Background Chronic synovial inflammation is a hallmark of rheumatoid arthritis (RA). During the long-term course of RA synovitis, chronic exposure of FLS to pro-inflammatory cytokines, such as TNF, induces a constellation of genes, which perpetuate synovial inflammation and provoke joint damage. Inflammation-induced gene expression evolves in a temporal order and involves two integrated processes: transcription and mRNA decay. Although the molecular mechanisms regulating transcription are well characterised, little is known about the impact of mRNA stability on gene expression and the genome-wide impact of chronic inflammation on decay rates of mRNA transcripts in RA FLS. Materials and methods Chronic synovitis was modelled ex vivo by long-term exposure of RA FLS to TNF. RNA-sequencing was performed, gene-transcripts were quantified, and DESeq2 was used for differential testing between TNF treated and control libraries. For pathway analysis, MSigDB and Panther were used. Differentially stable gene transcripts were identified between time points of TNF stimulation by specific statistical analytical tools. Results In our experimental system, long-term exposure of FLS to TNF induces a biphasic gene expression program. Initially, the inducible transcriptome consists primarily of unstable transcripts, but progressively switches and becomes dominated by very stable transcripts. This temporal switch is due to: a) TNF-induced prolonged stabilisation of previously unstable transcripts that enables progressive transcript accumulation over days and b) sustained expression and late induction of very stable transcripts. TNF-induced mRNA stabilisation in RA FLS occurs during the late phase of TNF response, is MAPK-dependent, and involves several genes with pathogenic potential, such as pro-inflammatory cytokines (eg, IL-6), a series of neutrophil-recruiting chemokines (eg, CXCL1, CXCL3, IL-8/CXCL8), CCL2, and PTGS2. In this paper we present a new computational primitive for convolution using coupled oscillator arrays. It is based on a Degree of Match (DOM) operation for pairs of vectors of analog voltage encoded data. The convolution operation is synthesized from these DOM circuits and computes the precise mathematical convolution of the two vectors. We present an example circuit design for the DOM operator and SPICE simulations of the circuit behavior. A MATLAB model is curve fit to an inverted copy of this output and shown to be roughly equivalent to the square of the Euclidian distance between the vectors. Using a parameterized version of this model we conducted a MATLAB study to analyze the accuracy of the DOM and convolution operations under variations in oscillator array symmetry, locking range, and additive noise. We also show a small example of convolution with an edge enhancement filter.

Managing UCONabc policies in modern distributed computing systems is a challenge for traditional approaches. The provisioning model has trouble to keep track and to synchronize large numbers of distributed policies, outsourcing model may suffer from network overhead and single point of failure. This paper describes an approach to manage distributed UCONabc policies, derived from the combination of authorization assertions and policy templates. It combines the benefits of provisioning and outsourcing, eliminating their respective drawbacks. Prototyping details and performance evaluation are shown, messages are 42.7% smaller than provisioning and response times are faster than outsourcing. © 2015 IEEE.

The nearby Sun-like star GJ 758 hosts a cold substellar companion, GJ 758 B, at a projected separation of 30 au, previously detected in high-contrast multi-band photometric observations. In order to better constrain the companion’s physical characteristics, we acquired the first low-resolution (R ∼ 50) near-infrared spectrum of it using the high-contrast hyperspectral imaging instrument Project 1640 on Palomar Observatory’s 5 m Hale telescope. We obtained simultaneous images in 32 wavelength channels covering the Y , J, and H bands (∼952–1770 nm), and used data processing techniques based on principal component analysis to efficiently subtract chromatic background speckle-noise. GJ 758 B was detected in four epochs during 2013 and 2014. Basic astrometric measurements confirm its apparent northwest trajectory relative to the primary star, with no clear signs of orbital curvature. Spectra of SpeX/IRTF observed T dwarfs were compared to the combined spectrum of GJ 758 B, with χ2 minimization suggesting a best fit for spectral type T7.0±1.0, but with a shallow minimum over T5–T8. Fitting of synthetic spectra from the BT-Settl13 model atmospheres gives an effective temperature Teff = 741 ± 25 K and surface gravity log g = 4.3 ± 0.5 dex (cgs). Our derived best-fit spectral type and effective temperature from modeling of the low-resolution spectrum suggest a slightly earlier and hotter companion than previous findings from photometric data, but do not rule out current results, and confirm GJ 758 B as one of the coolest sub-stellar companions to a Sun-like star to date. The task of designing WMSN real scenarios in order to evaluate the network performance is not trivial. This work sheds some light on this challenge by studying the impact of the load size when using ZigBee protocol in WMSN that transmits images. The main feature is to use real nodes for reading and dividing camera data into packages for transmission over the network. We have analyzed the number of packets transmitted by sensor node and the number of packets received by base station as a success parameter of the delivery. We have also analyzed data delay and data delivery for each load size: 64, 80 and 96 bytes. The results reveal that there is a trade-off between latency and success rate and that the packet size definition can promote the productivity of network resources and the quality of the provided services.

First day (onset) and last day (offset) of monsoon are nature phenomena which are important elements at cultivation stages in agriculture. These 2 sets of time value influent harvest performance but it is difficult to predict onset and offset at drought region. One of technique that can be used to solve mentioned problem is prediction technique which is one of data mining task. In this research, Feed Forward Backpropagation (BPNN) were combined with Moron method to predict onset and offset at drought region. Data used were daily rainfall data from 1983 to 2013. This experiment used 2 kind of BPNN models and they used 5 different values for learning rate (alpha) from range 0.01 to 0.2. Root Mean Square Error (RMSE) is used to evaluate resulted prediction models along with correlation value and standard deviation of error for better understanding. For BPNN onset model, lowest RMSE value at alpha 0.15 is 32,0546 and lowest RMSE value for BPNN offset is 26,6977 at alpha 0.05. Developed model has been able to use for prediction, but the result was still not close enough to actual data. In order to achieve a better model with lower RMSE, it is neccesary to improve model architecture and to specify some methods to obtain certain number of input layer based on Southern Oscillation Index (SOI) data.

Several works in the literature so far have been focused on deterministic point forecasts, which, usually, indicates the conditional mean of future observations. An increasing need for generating the entire conditional distribution of future observations has been required for the new generation of soft sensors. This study aims the probabilistic forecasts, reporting the use of a hybrid fuzzy forecasting model applied in two different forecasting problems. Our adapted model is applied to predict the rain of the city of Vitoria, in the state of Espírito Santo, Brazil. Real data from a wind farm, provided by the Irish EirGrid institute, was used for analyzing the proposal over a real time series with high fluctuations. Due to the stochasticity of the the hybrid model, which is calibrated through the use of an evolutionary metaheuristic, we adapted it in order to generate future using quantile regression. Computational experiments indicated the ability of the model in finding useful probabilistic quantiles, which were flexible enough in order to limit the lower and upper bounds of the historical datasets. While the probabilistic quantiles suggested the probability of rain and its magnitude, they were also able to predict expected ranges of the amount of energy generated from the wind farm.

Processing surges are fast and unexpected changes in the processing demand that commonly occur in cloud computing. The cloud elasticity enables to handle processing surges, increasing and decreasing resources as required. However, a surge can be very fast, so that the overhead to provide more resource is greater than the processing benefit. On the other hand, if the surge is slow and continuous, and the required resources are not provided, the application performance may be impaired or interrupted. This paper presents a machine learning-based approach to detect and classify processing surges, in order to improve the cloud resource management, minimizing losses for the application and cloud provider. We use a real cloud dataset to select features, to construct the classifier and to test our approach, which successfully detected and classified 99% of the processing surges.

Hyper-Heuristics are high-level methodologies developed to select or generate heuristics for solving complex problems. Despite their success, there is a lack of multi-objective hyper-heuristics. In the multi-objective optimization context, MOEA/D decomposes a problem into a number of sub problems handled by individuals in a collaborative manner. Our approach, named MOEA/D-HH SW , expands the MOEA/D framework with a multi-objective selection hyper-heuristic. It uses the proposed adaptive choice function with sliding window to determine which low-level heuristic (differential evolution operators) should be applied by each individual during MOEA/D execution. The proposed approach is tested in three known instances of the multi-objective environmental/economic dispatch problem, formulated as a non-linear constrained optimization problem with competing and non-commensurable objectives. MOEA/D-HH SW outperforms state-of-the-art algorithms reported in the literature for all considered instances.
