salpingitis could thus as justifiably be accepted as a diagnostic criterion for RS as cervicitis. These findings and clinical observations, including comprehensive histories of the patients concerned, serve as a basis for our hypothesis of rheumatic salpingitis. Further prospective and co-operative investigations will be needed to confirm or disprove this hypothesis.

A program using acoustically evoked neurophysiological responses recorded by signal averaging is presented. The system, also using an adaptive digital filtering, is now used in the audiology clinic for investigation of the auditory system without active cooperation of the patient. The present paper will first give the outline of the hospital computer's time sharing system, and next a detailed description of the audiometry system. In adddition to on-line generated displays, the system, in batch, can produce "hardcopy" documentation of all data contained in a session. © 1971.

The spatial ventricular gradient during alterations in the ventricular activation pathway

The spatial ventricular gradient was obtained from 71 ECG records exhibiting both normal sinus beats (NSB) and premature ventricular contractions (PVC). The difference in gradients between normal and extra beats was compared statistically with beat-to-beat variability of normal sinus rhythm. A significant difference was found, and Wilson's hypothesis on the constancy of the ventricular gradient in spite of changes in the ventricular activation pathway could be rejected. Two different corrected orthogonal lead systems were used in order to minimize the effect that a particular lead design might have had upon results. Methods for obtaining ECG time integrals were studied for accuracy and consistency. In planimetric determinations the width of records was found to influence results considerably. Digital or analog computations were found to be a preferable procedure for quantitative applications. Furthermore, the averaging of time integrals over at least one respiratory cycle was found to be necessary in order to eliminate beat-to-beat variability of the ventricular gradient. Since methods for obtaining ECG time integrals were found to be very critical, the question was raised whether simpler ECG measurements, such as maximal QRS vectors, are not preferable for clinical use. © 1966.
