Although melanoma is notorious for its high degree of heterogeneity and plasticity1,2,the origin and magnitude of cell-state diversity remains poorly ***,it is unclear whether growth and metastatic dissemination are supported by overlapping or distinct melanoma ***,by combining mouse genetics,single-cell and spatial transcriptomics,lineage tracing and quantitative modelling,we provide evidence of a hierarchical model of tumour growth that mirrors the cellular and molecular logic underlying the cell-fate specification and differentiation of the embryonic neural *** show that tumorigenic competence is associated with a spatially localized perivascular niche,a phenotype acquired through an intercellular communication pathway established by endothelial *** with a model in which only a fraction of cells are fated to fuel growth,temporal single-cell tracing of a population of melanoma cells with a mesenchymal-like state revealed that these cells do not contribute to primary tumour growth but,instead,constitute a pool of metastatic initiating cells that switch cell identity while disseminating to secondary *** data provide a spatially and temporally resolved map of the diversity and trajectories of melanoma cell states and suggest that the ability to support growth and metastasis are limited to distinct pools of *** observation that these phenotypic competencies can be dynamically acquired after exposure to specific niche signals warrant the development of therapeutic strategies that interfere with the cancer cell reprogramming activity of such microenvironmental cues.

The field of the study was the development of a microprocessor device for contactless measurement of a liquid level using the impedance method. The result was a noncontact liquid level meter on the PSoC4 microcontroller in the form of a working model. The results of tests of the developed liquid level sensor on tap water, distilled water, AdBlue, and diesel fuel are presented. © 2019 IEEE.

Fundamental investigations in non-locality have shown that while the no-signaling principle alone is not sufficient to single out the set of quantum non-local correlations, local quantum mechanics and no-signaling together exactly reproduce the set of quantum correlations in the two-party Bell scenario. Here, we introduce and study an intermediate hybrid no-signaling quantum set of non-local correlations that we term HNSQ in the multi-party Bell scenario where some subsystems are locally quantum while the remaining subsystems are only constrained by the no-signaling principle. Specifically, the set HNSQ is a super-quantum set of correlations derived from no-signaling assemblages by performing quantum measurements on the trusted subsystems. As a tool for optimization over the set HNSQ, we introduce an outer hierarchy of semi-definite programming approximations to the set following an approach put forward by Doherty-Parillo-Spedalieri. We then show that in contrast to the set NS of no-signaling correlations, there exist extreme points of HNSQ in the tripartite Bell scenario that admit quantum realization. We perform an extensive numerical analysis of the maximal violation of the facet Bell inequalities in the three-party binary input-output scenario and study the corresponding self-testing properties. In contrast to the usual no-signaling correlations, the new set allows for simple security proofs of (one-sided)-device-independent applications against super-quantum adversaries. The current paper proposes a Galerkin method for calculating spectral characteristics of microcavity lasers with piercing holes. If boundaries of the cavity and holes are circles, then entries of the matrix have explicit expressions computed in the present work. Using the proposed algorithm, we calculate directivities, spectra, and thresholds of laser modes. Numerical experiments demonstrate that, by varying the location of the hole, we can increase the directivity, while the threshold gain stays low.

Regional Image Perturbation Reduces Lp Norms of Adversarial Examples While Maintaining Model-to-model Transferability

Regional adversarial attacks often rely on complicated methods for generating adversarial perturbations, making it hard to compare their efficacy against well-known attacks. In this study, we show that effective regional perturbations can be generated without resorting to complex methods. We develop a very simple regional adversarial perturbation attack method using cross-entropy sign, one of the most commonly used losses in adversarial machine learning. Our experiments on ImageNet with multiple models reveal that, on average, 76% of the generated adversarial examples maintain model-to-model transferability when the perturbation is applied to local image regions. Depending on the selected region, these localized adversarial examples require significantly less Lp norm distortion (for p ∈ {0, 2, ∞}) compared to their non-local counterparts. These localized attacks therefore have the potential to undermine defenses that claim robustness under the aforementioned norms. Copyright © 2020, The Authors. All rights reserved.

Background Systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), systemic sclerosis (SSC), Sjögren's syndrome (SJS), mixed connective tissue disease (MCTD), primary antiphospholipid syndrome (PAPS) and undifferentiated connective tissue disease (UCTD) are classified as systemic autoimmune diseases (SADs). They are diagnosed based on different clinical and laboratory criteria. Due to their high internal heterogeneity and overlapping symptoms, SADs are difficult to diagnose. Therefore, molecular and cellular-based studies need to be undertaken to precisely classify the patients. Mass cytometry is a single-cell proteomics technology that measures approximately 50 markers per cell, thus it is a suitable tool to perform deep-phenotyping studies in SADs. Objectives Explore differences and similarities between SADs and build reclassification framework using high-dimensional cytometry data. Methods The whole blood samples collected from 129 individuals, including patients and controls were stained with a 39-plex antibody panel and acquired in 9 batches on a CyTOF (HELIOS) instrument. Data were cleaned, and normalized for batch effects using semi-automated cytof analysis pipeline. Cell frequencies and median signal intensities (MSI) for each population were extracted using FlowSOM for mononuclear cells (PBMC) and Phenograph for granulocytes. Secretion of 44 cytokines and chemokines were analyzed using a multiplexed luminex assay. Diseases were compared by Kruskal-Wallis analysis and hierarchical clustering and reclassification was done using unsupervised k-means clustering. Cytokine analysis across clusters was performed using Kruskal-Wallis test. Results Differently expressed features were observed between patient groups, regarding frequency of classical monocytes, B and T cells subpopulations, mature and immature granulocytes and intensities of CD38, HLA-DR and CD95 across various populations. The term Research Software Engineer, or RSE, emerged a little over 10 years ago as a way to represent individuals working in the research community but focusing on software development. The term has been widely adopted and there are a number of high-level definitions of what an RSE is. However, the roles of RSEs vary depending on the institutional context they work in. At one end of the spectrum, RSE roles may look similar to a traditional research role. At the other extreme, they resemble that of a software engineer in industry. Most RSE roles inhabit the space between these two extremes. Therefore, providing a straightforward, comprehensive definition of what an RSE does and what experience, skills and competencies are required to become one is challenging. In this community paper we define the broad notion of what an RSE is, explore the different types of work they undertake, and define a list of foundational competencies as well as values that outline the general profile of an RSE. Further research and training can build upon this foundation of skills and focus on various aspects in greater detail. We expect that graduates and practitioners will have a larger and more diverse set of skills than outlined here. On this basis, we elaborate on the progression of these skills along different dimensions. We look at specific types of RSE roles, propose recommendations for organisations, and give examples of future specialisations. Metal (Cu) and semiconductor (Cu 2 ZnSnS 4 ) nanoparticles were successfully synthesized by the chemical methods. Morphological and structural properties of nanoparticles were investigated. Synthesized nanoparticles were used for development of nanoinks, which are promising for 3D printing of the electronic circuit components and solar cells.

It is known that if the boundary of a planar domain is smooth enough, it has eigenvalues converging to two different points determined by Lamé parameters. We show that eigenvalues converge at a polynomial rate on smooth boundaries and the convergence rate is determined by smoothness of the boundary.
