Background: Age-related disparities in outcomes for pediatric and adolescent leukemia are multifactorial. While the prevalence of pre-existing medical morbidities is low in children and adolescents, cancer-associated organ dysfunction leading to increased acuity of illness at initial presentation has been noted in up to 24% of those with pediatric leukemia. We have noted an association between adolescent and young adult (AYA) age group and acuity of illness at initial presentation (Jain et al, Blood , 2023). The current study aims to examine (1) the association between acuity of illness and overall survival, and (2) whether acuity of illness mediates age-related differences in overall survival. Methods: We conducted a retrospective analysis of 688 patients aged 1-21 years who presented to Children's Healthcare of Atlanta between 2010-2018 with any newly diagnosed leukemia, including B-cell acute lymphoblastic leukemia (ALL), T-cell ALL, acute myeloid leukemia, and other leukemias (chronic myeloid leukemia, juvenile myelomonocytic leukemia, Burkitt's leukemia). We excluded patients who lacked information in the first 72 hours following initial presentation to the hospital due to incomplete records or initial treatment at another facility. Based on manual chart review, high acuity of illness (yes/no) was defined as any ICU admission or ICU-level resource utilization by organ system (i.e., cardiovascular, respiratory, renal, hematologic, and neurologic), regardless of physical location in the hospital. We assessed overall survival with Kaplan-Meier analysis, with censoring defined by date at last follow-up, 5 years post-diagnosis, or December 31, 2020, whichever came first; the analysis was stratified by acuity of illness (high versus low) at initial presentation. Objectives Although previous epidemiological studies have reported the effects of fetal exposure to phthalates and phenols on birth outcomes, evidence is limited. The objective of this study was to investigate whether prenatal exposure to endocrine-disrupting chemicals [EDCs;phthalates and bisphenol A (BPA)] is associated with birth outcomes and whether there are sex-specific *** We used data from the Korean CHildren’s ENvironmental health Study (Ko-CHENS) cohort. Exposure to phthalates and BPA was measured by urine tests, and the information on outcomes and confounders was obtained from delivery records, biomarker assessment, and a self-reported questionnaire. We performed multivariate logistic regression to estimate the association between exposure to individual EDCs and each birth outcome. Additionally, to evaluate the effects of EDC mixture exposure, we performed weighted quantile sum regression analysis. Subgroup analysis stratified by maternal age and sex at birth was performed to examine potential differences in *** This study included 2,176 mother-child pairs from the Ko-CHENS. We observed a significant association between the risk of PTB and exposure to BPA during early pregnancy and mono-carboxyoctyl phthalate (MCOP) during late pregnancy, with odds ratios of 1.23 (95% CI: 1.01, 1.49) and 1.29 (95% CI: 1.03, 1.62), respectively. Additionally, there was a positive association between exposure to MCOP during early pregnancy and the risk of LBW (OR: 1.39, 95% CI: 1.03, 1.89). For male infants, exposure to MCOP during early pregnancy was associated with the risk of LBW (OR: 2.44, 95% CI: 1.43, 4.15), and exposure to MCOP during late pregnancy was associated with the risk of PTB (OR 1.45, 95% CI: 1.05, 2.02).Conclusions This study presents quantitative evidence for the association between prenatal exposure to EDCs, especially BPA and MCOP, and PTB/LBW. Association of long non-coding RNA HOTAIR and MALAT1 variants with cervical cancer risk in Han Chinese women

Long noncoding RNA(lncRNA) HOTAIR and MALAT1 are implicated in the development of multiple cancers. Genetic variants within HOTAIR and MALAT1 may affect the gene expression, thereby modifying genetic susceptibility to cervical cancer. A case-control study was designed, including 1 486 cervical cancer patients and 1 536 healthy controls. Based on RegulomeDB database, 11 SNPs were selected and genotyped by using Sequenom’s Mass ARRAY. Univariate and multivariate logistic regression models were used to calculate the odds ratio(OR) and 95% confidence interval(CI). We found that the A allele of rs35643724 in HOTAIR was associated with increased risk of cervical cancer, while the C allele of rs1787666 in MALAT1 was associated with decreased risk. Compared to individuals with 0–1 unfavorable allele, those with 3–4 unfavorable alleles showed18% increased odds of having cervical cancer. Our findings suggest that HOTAIR rs35643724 and MALAT1 rs1787666 might represent potential biomarkers for cervical cancer susceptibility.
