Individuals with inflammatory bowel disease(IBD)have been reported to be at an increased risk of infertility and sexual *** the relationship between them remains unclear,IBD severity is suspected to affect hormone levels and *** analyze the impact of IBD severity on semen parameters and sex hormone levels in ulcerative colitis-type IBD(UC-IBD),we conducted a cross-sectional study involving 120 patients with UC-IBD in Adam Malik General Hospital,Medan,*** patients were classified into three groups based on the Mayo score for Uc,followed by a comparison of various semen and hormone parameters among these *** addition to the cross-sectional analysis,a simple correlation test was conducted irrespective of the patient *** concentration,motility,and morphology were found to decline significantly with an increase in IBD *** classifying patients with IBD into subgroups,the Mayo score showed negative correlations with sperm concentration(r=-0.375,P<0.0001),rapid progressive motility(r=-0.660,P<0.0001),free testosterone(r=-0.732,P<0.0001),and total testosterone(r=-0.721,P<0.0001),and positivecorrelations with immotile sperm(r=0.660,P<0.0001),abnormal morphology(r=0.657,P<0.0001),and sex hormone-binding globulin(SHBG;r=0.278,P=0.002).Sperm concentration,motility,and morphology declined significantly with the severity of *** study suggests a significant negative impact of IBD severityonsemenqualityand sexhormones.

Acromegaly is a challenging medical condition that arises from the excessive production of growth hormones and the insulin-like growth factor 1 in the pituitary gland. While surgery is the primary treatment for acromegaly, medication is increasingly being used in patients who are unsuitable for surgery or have experienced treatment failure. Despite advancements in medical and surgical therapies, the treatment of acromegaly remains challenging. In this research, a three-dimensional (3D) in-vitro cell culture model for pituitary adenoma research was developed using hydrogel fiber meshes (HFMs) and GH3 cells. Electrospun nanofibers based on polyvinyl alcohol and polyacrylic acid were converted into HFMs by hydrogelification with the leaching of electrosprayed cellulose acetate beads for porosity enhancement. GH3 cells grown in the 3D model exhibited increased dispersion and upregulation of the somatostatin receptor subtypes 2 and 5 compared to those grown in traditional 2D cultures, as well as high sensitivity to somatostatin analogs and tumor-like profiles (as indicated by functional assays and transcriptome analysis, respectively). Therefore, the proposed 3D model accurately represents the physiological response to pituitary-adenoma therapeutic agents. This study highlights the potential of HFMs as a versatile platform for 3D in-vitro cell culture models that can be employed for pituitary adenoma research. Moreover, the proposed 3D cell culture model may contribute to a deeper understanding of tumor biology and facilitate the development of effective therapeutic strategies for acromegaly. © 2024 The Authors

Bacteriophage-cocktail hydrogel dressing to prevent multiple bacterial infections and heal diabetic ulcers in mice

Bacteriophage (phage) has been reported to reduce the bacterial infection in delayed-healing wounds and, as a result, aiding in the healing of said wounds. In this study we investigated whether the presence of phage itself could help repair delayed-healing wounds in diabetic mice. Three strains of phage that target Salmonella enterica, Escherichia coli, and Pseudomonas aeruginosa were used. To prevent the phage liquid from running off the wound, the mixture of phage (phage-cocktail) was encapsulated in a porous hydrogel dressing made with three-dimensional printing. The phage-cocktail dressing was tested for its phage preservation and release efficacy, bacterial reduction, cytotoxicity with 3T3 fibroblast, and performance in repairing a sterile full-thickness skin wound in diabetic mice. The phage-cocktail dressing released 1.7%–5.7% of the phages embedded in 24 h, and reduced between 37%–79% of the surface bacteria compared with the blank dressing (p © 2024 Wiley Periodicals LLC.

Inflammatory Bowel Disease (IBD) is a term that includes two chronic conditions characterized by inflammation of the gastrointestinal tract, i.e., Ulcerative Colitis and Crohn's Disease. IBD can be associated to symptoms that significantly reduce quality of life and can lead to serious complications, therefore proper disease monitoring and treatment is necessary. To this end, Telemedicine has been identified as a useful tool, since it can ease the patients' burden on the management of nutrition and pharmacological treatment, and can improve the connection with the physicians. In this paper, we present NutriA, a new responsive web application for the management of IBD patients in Italy, created to assist patients with diet, medications and symptoms monitoring, optimizing the follow-ups frequency and providing direct assistance by the doctors. Besides being useful to improve the patients' well-being, NutriA is also designed to be used in a new clinical study on the effects of malnutrition on IBD treatment. © 2024 IEEE.

A Novel SERS Substrate Containing Silver Nanoparticle-Deposited Photoresist Microcone Array for Sensitive Biosensing

In this study, we developed a novel surface-enhanced Raman scattering (SERS) substrate formed by depositing silver nanoparticles (AgNPs) on a silver thin film-coated photoresist (PR) microcone array. The semiconductor photolithographic process was adopted for fabricating the PR microcone array. The hotspot enhancement effect of the SERS substrates was optimized by adjusting the concentration of the trisodium citrate, which was used as a reducing agent and a stabilizing agent for the synthesis of AgNPs. Experimental results showed that the proposed SERS substrate could detect the indicating peaks of Rhodamine 6G (R6G) at a relatively low concentration of 10-12 M. A standard detection curve for HbA1c detection with a linear detection range of 5000-20000 ng/mL with good linearity of 0.994 and a limit of detection (LOD) of 850.2 ng/mL was further established by adding HbA1c agent to artificial blood to simulate a real environmental condition. Recovery rate detection experiments confirmed the reproducibility of the established standard detection curve. © 2001-2012 IEEE.
