Objective: Osteoarthritis (OA) is a chronic, degenerative disease of the articular joints. The disease presents an enormous clinical and economic burden globally, due in part to the lack of disease modifying therapies. For over a decade, OA researchers have been working to determine epigenetic mechanisms underlying the disease to better understand pathology, identify biomarkers of progression, and pinpoint novel targets for therapeutic intervention. Design: This article presents a summary of the 3rd International Workshop on the Epigenetics of Osteoarthritis held in Toronto, Ontario, Canada, in September 2024. The purpose of this meeting was to gather the international community to discuss the status of OA epigenetic research and share expertise on innovative techniques for future. Results: Since the two previous meetings, there has been increasing adoption of advanced single-cell and spatial sequencing technologies and bioinfomatic analyses. Furthermore, investigations of multiple joint tissues has highlighted the shifting paradigm from OA as a cartilage centric disease to the consideration of all joint tissues. Conclusions: The workshop provided a unique opportunity for early-career researchers to expand their network, and for all participants to discuss new or improved approaches to advance the field, including international consortia and data sharing. The highlights and outcomes from this OA epigenetics workshop are described in this report. © 2025 The Authors

The optical vortex has recently attracted scholars to implement it in optical tweezers, microscopy, optical communications, quantum information processing, optical trapping, and laser machining. Optical vortex beam applied classically that can be transferred to the transverse amplitude of a heralded single-photon, and optical vortex possesses a helical wavefront and carries orbital angular momentum. In this study, Optical vortex is applied in optical-CDMA (optical code-division multiple-access) in conjunction with WDM (wavelength division multiplexing). This mechanism aims to increase the capacity and security in optical communication significantly. The implementation of Laguerre-Gaussian (LG) modes with optical vortex based on one dimension zero cross-correlation (ZCC) code shows that mode coupling reduces effectively. Consequently, a positive increase in channel performance and response. Accordingly, the LG modes based on the 1D-ZCC code are investigated and propagated over multi-mode fiber (MMF) based on an optical vortex, which also substantially reduces channel effects. Consequently, all these attributes combined will result in a hybrid WDM-Optical-CDMA with an optical vortex system over MMF.

The Brain Imaging Data Structure (BIDS) is a community-driven standard for the organization of data and metadata from a growing range of neuroscience modalities. This paper is meant as a history of how the standard has developed and grown over time. We outline the principles behind the project, the mechanisms by which it has been extended, and some of the challenges being addressed as it evolves. We also discuss the lessons learned through the project, with the aim of enabling researchers in other domains to learn from the success of BIDS. © 2023, CC BY.

Small-animal diffusion MRI (dMRI) has been used for methodological development and validation, characterizing the biological basis of diffusion phenomena, and comparative anatomy. The steps from animal setup and monitoring, to acquisition, analysis, and interpretation are complex, with many decisions that may ultimately affect what questions can be answered using the resultant data. This work aims to present selected recommendations and guidelines from the diffusion community, on best practices for preclinical dMRI of in vivo animals. We describe the general considerations and foundational knowledge that must be considered when designing experiments. We briefly describe differences in animal species and disease models and discuss why some may be more or less appropriate for different studies. We then give guidelines for in vivo acquisition protocols, including decisions on hardware, animal preparation, and imaging sequences, followed by advice for data processing including pre-processing, model-fitting, and tractography. Finally, we provide an online resource which lists publicly available preclinical dMRI datasets and software packages, to promote responsible and reproducible research. In each section, we attempt to provide guides and recommendations, but also highlight areas for which no guidelines exist (and why), and where future work should focus. While we mainly cover the central nervous system (on which most preclinical dMRI studies are focused), we also provide, where possible and applicable, recommendations for other organs of interest. An overarching goal herein is to enhance the rigor and reproducibility of small animal dMRI acquisitions and analyses, and thereby advance biomedical knowledge. © 2022, CC BY-NC-ND.

Infants learn the sound categories of their language and adults successfully process the sounds they hear, even though sound categories often overlap in their acoustics. Most researchers agree that listeners use context to disambiguate overlapping categories. However, they differ in their ideas about how context is used. One idea is that listeners normalize out the systematic effects of context from the acoustics of a sound. Another idea is that contextual information may itself be an informative cue to category membership, due to patterns in the types of contexts that particular sounds occur in. We directly contrast these two ways of using context by applying each one to the test case of Japanese vowel length. We find that normalizing out contextual variability from the acoustics does not improve categorization, but using context in a top-down fashion does so substantially. This reveals a limitation of normalization in phonetic acquisition and processing and suggests that approaches that make use of top-down contextual information are promising to pursue. © 2018 Proceedings of the 40th Annual Meeting of the Cognitive science Society, CogSci 2018. The emergence of SARS-CoV in 2002 and SARS-CoV-2 in 2019 led to increased sampling of sarbecoviruses circulating in horseshoe bats. Employing phylogenetic inference while accounting for recombination of bat sarbecoviruses, we find that the closest-inferred bat virus ancestors of SARS-CoV and SARS-CoV-2 existed less than a decade prior to their emergence in humans. Phylogeographic analyses show bat sarbecoviruses traveled at rates approximating their horseshoe bat hosts and circulated in Asia for millennia. We find that the direct ancestors of SARS-CoV and SARS-CoV-2 are unlikely to have reached their respective sites of emergence via dispersal in the bat reservoir alone, supporting interactions with intermediate hosts through wildlife trade playing a role in zoonotic spillover. Summary Background Across low-income and middle-income countries (LMICs), one in ten deaths in children younger than 5 years is attributable to diarrhoea. The substantial between-country variation in both diarrhoea incidence and mortality is attributable to interventions that protect children, prevent infection, and treat disease. Identifying subnational regions with the highest burden and mapping associated risk factors can aid in reducing preventable childhood *** We used Bayesian model-based geostatistics and a geolocated dataset comprising 15 072 746 children younger than 5 years from 466 surveys in 94 LMICs, in combination with findings of the Global Burden of Diseases, Injuries, and Risk Factors Study (GBD) 2017, to estimate posterior distributions of diarrhoea prevalence, incidence, and mortality from 2000 to 2017. From these data, we estimated the burden of diarrhoea at varying subnational levels (termed units) by spatially aggregating draws, and we investigated the drivers of subnational patterns by creating aggregated risk factor *** The greatest declines in diarrhoeal mortality were seen in south and southeast Asia and South America, where 54·0% (95% uncertainty interval [UI] 38·1-65·8), 17·4% (7·7-28·4), and 59·5% (34·2-86·9) of units, respectively, recorded decreases in deaths from diarrhoea greater than 10%. Although children in much of Africa remain at high risk of death due to diarrhoea, regions with the most deaths were outside Africa, with the highest mortality units located in Pakistan. Indonesia showed the greatest within-country geographical inequality; some regions had mortality rates nearly four times the average country rate. Reductions in mortality were correlated to improvements in water, sanitation, and hygiene (WASH) or reductions in child growth failure (CGF). This study investigates the potential of deep learning methods to identify individuals with suspected COVID-19 infection using remotely collected heart-rate data. The study utilises data from the ongoing EU IMI RADAR-CNS research project that is investigating the feasibility of wearable devices and smartphones to monitor individuals with multiple sclerosis (MS), depression or epilepsy. As part of the project protocol, heart-rate data was collected from participants using a Fitbit wristband. The presence of COVID-19 in the cohort in this work was either confirmed through a positive swab test, or inferred through the selfreporting of a combination of symptoms including fever, respiratory symptoms, loss of smell or taste, tiredness and gastrointestinal symptoms. Experimental results indicate that our proposed contrastive convolutional auto-encoder (contrastive CAE), i. e., a combined architecture of an auto-encoder and contrastive loss, outperforms a conventional convolutional neural network (CNN), as well as a convolutional auto-encoder (CAE) without using contrastive loss. Our final contrastive CAE achieves 95:3% unweighted average recall, 86:4% precision, an F1 measure of 88:2%, a sensitivity of 100% and a specificity of 90:6% on a test set of 19 participants with MS who reported symptoms of COVID-19.
