Background:Screening using low-dose computed tomography(LDCT)is a more effective approach and has the potential to detect lung cancer more *** aimed to conduct a meta-analysis to estimate the accuracy of population-based screening studies primarily assessing baseline LDCT screening for lung ***:MEDLINE,Excerpta Medica Database,and Web of Science were searched for articles published up to April 10,*** to the inclusion and exclusion criteria,the data of true positives,false-positives,false negatives,and true negatives in the screening test were *** Assessment of Diagnostic Accuracy Studies-2 was used to evaluate the quality of the literature.A bivariate random effects model was used to estimate pooled sensitivity and *** area under the curve(AUC)was calculated by using hierarchical summary receiver-operating characteristics *** between studies was measured using the Higgins I 2 statistic,and publication bias was evaluated using a Deeks’funnel plot and linear regression ***:A total of 49 studies with 157,762 individuals were identified for the final qualitative synthesis;most of them were from Europe and America(38 studies),ten were from Asia,and one was from *** recruitment period was 1992 to 2018,and most of the subjects were 40 to 75 years *** analysis showed that the AUC of lung cancer screening by LDCT was 0.98(95%CI:0.96-0.99),and the overall sensitivity and specificity were 0.97(95%CI:0.94-0.98)and 0.87(95%CI:0.82-0.91),*** funnel plot and test results showed that there was no significant publication bias among the included ***:Baseline LDCT has high sensitivity and specificity as a screening technique for lung ***,long-term follow-up of the whole study population(including those with a negative baseline screening result)should be performed to enhance the accuracy of LDCT screening.

Time-dependent Predictive Accuracy Metrics in the Context of Interval Censoring and Competing Risks

Evaluating the performance of a prediction model is a common task in medical statistics. Standard accuracy metrics require the observation of the true outcomes. This is typically not possible in the setting with time-to-event outcomes due to censoring. Interval censoring, the presence of time-varying covariates, and competing risks present additional challenges in obtaining those accuracy metrics. In this study, we propose two methods to deal with interval censoring in a time-varying competing risk setting: a model-based approach and the inverse probability of censoring weighting (IPCW) approach, focusing on three key time-dependent metrics: area under the receiver-operating characteristic curve (AUC), Brier score, and expected predictive cross-entropy (EPCE). The evaluation is conducted over a medically relevant time interval of interest, [t,∆t). The model-based approach includes all subjects in the risk set, using their predicted risks to contribute to the accuracy metrics. In contrast, the IPCW approach only considers the subset of subjects who are known to be event-free or experience the event within the interval of interest. we performed a simulation study to compare the performance of the two approaches with regard to the three metrics. © 2025, CC BY-NC-ND.

Background:Biliary tract carcinomas(BTCs)are relatively rare but lethal primary malignant tumors derived from the biliary tract *** burden of BTCs varies according to sex,age,region,and country,but limited attention has been paid to the burden of *** sought to explore the up-to-date data from the Global Burden of Disease Study(GBD)and expand findings by accessing the demographic features of BTC disease ***:Using the latest data from the GBD 2021,we evaluated and analyzed the distributions and patterns of BTC disease burden in various age groups,sexes,regions,and ***:The number of incident cases,deaths,and disability-adjusted life-years(DALYs)tended to increase and peaked at 216,770(95%uncertainty interval[UI]:181,890-245,240),171,960(95%UI:142,350-194,240),and 3,732,100(95%UI:3,102,900-4,317,000)person-years,respectively,in ***,the average global age-standardized rates(ASRs)of incident cases,deaths,and DALYs shrunk by−11.46%(95%UI:−21.91 to 3.35%),−24.09%(95%UI:−33.19 to 16.88%),and−26.25%(95%UI:−35.53 to 18.36%),respectively,from 1990 to ***,the male/female ratio(male per 100 female)of incidence,deaths,and DALYs changed from 76.40,75.41,and 74.72 to 86.89,79.11,and 82.29,*** 2021,the highest number of incident cases,deaths,and DALYs occurred in East *** top three highest incidences,deaths,and DALYs were observed in China,India,and Japan,and the highest ASRs were observed in Chile in *** of the Human Development Index along with disease burden estimates of BTCs also suggests that the burden of the disease is related to the level of comprehensive development of the ***:This study provided a comprehensive comparison of differences in the burden of disease across populations and over time,and further presented evidence concerning the formulation of prevention and control policies and etiologic studies for BTCs and proposed logical hypotheses to investigate.

Background: Precision medicine has become a mainstay of cancer care in recent years. The National cancer Institute (NCI) Surveillance, epidemiology, and End Results (SEER) program has been an authoritative source of cancer statistics and data since 1973. However, tumor genomic information has not been adequately captured in the cancer surveillance data, which impedes population-based research on molecular subtypes. To address this, the SEER program has developed and implemented a centralized process to link SEER registries' tumor cases with genomic test results that are provided by molecular laboratories to the registries. Methods: Data linkages were carried out following operating procedures for centralized linkages established by the SEER program. The linkages used Match∗Pro, a probabilistic linkage software, and were facilitated by the registries' trusted third party (an honest broker). The SEER registries provide to NCI limited datasets that undergo preliminary evaluation prior to their release to the research community. Results: Recently conducted genomic linkages included OncotypeDX Breast Recurrence Score, OncotypeDX Breast Ductal Carcinoma in Situ, OncotypeDX Genomic Prostate Score, Decipher Prostate Genomic Classifier, DecisionDX Uveal Melanoma, DecisionDX Preferentially Expressed Antigen in Melanoma, DecisionDX Melanoma, and germline tests results in Georgia and California SEER registries. Conclusions: The linkages of cancer cases from SEER registries with genomic test results obtained from molecular laboratories offer an effective approach for data collection in cancer surveillance. By providing de-identified data to the research community, the NCI's SEER program enables scientists to investigate numerous research inquiries. © 2024 The Author(s). Cervical cancer is one of the leading causes of cancer death among females worldwide and its behavior epidemiologically likes a venereal disease of low infectiousness. Early age at first intercourse and multiple sexual partners have been shown to exert strong effects on risk. The wide differences in the incidence among different countries also influenced by the introduction of screening. Although the general picture remains one of decreasing incidence and mortality, there are signs of an increasing cervical cancer risk probably due to changes in sexual behavior. Smoking and human papillomavirus(HPV) 16/18 are currently important issues in a concept of multifactorial, stepwise carcinogenesis at the cervix uteri. Therefore, society-based preventive and control measures, screening activities and HPV vaccination are recommended. Cervical cancer screening methods have evolved from cell morphology observation to molecular testing. High-risk HPV genotyping and liquid-based cytology are common methods which have been widely recommended and used worldwide. In future, accurate,cheap, fast and easy-to-use methods would be more popular. Artificial intelligence also shows to be promising in cervical cancer screening by integrating image recognition with big data technology. Meanwhile, China has achieved numerous breakthroughs in cervical cancer prevention and control which could be a great demonstration for other developing and resource-limited areas. In conclusion, although cervical cancer threatens female health, it could be the first cancer that would be eliminated by human beings with comprehensive preventive and control strategy.

Colorectal cancer (CRC) is a leading cause of mortality worldwide. We conducted a genome-wide association study meta-analysis of 100,204 CRC cases and 154,587 controls of European and east Asian ancestry, identifying 205 independent risk associations, of which 50 were unreported. We performed integrative genomic, transcriptomic and methylomic analyses across large bowel mucosa and other tissues. Transcriptome- and methylome-wide association studies revealed an additional 53 risk associations. We identified 155 high-confidence effector genes functionally linked to CRC risk, many of which had no previously established role in CRC. These have multiple different functions and specifically indicate that variation in normal colorectal homeostasis, proliferation, cell adhesion, migration, immunity and microbial interactions determines CRC risk. Crosstissue analyses indicated that over a third of effector genes most probably act outside the colonic mucosa. Our findings provide insights into colorectal oncogenesis and highlight potential targets across tissues for new CRC treatment and chemoprevention strategies.

A cercarial dermatitis outbreak occurred in Chana district, Songkhla Province, South Thailand, between August and October 2020. A total of 359 cases with cercarial dermatitis were confirmed with three cases of skin biopsy. The species of potential trematodes from infected snails were investigated, and the prevalence of infestation with schistosomes was described. As part of our ongoing studies of trematode diversity in freshwater systems, using morphological traits and sequence data to differentiate species, this study aimed to provide insights into the parasite species that cause cercarial dermatitis in the outbreak area and improve our understanding of parasite species distribution. Snail samples were collected in December 2020 and September and October 2021. Five main areas of outbreaks were investigated, and snails were collected by scooping and examined for infection with cercariae. The cercariae were characterized on the basis of morphological features. We found two species of snails to be infected, viz. Indoplanorbis exustus and Bithynia siamensis siamensis, with infection rates of 2.05% (12/586) and 7.93% (23/290), respectively. Three species of trematodes were found in B. s. siamensis, viz. Gastrothylax crumenifer, Astiotrema monticellii, and Loxogenes liberum. Moreover, three species of trematodes were found in Indoplanorbis exustus, viz. Clinostomum giganticum, Echinostoma spiniferum, and Schistosoma indicum. The latter is a ruminant schistosome, which causes the outbreak of cercarial dermatitis in the area. They were genetically analyzed using the internal transcribed spacer subunit II region to confirm the species identity at generic and infrageneric levels. Copyright Duangduen Krailas et al.
